Sequential neoadjuvant chemotherapy using pegylated liposomal doxorubicin and cyclophosphamide followed by taxanes with complete trastuzumab and pertuzumab treatment for HER2-positive breast cancer: A phase Ⅱ single-arm study

Objective: Despite cardiotoxicity overlap, the trastuzumab/pertuzumab and anthracycline combination remains crucial due to significant benefits. Pegylated liposomal doxorubicin(PLD), a less cardiotoxic anthracycline, was evaluated for efficacy and cardiac safety when combined with cyclophosphamide and followed by taxanes with trastuzumab/pertuzumab in human epidermal growth factor receptor-2(HER2)-positive early breast cancer(BC).Methods: In this multicenter, phase II study, patients with confirmed HER2-positive early BC received four cycles of PLD(30-35 mg/m^(2)) and cyclophosphamide(600 mg/m^(2)), followed by four cycles of taxanes(docetaxel,90-100 mg/m^(2) or nab-paclitaxel, 260 mg/m^(2)), concomitant with eight cycles of trastuzumab(8 mg/kg loading dose,then 6 mg/kg) and pertuzumab(840 mg loading dose, then 420 mg) every 3 weeks. The primary endpoint was total pathological complete response(tp CR, yp T0/is yp N0). Secondary endpoints included breast p CR(bp CR),objective response rate(ORR), disease control rate, rate of breast-conserving surgery(BCS), and safety(with a focus on cardiotoxicity).Results: Between May 27, 2020 and May 11, 2022, 78 patients were treated with surgery, 42(53.8%) of whom had BCS. After neoadjuvant therapy, 47 [60.3%, 95% confidence interval(95% CI), 48.5%-71.2%] patients achieved tp CR, and 49(62.8%) achieved bp CR. ORRs were 76.9%(95% CI, 66.0%-85.7%) and 93.6%(95% CI,85.7%-97.9%) after 4-cycle and 8-cycle neoadjuvant therapy, respectively. Nine(11.5%) patients experienced asymptomatic left ventricular ejection fraction(LVEF) reductions of ≥10% from baseline, all with a minimum value of >55%. No treatment-related abnormal cardiac function changes were observed in mean N-terminal pro-BNP(NT-pro BNP), troponin I, or high-sensitivity ***: This dual HER2-blockade with sequential polychemotherapy showed promising activity with rapid tumor regression in HER2-positive BC. We operated a p-type point contact high purity germanium(PPCGe)detector(CDEX-1B,1.008 kg)in the China jinping Underground Laboratory(CJPL)for 500.3 days to search for neutrinoless double beta(0νββ)decay of^(76)Ge.A total of 504.3 kg⋅day effective exposure data was *** anti-coincidence and the multi/single-site event(MSE/SSE)discrimination methods were used to suppress the background in the energy region of interest(ROI,1989–2089 keV for this work)with a factor of 23.A background level of 0.33 counts/(keV⋅kg⋅yr)was *** lower limit on the half life of^(76)Ge 0νββdecay was constrained as T_(1/2)^(0ν)>1.0×10^(23)yr(90%C.L.),corresponding to the upper limits on the effective Majorana neutrino mass:<3.2–7.5 eV.

Dwarfing is a pivotal agronomic trait affecting both yield and *** species exhibit substantial variation in plant height,among which internode length is a core ***,the molecular mechanism governing internode elongation remains ***,we unveiled that the transcriptional cascade consisting of B-BOX DOMAIN PROTEIN 22(BBX22)and ELONGATED HYPOCOTYL 5(HY5)finely tunes plant height and internode elongation in ***-of-function mutations of BBX22 in an early-flowering citrus(Citrus hindsii“SJG”)promoted internode elongation and reduced pigment accumulation,whereas ectopic expression of BBX22 in SJG,sweet orange(***),pomelo(***)or heterologous expression of BBX22 in tomato(Solanum lycopersicum)significantly decreased internode ***,exogenous application of gibberellin A3(GA3)rescued the shortened internode and dwarf phenotype caused by *** experiments revealed that BBX22 played a dual role in regulation internode elongation and pigmentation in *** the one hand,it directly bound to and activated the expression of HY5,GA metabolism gene(GA2 OXIDASE 8,GA2ox8),carotenoid biosynthesis gene(PHYTOENE SYNTHASE 1,PSY1)and anthocyanin regulatory gene(Ruby1,a MYB DOMAIN PROTEIN).On the other hand,it acted as a cofactor of HY5,enhancing the ability of HY5 to regulate target genes ***,our results reveal the critical role of the transcriptional cascade consisting of BBX22 and HY5 in controlling internode elongation and pigment accumulation in *** the crosstalk regulatory mechanism between internode elongation and fruit pigmentation provides key genes for breeding of novel types with both dwarf and health-beneficial fortification in citrus.

A Study of the Stability Properties in Simulation of Wave Propagation with SPH Method

When ordinary Smoothed Particle Hydrodynamics （SPH） method is used to simulate wave propagation in a wave tank, it is usually observed that the wave height decays and the wave length elongates along the direction of wave propagation. Accompanied with this phenomenon, the pressure under water decays either and shows a big oscillation simultaneously. The reason is the natural potential tensile instability of modeling water motion with ordinary SPH which is caused by particle negative stress in the computation. I＇o deal with the problems, a new sextic kernel function is proposed to reduce this instability. An appropriate smooth length is given and its computation criterion is also suggested. At the same time, a new kind dynamic boundary condition is introduced. Based on these improvements, the new SPH method named stability improved SPH （SISPH） can simulate the wave propagation well. Both the water surface and pressure can be well expressed and the oscillation of pressure is nearly eliminated. Compared with other improved methods, SISPH can truly reveal the physical reality without bringing some new problems in a simple way.

为了深入了解防风多糖能否在胃液中代谢及代谢情况,该文以防风多糖的人工胃液代谢产物为研究对象,结合斑马鱼免疫活性评价,采用高效液相色谱-离子阱-飞行时间质谱(HPLC-IT-TOF-MS)技术系统分析了防风多糖的人工胃液代谢产物。根据所获得的精确相对分子质量、色谱保留行为、质谱裂解规律、对照品比对以及相关文献报道,共分析鉴定了19种代谢产物。其中,代谢产生的单糖5种,寡糖14种。胃液代谢产物中可以准确鉴定出多种还原糖,包括甘露糖、葡萄糖、鼠李糖以及木糖。斑马鱼药效评价结果显示,防风多糖经过胃液代谢后仍具有较好的免疫活性,W3(人工胃液代谢2 h)提高免疫细胞密度的作用最为显著。其中,胃液代谢物M1和M3(Hex-Hex-Man)可能与药理活性关系最为密切,可将其作为潜在的活性片段进行进一步研究。

Effects of oat β-glucan–based prebiotic blended formulas on defecation and gut microbiota in Chinese adults: a randomized clinical trial

Digestive health is important for overall well-being， but gut health problems are common in the Chinese population. The objective of this study was to investigate the effects of oat β-glucan–based prebiotic blended formulas in humans on digestive health by assessing defecation frequen-cy， defecation symptoms， stool quality， self-reported digestive feelings， defecation satisfaction， and gut bacteria. A parallel， randomized， double-blind， controlled trial was conducted in 168 eli-gible adults randomized to three groups for 14 days: Intervention Groups A and B received two different doses of oat β-glucan–based prebiotic blended formulas and a Control Group received oat β-glucan mixed with hydroxypropylmethyl-cellulose and microcrystalline cellulose. Partici-pants completed a defecation diary and Bristol stool chart daily and digestive system question-naires weekly. Fecal samples were collected on Day 0 and Day 14 to evaluate gut bacteria using 16S rDNA analysis. Both formulas improved defecation frequency， stool quality， defecation symptoms， defecation satisfaction， and on digestive health of human， compared with baseline (P < 0.05). Both formulas led to increased quantities of Bifidobacterium and Akkermansia bacteria， compared to control， on Day 14. This study therefore demonstrated the beneficial effects of oat β-glucan–based prebiotic blended formulas on gut health in generally healthy Chinese adults. These formulas are a feasible nutritional strategy for digestive health.
