BACKGROUNd One of the primary reasons for the dismal survival rates in pancreatic ductal adenocarcinoma(PdAC)is that most patients are usually diagnosed at late *** is an urgent unmet clinical need to identify and develop diagnostic methods that could precisely detect PdAC at its earliest *** A total of 71 patients with pathologically proved PdAC based on surgical resection who underwent contrast-enhanced computed tomography(CT)within 30 d prior to surgery were included in the *** staging was performed in accordance with the 8th edition of the American Joint Committee on Cancer staging *** features were extracted from the region of interest(ROI)for each patient using Analysis Kit *** most important and predictive radiomics features were selected using Mann-Whitney U test,univar-iate logistic regression analysis,and minimum redundancy maximum relevance(MRMR)*** forest(RF)method was used to construct the radiomics model,and 10-times leave group out cross-validation(LGOCV)method was used to validate the robustness and reproducibility of the *** A total of 792 radiomics features(396 from late arterial phase and 396 from portal venous phase)were extracted from the ROI for each patient using Analysis Kit *** most important and predictive features were selected using Mann-Whitney U test,univariate logistic regression analysis,and MRMR *** method was used to construct the radiomics model with the nine most predictive radiomics features,which showed a high discriminative ability with 97.7%accuracy,97.6%sensitivity,97.8%specificity,98.4%positive predictive value,and 96.8%negative predictive *** radiomics model was proved to be robust and reproducible using 10-times LGOCV method with an average area under the curve of 0.75 by the average performance of the 10 newly built *** The radiomics model based on CT could serve as a promising non-invasive method in differential diag

Extreme droughts are anticipated to have detrimental impacts on forest ecosystems,especially in water-limited regions,due to the influence of climate ***,considerable uncertainty remains regarding the patterns in species-specific responses to extreme ***,we conducted a study integrating dendrochronology and remote sensing methods to investigate the mosaic-distributed maple-oak(native)natural forests and poplar plantations(introduced)in the Horqin sa.dy Land,Northeast *** assessed the impacts of extreme droughts on tree performances by measuring interannual variations in radial growth and vegetation *** results showed that precipitation and self-calibrated palmer drought severity index(scPdSI)are the major factors influencing tree-ring width index(RWI)and normalized difference vegetation index(NdVI).The severe droughts between 2000 and 2004 resulted in reduced RWI in the three studied tree species as well as led to NdVI reductions in both the maple-oak natural forests and the poplar *** RWI reached the na.ir during the2000-2004 severe droughts and remained at low levels two years after the severe drought,creating a legacy *** contrast to the lack of significant correlation between RWI and scPdSI,NdVI exhibited a significant positive correlation with scPdSI indicating the greater sensitivity of canopy performance to droughts than radial ***,interspecific differences in RWI and NdVI responses were observed,with the fast-growing poplar species experiencing a more significant RWI decrease and more negative NdVI anomaly during severe droughts than native species,highlighting the species-specific trade-offs between drought resilience and growth *** study emphasizes the importance of combining tree-level radial growth with landscape-scale canopy remote sensing to understand forest resilience and *** study improves our understanding of forest responses to extreme drought and highlights species di

采用水提醇沉法提取刺玫根粗多糖,以Sevage法和TCA法脱蛋白,进一步分级醇沉法分离纯化得到较纯的60%醇沉物(RdRP)进行结构鉴定和活性评价。苯酚浓硫酸法测定出刺玫根多糖RdRP总糖含量为94.1%;高效凝胶渗透色谱法测定表明主要含有重均分子量为13.1 kda,相对分散度为1.2的多糖;通过高效离子色谱法和红外光谱测定,鉴定刺玫根多糖为由葡萄糖、阿拉伯糖、半乳糖、甘露糖、木糖等5种单糖组成的含有α-构型的中性吡喃多糖。环磷酰胺诱导的免疫抑制小鼠模型实验结果表明,与模型组相比,刺玫根多糖各剂量组小鼠的脾脏和胸腺指数明显升高(Psa法检测血清中免疫相关蛋白IgG、IgM、IgA以及细胞因子TNF-α、IFN-γ、IL-6与IL-2的分泌水平,结果显示,与模型组比较,刺玫根多糖组显著上调小鼠血清中免疫球蛋白IgA、IgG、IgM浓度水平(Pd88、TRAF-6和NF-κB p65蛋白的表达水平,发现刺玫根多糖各剂量组脾脏中TLR4、TRAF-6蛋白极显著上升(Pd88蛋白极显著上升(P<0.01),其他组呈现上升趋势;刺玫根多糖各剂量组NF-κB p65蛋白均有上升趋势,因此其免疫调节作用机制可能与TLR/NF-κB通道有关。本研究初步阐明刺玫根多糖的基本结构特征和其免疫调节作用及其机制,为刺玫根多糖的开发应用提供一定价值的实验依据。

We revisit the γ-ray emission above 300 Me V towards the massive star-forming region of Orion B by adopting14 yr observations with the Fermi Large Area Telescope and utilizing the updated software *** extended γ-ray emission region around Orion B is resolved into two components(region Ⅰ and region Ⅱ).The γ-ray spectrum of region I agrees with the predicted γ-ray spectrum assuming the cosmic ray(CR)density is the same as that of Alpha Magnetic Spectrometer(AMS-02)measured ***γ-ray emissivity of region II appears to be deficit at low energy band(Edeling we find that CR densities exhibit a significant deficit below 20 Ge V,which may be caused by a slow diffusion inside the dense *** is probably caused by an increased magnetic field whose strength increases with the gas density.

The Ebola virus(EBOV)is a member of the Orthoebolavirus genus,Filoviridae family,which causes severe hemorrhagic diseases in humans and non-human primates(NHPs),with a case fatality rate of up to 90%.The development of countermeasures against EBOV has been hindered by the lack of ideal animal models,as EBOV requires handling in biosafety level(BSL)-4 ***,accessible and convenient animal models are urgently needed to promote prophylactic and therapeutic approaches against *** this study,a recombinant vesicular stomatitis virus expressing Ebola virus glycoprotein(VSV-EBOV/GP)was constructed and applied as a surrogate virus,establishing a lethal infection in *** infection with VSV-EBOV/GP,3-week-old female Syrian hamsters exhibited disease signs such as weight loss,multi-organ failure,severe uveitis,high viral loads,and developed severe systemic diseases similar to those observed in human EBOV *** animals succumbed at 2–3 days post-infection(dpi).Histopathological changes indicated that VSV-EBOV/GP targeted liver cells,suggesting that the tissue tropism of VSV-EBOV/GP was comparable to wild-type EBOV(WT EBOV).Notably,the pathogenicity of the VSV-EBOV/GP was found to be species-specific,age-related,gender-associated,and challenge ***,equine anti-EBOV immunoglobulins and a subunit vaccine were validated using this ***,this surrogate model represents a safe,effective,and economical tool for rapid preclinical evaluation of medical countermeasures against EBOV under BSL-2 conditions,which would accelerate technological advances and breakthroughs in confronting Ebola virus disease.
