Tetracycline repressor(TetR)family regulators(TFRs)are the largest group of DNA-binding transcription factors and are widely distributed in bacteria and *** play vital roles in controlling the expression of various genes and regulating diverse physiological ***,a TFR protein Pseudomonas virulence regulator A(PvrA),was identified from Pseudomonas aeruginosa as the transcriptional activator of genes involved in fatty acid utilization and bacterial ***,we show that PvrA can simultaneously bind to multiple pseudo-palindromic sites and upregulate the expression levels of target ***-electron microscopy(cryo-EM)analysis indicates the simultaneous DNA recognition mechanism of PvrA and suggests that the bound DNA fragments consist of a distorted B-DNA double *** crystal structure and functional analysis of PvrA reveal a hinge region that secures the correct domain motion for recognition of the promiscuous ***,our results showed that mutations disrupting the regulatory hinge region have differential effects on biofilm formation and pyocyanin biosynthesis,resulting in attenuated bacterial ***,these findings will improve the understanding of the relationship between the structure and function of the TetR family and provide new insights into the mechanism of regulation of *** virulence.

党的十八大以来,中国人才强国战略位置被进一步提升,其实施可划分为人才优先发展战略布局推进落地期、人才发展体制机制深化改革肇始期、人才引领发展战略地位形成转换期和新时代人才强国战略部署启动期等四个阶段。党的十八大以来,我国人才工作取得历史性成就、发生历史性变革。其中,主要人才发展指标大幅增长,人才规模、素质和创新效能达到历史新高位;人才体制机制改革和政策深度破题,人才创新创业活力进一步释放;重大人才计划工程有效实施,成为高层次人才梯队建设的有力抓手;国家人才中心城市正在加速崛起,以点带面对周边区域产生辐射带动效应。本文认为,实施人才强国战略是中国式现代化发展模式的有效替代物,其战略内核和重心随国家总体战略布局而动态调整进阶,面向新时代新阶段实施人才强国战略需要亟需构建与之相匹配的国家人才发展治理新体系。

Environmental factors such as diet and lifestyle can influence the health of both mothers and ***,its transgenerational transmission and underlying mechanisms remain largely ***,using a maternal lactation-period low-protein diet(LPD)mouse model,we show that maternal LPD during lactation causes decreased survival and stunted growth,significantly reduces ovulation and litter size,and alters the gut microbiome in the female LPD-F1 *** transcriptome of LPD-F1 metaphase II(MII)oocytes shows that differentially expressed genes are enriched in female pregnancy and multiple metabolic ***,maternal LPD causes early stunted growth and impairs metabolic health,which is transmitted over two *** methylome alteration of LPD-F1 oocytes can be partly transmitted to the F2 ***,our results reveal that LPD during lactation transgenerationally affects offspring health,probably via oocyte epigenetic changes.

Lipogenesis is often highly upregulated in breast cancer brain metastases to adapt to intracranial low lipid *** inhibitors hold therapeutic potential but their intra-tumoral distribution is often blocked by the blood-tumor barrier(BTB).BTB activates its Wnt signaling to maintain barrier properties,e.g.,Mfsd2a-mediated BTB low ***,we reported VCAM-1-targeting nano-wogonin(W@V-NPs)as an adjuvant of nano-orlistat(O@V-NPs)to intensify drug delivery and inhibit lipogenesis of brain metastases.W@V-NPs were proven to be able to inactivate BTB Wnt signaling,downregulate BTB Mfsd2a,accelerate BTB vesicular transport,and enhance tumor accumulation of O@*** the ability to specifically kill cancer cells in a lipid-deprived environment with IC_(50) at 48 ng/mL,W@V-NPs plus O@V-NPs inhibited the progression of brain metastases with prolonged survival of model *** combination did not induce brain edema,cognitive impairment,and systemic toxicity in healthy *** Wnt signaling could safely modulate the BTB to improve drug delivery and metabolic therapy against brain metastases.

Nucleotide second messengers are highly versatile signaling molecules that regulate a variety of key biological processes in *** best-studied examples are cyclic AMP(cAMP)and bis-(3'-5')-cyclic dimeric guanosine monophosphate(c-di-GMP),which both act as global *** regulatory frameworks of c-di-GMP and cAMP in bacteria show several parallels but also significant *** this review,we llustrate the global regulatory models of the two nucleotide second messengers,compare the different regulatory frameworks between c-di-GMP and cAMP,and discuss the mechanisms and physiological significance of cross-regulation between c-di-GMP and cAMP.c-di-GMP responds to numerous signals de-pendent on a great number of metabolic enzymes,and it regulates various signal transduction pathways through its huge number of effectors with varying *** contrast,due to the limited quantity,the cAMP metabolic enzymes and its major effector are regulated at different levels by diverse *** performs its global regulatory function primarily by controlling the transcription of a large number of genes via cAMP receptor protein(CRP)in most *** review can help us understand how bacteria use the two typical nucleotide second messengers to effectively coordinate and integrate various physiological processes,providing theoretical guidelines for future research.

Divergent responses of runoff to climate change in the upper basins of the Third Pole dominated by westerlies and monsoon

The diverse climates,distribution of snow and glaciers,and geographic locations directly affect the runoff response to climate change in the upper basins of the Third *** present,a comprehensive analysis of runoff variations and their distinct responses to climate change in the westerlies-and monsoon-dominated upper basins is still *** study comprehensively analyzed annual runoff variations in westerlies-dominated basins(the upper basins of the Aksu(UAKS),Syr Darya(USRD),Yarkant(UYK),Hotan(UHT),Amu Darya(UAMD),and Indus(UI))and monsoon-dominated basins(the upper basins of the Yangtze(UYA),Yellow(UYE),Lancang(ULC),Nujiang(UNJ),and Yarlung Zangbo(UYZ))of the Third Pole from 1961 *** multi-source meteorological data and large-scale circulation factors,this study investigated the divergent responses of runoff in the upper basins to climate change,and explored the large-scale circulation mechanisms underlying runoff variations in these upper *** results showed that:(1)The annual runoff in the majority of upper basins(except for the UYE and UYZ)exhibited an increasing trend,and the annual runoff in the UAKS,UYK,and UI showed a significant increasing trend from1961 to *** annual runoff in the upper basins of the Third Pole changed abruptly from decreasing to increasing between the 1980s and 2000s,with the exception of the UYE.(2)The runoff in the monsoon-dominated upper basins has been controlled primarily by changes in precipitation over the past 55 *** contrast,the runoff in the westerlies-dominated upper basins exhibited three distinct long-term responses to climate change:temperature-dominated(UYK and UHT),precipitation-dominated(USRD and UAMD),and the combined influence of precipitation and temperature(UAKS and UI).Since the 1960s,the sensitivity of runoff to warm season temperature changes in the most westerlies-dominated upper basins has decreased,while the response of runoff to precipitation changes has intensified.(3)The stud

Correlation between abdominal computed tomography signs and postoperative prognosis for patients with colorectal cancer

BACKGROUND Patients with different stages of colorectal cancer(CRC)exhibit different abdominal computed tomography(CT)***,the influence of CT signs on CRC prognosis must be *** To observe abdominal CT signs in patients with CRC and analyze the correlation between the CT signs and postoperative *** The clinical history and CT imaging results of 88 patients with CRC who underwent radical surgery at Xingtan Hospital Affiliated to Shunde Hospital of Southern Medical University were retrospectively *** and multivariate Cox regression analyses were used to explore the independent risk factors for postoperative death in patients with *** three-year survival rate was analyzed using the Kaplan-Meier curve,and the correlation between postoperative survival time and abdominal CT signs in patients with CRC was analyzed using Spearman correlation *** For patients with CRC,the three-year survival rate was 73.86%.The death group exhibited more severe characteristics than the survival group.A multivariate Cox regression model analysis showed that body mass index(BMI),degree of periintestinal infiltration,tumor size,and lymph node CT value were independent factors influencing postoperative death(Ptients with characteristics typical to the death group had a low three-year survival rate(log-rankχ2=66.487,11.346,12.500,and 27.672,respectively,Ptients was negatively correlated with BMI,degree of periintestinal infiltration,tumor size,lymph node CT value,mean tumor long-axis diameter,and mean tumor short-axis diameter(r=-0.559,0.679,-0.430,-0.585,-0.425,and-0.385,respectively,P<0.05 for all).BMI was positively correlated with the degree of periintestinal invasion,lymph node CT value,and mean tumor short-axis diameter(r=0.303,0.431,and 0.437,respectively,P<0.05 for all).CONCLUSION The degree of periintestinal infiltration,tumor size,and lymph node CT value are c
