Protective role of Triphala, an Indian traditional herbal formulation, against the nephrotoxic effects of bromobenzene in Wistar albino rats

OBJECTIVE： The purpose of the present study was to evaluate the nephroprotective and antioxidant properties of Triphala against bromobenzene-induced nephrotoxicity in female Wistar albino rats. METHODS： Animals were divided into five groups of six rats and treated as follows： Group I was a normal control and received no treatment, Group II received only bromobenzene （10 mmol/kg）, Groups III and IV received bromobenzene and Triphala （250 and 500 mg/kg, respectively）, Group V received Triphala alone （500 mg/kg）, and Group VI received bromobenzene and silymarin （100 mg/kg） Antioxidant status and serum kidney functional markers were analyzed. RESULTS： Bromobenzene treatment resulted in significant （P 〈 0.05） decreases in the activities of antioxidant enzymes such as catalase, superoxide dismutase, glutathione-S-transferase and glutathione peroxidase as well as total reduced glutathione. There was a significant （P 〈 0.05） increase in lipid peroxidation in kidney tissue homogenates. There were significant （P 〈 0.05） reductions in the levels of serum total protein and albumin as well as significant （P 〈 0.05） increases in serum creatinine, urea and uric acid. The oral administration of two different doses （250 and 500 mg/kg） of Triphala in bromobenzene-treated rats normalized the tested parameters. The histopathological examinations of kidney sections of the experimental rats support the biochemical observations. CONCLUSION： Triphala treatment alleviated the nephrotoxic effects of bromobenzene by increasing the activities of antioxidant enzymes and reducing the levels of lipid peroxidation and kidney functional markers.

Clinical and experimental research in antituberculosis drug-induced hepatotoxicity:a review

Drug-induced liver injury is the common adverse effect seen in patients receiving antituberculosis drugs （ATDs）. There are several risk factors associated with the development of hepatotoxicity in such patients. Though there have been appreciable efforts taken by carrying out studies investigating the efficacy of several natural and synthetic compounds in minimising this effect, the only choice available for clinicians is withdrawal of drugs. This review would give a precise idea of ATD-induced hepatotoxicity its underlying mechanisms and alternative therapies for the same.

Efficacy of boswellic acid on lysosomal acid hydrolases,lipid peroxidation and anti-oxidant status in gouty arthritic mice

Objective:To evaluate the efficacy of boswellic acid against monosodium urate crystal-induced inflammation in ***:The mice were divided into four experimental ***Ⅰserved as control;mice in groupⅡwere injected with monosodium urate crystal;groupⅢconsisted of monosodium urate crystal-induced mice who were treated with boswellic acid(30mg/kg/b.w.);groupⅣcomprised monosodium urate crystal-induced mice who were treated with indomethacin(3mg/kg/b.w.).Paw volume and levels/activities of lysosomal enzymes,lipid peroxidation,anti-oxidant status and inflammatory mediator TNF-αwere determined in control and monosodium urate crystal-induced *** addition,the levels ofβ-glucuronidase and lactate dehydrogenase were also measured in monosodium urate crystal-incubated polymorphonuclear leucocytes(PMNL)in ***:The activities of lysosomal enzymes,lipid peroxidation,and tumour necrosis factor-αlevels and paw volume were increased significantly in monosodium urate crystal-induced mice,whereas the activities of antioxidant status were in turn ***,these changes were modulated to near normal levels upon boswellic acid *** vitro,boswellic acid reduced the level ofβ-glucuronidase and lactate dehydrogenase in monosodium urate crystal-incubated PMNL in concentration dependent manner when compared with control ***:The results obtained in this study further strengthen the anti-inflammatory/antiarthritic effect of boswellic acid,which was already well established by several investigators.

Piperine,an active ingredient of black pepper attenuates acetaminophen-induced hepatotoxicity in mice

Objective:To explore the hepatoprotective and antioxidant effects of piperine against acetaminophen-induced hepatotoxicity in ***:In mice,hepatotoxicity was induced by a single dose of acetaminophen(900 mg/kg b.w.i.p.).Piperine(25 mg/kg b.w.i.p.) and standard drug silymarin(25 mg/kg b.w.i.p.) were given to mice,30 min after the single injection of *** 4 h,the mice were *** of liver marker enzymes [(aspartate transaminase(AST),alanine transaminase(ALT),and alkaline phosphatase(ALP)]and inflammatory mediator tumour necrosis factor-alpha(TNF-α) were estimated in serum,while lipid peroxidation and antioxidant status(superoxide dismutase,catalase,glutathione peroxidase, glutathione reductase,glutathione-s-transferase and glutathione) were determined in liver homogenate of control and experimental ***:Acetaminophen induction(900 mg/kg b.w.i.p.) significantly increased the levels of liver marker enzymes,TNF-α,and lipid peroxidation,and caused the depletion of antioxidant *** and silymarin treatment to acetaminophen challenged mice resulted in decreased liver marker enzymes activity,TNF-αand lipid peroxidation levels with increase in antioxidant ***:The results clearly demonstrate that piperine shows promising hepatoprotective effect as comparable to standard drug silymarin.

目的:研究对香豆酸,一种食物中含有的酚酸对佐剂性关节炎大鼠的抗过氧化治疗作用。方法:大鼠右后肢脚垫皮内注射0.1mL弗氏完全佐剂诱发关节炎。于佐剂注射后第11～18天连续给药8d,治疗组用药为对香豆酸(100mg/kg大鼠体质量,腹腔内给药),阳性对照组药物为吲哚美辛(3mg/kg大鼠体质量,腹腔内给药)。对佐剂性关节炎大鼠的肝脏、脾脏、血浆内脂质过氧化作用水平和抗氧化水平的变化进行测量,同时分析大鼠后肢的影像学改变以衡量对香豆酸的抗氧化作用。结果:佐剂性关节炎大鼠经对香豆酸治疗后,脂质过氧化作用和抗氧化水平恢复近正常值。影像学观察发现,治疗组大鼠的软组织肿胀、骨侵蚀及关节间隙狭窄情况得到缓解。结论:对香豆酸对佐剂性关节炎大鼠有明显的治疗作用,这种治疗作用可能与其抗氧化作用相关。

目的:研究生姜提取物6-姜酚对对乙酰氨基酚致小鼠肝脏毒性的保护作用。方法:实验组小鼠在腹腔注射对乙酰氨基酚(900 mg/kg)30 min后,分别给予6-姜酚(30 mg/kg)或标准对照药水飞蓟素(25 mg/kg)。注射对乙酰氨基酚4 h后处死小鼠,检测血清中天冬氨酸氨基转移酶、丙氨酸氨基转移酶、碱性磷酸酶活性,总胆红素的含量及肝匀浆中脂质过氧化及抗氧化情况,如超氧化物歧化酶、过氧化氢酶、谷胱甘肽过氧化物酶、谷胱甘肽还原酶、谷胱甘肽转移酶活性及还原型谷胱甘肽含量。结果:与对照组相比,6-姜酚及水飞蓟素均能显著降低对乙酰氨基酚引起的小鼠血清天冬氨酸氨基转移酶、丙氨酸氨基转移酶、碱性磷酸酶活性及总胆红素含量的升高(P<0.05)。此外,6-姜酚及水飞蓟素有效控制了肝脏内丙二醛的形成及各种抗氧化酶的降低(P<0.05)。结论:本研究的结果证实了6-姜酚具有与水飞蓟素相当的保肝作用。

Further Discussion on the ＂Two Suns Theory＂ and the Monarchia of Dante＂

The De Monarchia can be considered the summa of Dante＇s political thought, of which we can also find some starting points in the Convivio, in the Epistles and in the Divine Comedy. Here, in Purgatorio XVI, Marco Lombardo, after stating that the misrule of the popes led the world to the sin, articulates Dante＇s view of the Empire and Papacy as separate authorities and cites the instance of Rome at the pagan Age, when Rome used to possess two autonomous institutions to drive mankind both towards the material happiness and the spiritual one （vv. 106 ff. ＂soleva Roma, che ＇1 buon mondo feo/due soli aver, che l＇una e l＇altra strada/facean vedere, e del mondo e di Dio＂）. The image of＂two suns＂ also returns in the III Book of De Monarchia by Dante. Here Dante, inquiring into the relationship between ＂the two greatest luminaries＂, that is the Roman Pontiff and the Roman prince, wonders if the authority of the Roman ruler descends directly from God or from someone of His ministers. For Dante the Emperor, whose authority is given to him directly by God, does not depend on the Pope, but the Emperor is absolutely independent of the Pope.

Estrogen deficiency reversibly induces telomere shortening in mouse granulosa cells and ovarian aging in vivo

Estrogen is implicated as playing an important role in aging and tumorigenesis of estrogen responsive tissues;however the mechanisms underlying the mitogenic actions of estrogen are not fully *** we report that estrogen deficiency in mice caused by targeted disruption of the aromatase gene results in a signi-ficant inhibition of telomerase maintenance of telomeres in mouse ovaries in a tissue-specific *** inhibition entails a significant shortening of telomeres and compromised proliferation in the follicular granulosa cell compartment of *** expression analysis showed decreased levels of proto-oncogene c-Myc and the telomerase catalytic subunit,telomerase reverse transcriptase(TERT),in response to estrogen *** replacement therapy led to increases in TERT gene expression,telomerase activity,telomere length and ovarian tissue growth,thereby reinstating ovary development to normal in four *** data demonstrate for the first time that telomere maintenance is the primary mechanism mediating the mitogenic effect of estrogen on ovarian granulosa cell proliferation by upregulating the genes of c-Myc and TERT in *** deficiency or over-activity may cause ovarian tissue aging or tumorigenesis,respectively,through estrogen regulation of telomere remodeling.

Corneal transplantation: Beyond the horizon

Evolving techniques in keratoplasty have undoubtedly led to thinner corneal grafts. These newer iterations of keratoplasty aim to reduce graft rejections, improve visual acuity and visual rehabilitation. Each technique poses its own advantages and disadvantages; the surgeon should select patients suitable for a particular technique while accounting for their surgical competency given the learning curve associated with these newer techniques. Alternatives to corneal transplant may have a role in addressing the shortages of corneal graft, these bioengineered material and medical treatment still need further studies to demonstrate its clinical applicability.
