SArS-CoV-2 is an emerging viral pathogen and a major global public health challenge since December of 2019, with limited effective treatments throughout the *** part of the innate immune response to viral infection, type Ⅰ interferons(IFN-Ⅰ) trigger a signaling cascade that culminates in the activation of hundreds of genes, known as interferon stimulated genes(ISGs), that collectively foster an antiviral *** report here the identification of a group of type Ⅰ interferon suppressed genes,including fatty acid synthase(FASN), which are involved in lipid *** of FASN or the addition of its downstream product, palmitate, increased viral infection while knockout or knockdown of FASN reduced *** importantly, pharmacological inhibitors of FASN effectively blocked infections with a broad range of viruses, including SArS-CoV-2 and its variants of ***, our studies not only suggest that downregulation of metabolic genes may present an antiviral strategy by type Ⅰ interferon, but they also introduce the potential for FASN inhibitors to have a therapeutic application in combating emerging infectious diseases such as COVID-19.

SArS-CoV-2 virus NSP14 Impairs NrF2/HMOX1 activation by targeting Sirtuin 1

Most deaths from the COVID-19 pandemic are due to acute respiratory distress syndrome(ArDS)-related respiratory *** storms and oxidative stress are the major players in ArDS development during respiratory virus ***,it is still unknown how oxidative stress is regulated by viral and host factors in response to SArS-CoV-2 ***,we found that activation of NrF2/HMOX1 significantly suppressed SArS-CoV-2 replication in multiple cell types by producing the metabolite biliverdin,whereas SArS-CoV-2 impaired the NrF2/HMOX1 axis through the action of the nonstructural viral protein ***,NSP14 interacts with the catalytic domain of the NAD-dependent deacetylase Sirtuin 1(SIrT1)and inhibits its ability to activate the NrF2/HMOX1 ***,both genetic and pharmaceutical evidence corroborated the novel antiviral activity of SIrT1 against ***,our findings reveal a novel mechanism by which SArS-CoV-2 dysregulates the host antioxidant defense system and emphasize the vital role played by the SIrT1/NrF2 axis in host defense against SArS-CoV-2.

Osteosarcomas are malignant neoplasms derived from undifferentiated osteogenic mesenchymal cells. It causes severe and permanent damage to human tissue and has a high mortality rate. The condition has the capacity to occur in any bone;however, it often impacts long bones like the arms and legs. Prompt identification and prompt intervention are essential for augmenting patient longevity. However, the intricate composition and erratic placement of osteosarcoma provide difficulties for clinicians in accurately determining the scope of the afflicted area. There is a pressing requirement for developing an algorithm that can automatically detect bone tumors with tremendous accuracy. Therefore, in this study, we proposed a novel feature extractor framework associated with a supervised three-class XGBoost algorithm for the detection of osteosarcoma in whole slide histopathology images. This method allows for quicker and more effective data analysis. The first step involves preprocessing the imbalanced histopathology dataset, followed by augmentation and balancing utilizing two techniques: SMOTE and ADASYN. Next, a unique feature extraction framework is used to extract features, which are then inputted into the supervised three-class XGBoost algorithm for classification into three categories: non-tumor, viable tumor, and non-viable tumor. The experimental findings indicate that the proposed model exhibits superior efficiency, accuracy, and a more lightweight design in comparison to other current models for osteosarcoma detection.

Particle shape consideration in numerical simulation of assemblies of irregularly shaped particles

The mechanical behavior of granular materials depends much on the shape of the constituent particles. Therefore appropriate modeling of particle, or grain, shape is quite important. This study employed the method of direct modeling of grain shape （Matsushima ＆ Saomto, 2002）, in which, the real shape of a grain is modeled by combining arbitrary number of overlapping circular elements which are connected to each other in a rigid way. Then, accordingly, a discrete-element program is used to simulate the assembly of grains. In order to measure the effects of grain shape on mechanical properties of assembly of grains, three types of grains-high angular grains, medium angular grains and round grains are considered where several biaxial tests are conducted on assemblies with different grain types, The results show that the angularity of grains greatly affects the behavior of granular soil.

Highway safety researchers focus on crash injury severity,utilizing deep learning—specifically,deep neural networks(DNN),deep convolutional neural networks(D-CNN),and deep recurrent neural networks(D-rNN)—as the preferred method for modeling accident *** learning’s strength lies in handling intricate relation-ships within extensive datasets,making it popular for accident severity level(ASL)prediction and *** prior success,there is a need for an efficient system recognizing ASL in diverse road *** address this,we present an innovative Accident Severity Level Prediction Deep Learning(ASLP-DL)framework,incorporating DNN,D-CNN,and D-rNN models fine-tuned through iterative hyperparameter selection with Stochastic Gradient *** framework optimizes hidden layers and integrates data augmentation,Gaussian noise,and dropout regularization for improved *** and factor contribution analyses identify influential *** on three diverse crash record databases—NCDB 2018–2019,UK 2015–2020,and US 2016–2021—the D-rNN model excels with an ACC score of 89.0281%,a roc Area of 0.751,an F-estimate of 0.941,and a Kappa score of 0.0629 over the NCDB *** proposed framework consistently outperforms traditional methods,existing machine learning,and deep learning techniques.

Trustworthy Explainable recommendation Framework for relevancy

Explainable recommendation systems deal with the problem of‘Why’.Besides providing the user with the recommendation,it is also explained why such an object is being *** helps to improve trustworthiness,effectiveness,efficiency,persuasiveness,and user satisfaction towards the *** recommend the relevant information with an explanation to the user is *** systems provide the top-k recommendation options to the user based on ratings and reviews about the required object but unable to explain the matched-attribute-based recommendation to the user.A framework is proposed to fetch the most specific information that matches the user requirements based on Formal Concept Analysis(FCA).The ranking quality of the recommendation list for the proposed system is evaluated quantitatively with Normalized Discounted Cumulative Gain(NDCG)@k,which is better than the existing *** is provided qualitatively by considering trustworthiness criterion i.e.,among the seven explainability evaluation criteria,and its metric satisfies the results of proposed *** framework can be enhanced to accommodate for more effectiveness and trustworthiness.

To determine the individual circumstances that account for a road traffic accident,it is crucial to consider the unplanned connections amongst various factors related to a crash that results in high casualty *** of the road accident data concentrated mainly on categorizing accidents into different types using individually built classification methods which limit the prediction accuracy and fitness of the *** this article,we proposed a multi-model hybrid framework of the weighted majority voting(WMV)scheme with parallel structure,which is designed by integrating individually implemented multinomial logistic regression(MLr)and multilayer perceptron(MLP)classifiers using three different accident datasets i.e.,IrTAD,NCDB,and *** proposed WMV hybrid scheme overtook individual classifiers in terms of modern evaluation measures like rOC,rMSE,Kappa rate,classification accuracy,and performs better than state-of-theart approaches for the prediction of casualty severity ***,the proposed WMV hybrid scheme adds up to accident severity analysis through knowledge representation by revealing the role of different accident-related factors which expand the risk of casualty in a road *** aspects related to casualty severity recognized by the proposed WMV hybrid approach can surely support the traffic enforcement agencies to develop better road safety plans and ultimately save lives.
