GAPDH expression as a measurement of transfection efficiency for p16INk4agene silencing (siRNA) in senescent human diploid fibroblasts

Human diploid fibroblasts (HDFs) undergo a limited number of cell divisions in culture. After certain population doublings, they reach a state of irreversible growth arrest known as replicative senescence. Senescent HDFs showed several molecular and cytological changes such as large flat morphology, expression of senescence-associated β-galactosidase (SA β-gal) activity and altered gene expression. Small interfering RNA (siRNA) has been demonstrated to be a potential research tool to analyse gene function and pathway. Expression of an appropriate housekeeping or reference gene can be used as a measurement of transfection efficiency in siRNA. Therefore this study was designed to determine the suitability of GAPDH expression as a measurement of transfection efficiency for p16INk4a gene silencing in HDFs aging model. GAPDH knockdown with an appropriate transfection reagent was measured by quantitative real time RT-PCR while cellular senescence was characterized based on morphological changes, expression of SA β-gal and p16INk4a expression levels. Our findings showed that GAPDH knockdown represents silencing efficiency and down regulation of p16INk4a in senescent transfected HDFs caused morphological alterations which results in the formation of spindle shaped fibroblasts. This study demonstrated the suitability of GAPDH expression as a measurement of transfection efficiency for p16INk4a gene silencing in HDFs aging model.

This study aimed at prioritizing medicinal plants used to manage HIV/AIDS opportunistic infections and assessing their wild population status. Data were collected using focus group discussions and inventories. Preference ranking and Microsoft Excel and QGIS software were used for data analysis. Up to seven species were prioritized as the most important in the disease management. It was also found that the species dominated the landscape with elevation between 1950 to 2050 masl. Moreover, the species displayed linear pattern distribution adjacent rivers. With regards to population structure, the species revealed J-shaped curves. The species density was 200/ha for Hagenia abyssinica and 28/ha for Myrica salicifolia. The dominance of species in higher altitudes as cited above indicates that they flourish well in highlands. The tendency of them to grow adjacent water sources reveals their water or moisture loving. Moreover, the J-shaped curves observed imply poor recruitment and hence unsustainable. The study recommends for urgent conservation plans especially on the sampled species. There is a need to ensure strict measures are put in place to safeguard the medicinal plant species to ensure their sustainability.

Nowadays consumers seek foods that not only provide basic nutrition but also contain compounds that contribute to health. Thus, different products have been designed to meet this demand, especially those with probiotic and prebiotic characteristics. This study aimed to evaluate different formulations of a soy-based synbiotic beverage. The composition and process conditions were defined using a Placket & Burman factorial design in which the independent variables were process temperature, concentrations of the soy hydrosoluble extract and fructooligosaccharides (FOS) and the initial concentration of the probiotics Bifidobaterium longum, Lactobacillus acidophilus, Lactobacillus paracasei subsp. Paracasei, Lactobacillus rhamnosus. Changes to the product by addition of sucrose, Streptococcus thermophilus and Lactobacillus delbrueckii subsp. Bulgaricus were also studied. Response parameters were: pH, cell concentration (plate counting), and quantification of fructooligosaccharides, acetic acid and lactic acid. The specific growth rate (μx) and change in cell count (Δx) were determined in each formulation. The soybean hydrosoluble extract was considered a good substrate for beverage production;FOS had a greater effect at lower concentrations. At the end of all fermentation tests the total probiotic count was greater than 7 log CFU/mL, meeting the legal requirements of a functional food. Lower temperatures were better for the growth of probiotics. The addition of sucrose resulted in a lower consumption of FOS by the fermenting microorganisms, while the presence of Streptococcus thermophilus and Lactobacillus delbrueckii subsp. bulgaricus represented little change concerning the evaluated characteristics. Therefore, according to the results of this study, the production of a soy-based symbiotic fermented beverage can be performed using soybean hydrosoluble extract at 10% (w/v), fructooligosaccharides at about 3% (w/v), sucrose at 12% (w/v), and the process temperature should

Therefore,the concern for the human health,when exposed on EMF influence,occupies more and more attention and leads to series of investigations,related to EMF exposure. In this paper,a method for its determination of such an influence（when a human is exposed on high frequency EMF） is *** problems to be solved are modeling of the human exposed field,finding appropriate human model for presentation of his structure taking account his geometrical and electrical characteristics,finding suitable solving method for field determination and appropriate presentation of the results. Several studies indicate that music has soothing effects and is beneficial in reducing stress and anxiety in coronary patients. It is known that reactions to music are considered to be subjective, however, results of previous studies suggest that variables such as changes in heart rate, in cerebral blood flow and heart rate variability increase with a faster musical tempo, regardless of individual preferences. The effects of stress on the cardiovascular system have also been proven. However, the meanings assigned to music when used during hemodynamic procedures are unknown, as are the meanings of the experience of these procedures. The aim of this research is to understand the senses and feelings of music for patients undergoing hemodynamic procedures, identify and interpret the related fantasies and emotions and study the possibility of deploying in hospitals the “Musical Method for Hemodynamic Procedures”, being developed by the author. This research is based on a Clinical-Qualitative Methodology. The sampling method is the theoretical saturation. The semi structured interview was used in order to obtain data that were submitted to content analysis. The subjects are patients undergoing hemodynamic procedures in the Hospital SEMPER (Permanent Medical Service), Belo Horizonte, Brazil. We conclude that within the experience of listening to music while undergoing catheterization, 100% of the patients interviewed claimed they had overcome the experience of stress and felt calm, tranquility, peace and happiness. Some patients described the music as a companion, as something that diverts their attention from fear, transporting them to an imaginary place, to another dimension. The episodic memory, the capacity to recognize a musical excerpt for which the spatiotemporal context surrounding its former encounter could be recalled, was also important, with surprising results, in the case of patients who underwent catheterization in the presence of music and, later, angioplast

This article is the result of a research carried out in a private hospital of Belo Horizonte, Brazil. The patient care model of the Mastology Department includes an interdisciplinary staff that aims for the comprehensive treatment of these surgical patients. The research is based on clinical approach, under the framework of Analytical Psychology. Feminine archetypal images are symbolically represented by the Main Goddesses of the Olympus. It is natural for one of these expressions to prevail throughout a woman’s life, or even in specific moments of her life, but it is important to recognize that all of them are part of the feminine soul. We can, therefore, define and understand the Goddesses from Greek Mythology symbolically as being representative of the various feminine psychogenic forces that are projected. In reality, they are internal images that can be used to help know and understand personal issues and changes that are necessary in a woman’s life, enabling personal growth and the possibility to identify herself with the more authentic elements of her being. In this work experience, the feminine force showed itself to be a force of creation, of expression and of overcoming.
