Meiosis is a highly complex process significantly influenced by transcriptional ***,studies on the mechanisms that govern transcriptomic changes during meiosis,especially in prophase I,are ***,we performed single-cell ATAC-seq of human testis tissues and observed reprogramming during the transition from zygotene to pachytene *** event,conserved in mice,involved the deactivation of genes associated with meiosis after reprogramming and the activation of those related to spermatogenesis before their functional ***,we identified 282 transcriptional regulators(TRs)that underwent activation or deactivation subsequent to this *** suggested that physical contact signals from Sertoli cells may regulate these TRs in spermatocytes,while secreted ENHO signals may alter metabolic patterns in these *** results further indicated that defective transcriptional reprogramming may be associated with non-obstructive azoospermia(NOA).This study revealed the importance of both physical contact and secreted signals between Sertoli cells and germ cells in meiotic progression.

Background This study aimed to identify survival risk factors in Chinese children with hepatoblastoma(HB)and assess the effectiveness of the new treatment protocol proposed by the Chinese Children's Cancer Group(CCCG)in *** A multicenter,prospective study that included 399 patients with HB from January 2015 to june 2020 was *** demographics,treatment protocols,and other related information were *** regression models and Kaplan-Meier curve methods were *** The 4-year event-free survival(EFS)and overall survival(OS)were 76.9 and 93.5%,*** 4-year EFS rates for the very-low-risk,low-risk,intermediate-risk,and high-risk groups were 100%,91.6%,81.7%,and 51.0%,*** 4-year 0S was 100%,97.3%,94.4%,and 86.8%,*** regression analysis found that age,tumor rupture(R+),and extrahepatic tumor extension(E+)were independent prognostic factors.A total of 299 patients had complete remission,and 19 *** with declining alpha-fetoprotein(AFP)>75%after the first two cycles of neoadjuvant chemotherapy had a better EFS and OS than those≤75%.Conclusions The survival outcome of HB children has dramatically improved since the implementation of CCCG-HB-2016 ***≥8 years,R+,and E+were independent risk factors for *** with a declining AFP>75%after the first two cycles of neoadjuvant chemotherapy had better EFS and OS.

生菜是人们经常食用的蔬菜之一,生菜的储藏时间是影响生菜新鲜程度的重要因素。所以研究一种简单、快速、非破坏性的生菜储藏时间的鉴别方法是非常必要的。近红外光谱(NIR)分析能快速和准确的获取生菜的近红外光谱,从而实现无损鉴别生菜储藏时间。但是生菜的NIR数据中存在噪声信号和冗余信号,为了消除光谱的噪声信号并提取特征信息,提出了一种基于模糊非相关QR分析(FUQRA)的近红外光谱生菜储藏时间鉴别新方法。首先,需要降低原始NIR数据的维数,通过使用主成分分析(PCA)将包含1557个维度的光谱数据降至包含22个维度。然后通过模糊非相关判别转换(FUDT)计算出特征向量,利用特征向量建立鉴别向量矩阵,并进行QR分解,得到最终的鉴别向量矩阵。最后以60个新鲜生菜样本为研究样本,使用K近邻(KNN)方法进行分类,用AntarisⅡ型NIR光谱仪对生菜样品进行近红外光谱检测和数据收集。实验过程中每隔12小时对每个样本进行3次重复检测,将这些数据取平均值作为实验数据。随后利用多元散射校正(MSC)减少近红外光谱中的噪声信号。为了验证所提出方法的有效性,分别将主成分分析(PCA)结合KNN、主成分分析和模糊线性判别分析(FLDA)结合KNN、主成分分析和模糊非相关判别转换(FUDT)结合KNN以及主成分分析和模糊非相关QR分析(FUQRA)结合KNN四种算法分析结果进行比较。将权重指数m的不同取值产生的分类准确率进行比较,选出最合适的权重指数和KNN的参数K:m=2,K=3。最终得到的分类准确率分别为43.33%、96.67%、96.67%和98.33%。可以看出,相比其他三个算法,模糊非相关QR分析可以更好地实现对生菜储藏时间的鉴别。

To the Editor:Major depressive disorder(MDD)is a mood disorder characterized by complex patterns of emotional,cognitive,and behavioral symptomology and deficits in daily *** symptoms,including a reduction in libido and menstrual disturbances,have been considered to be a classic symptom of MDD for many *** evidence has reached a broad consensus that the incidence of genital symptoms is higher in patients with MDD than in the general population.A systematic review and meta-analysis found a bidirectional association between MDD and genital symptoms,with patients with MDD showing a 50–70%increased risk of developing genital symptoms,while individuals with genital symptoms had a 130–210%increased risk of developing MDD.^([1])As previously reported,50–70%of people with MDD experience sexual dysfunction.^([2])To date,few studies have focused on the comparison of clinical features between patients with MDD with and without genital symptoms,and the longitudinal prognosis.

Background Pediatric antineutrophil cytoplasmic antibody-associated vasculitis(AAV)is a life-threatening systemic vasculitis featured by liability to renal ***,there are few studies on the risk factors and predictive models for renal outcomes of AAV in *** Data from 179 AAV children in multiple centers between January 2012 and March 2020 were collected *** risk factors and predictive model of end-stage renal disease(ESRD)in AAV were *** Renal involvement was the most typical manifestation(95.5%),and the crescent was the predominant pathological lesion(84.9%).The estimated glomerular filtration rate(eGFR)was evaluated in 114 patients,of whom 59.6%developed ESRD,and the median time to ESRD was 3.20 *** eGFR[P=0.006,odds ratio(OR)=0.955,95%confidence interval(CI)=0.924–0.987]and the percentages of global glomerulosclerosis(pGGS;P=0.018,OR=1.060,95%CI=1.010–1.112)were independent risk factors for ESRD of renal *** on the pGGS and eGFR at renal biopsy,we developed three risk grades of ESRD and one predictive *** Kaplan‒Meier curve indicated that renal outcomes were significantly different in different risk grades(P china,of which more than half deteriorates into *** predictive model based on eGFR at renal biopsy and the pGGS may be stable and accurate in speculating the risk of ESRD in AAV children.

Alzheimer's disease(AD) is a neurodegenerative disease and a major threat to human health worldwide. The association between aluminum exposure and AD has been widely reported. Owing to the ubiquitous presence of aluminum in daily life, aluminum exposure can easily occur whenever and wherever possible. Thus, a rapid and sensitive reagent for detecting aluminum and assist in AD daily prevention for potential AD patient population is extremely needed. However, existing aluminum detection methods rely on precise instruments, which are impractical for household use. Herein, a series of aggregation-induced emission-based covalent-organic framework(AIE-COF) fluorescent probes has been designed with progressively tuned sizes and screened for aluminum detection. Among them, COF-N2 was found to have the highest response towards aluminum specifically, with a fluorescence intensity change of 19.14 times before and after chelation, which could determine the aluminum concentration by naked eye. Then, the molecular mechanism of COF-N2 fluorescence changes was explained and COF-N2 was used for both diagnose the aluminum distribution in various organs of APP/PS1 transgenic mice and quickly determine the aluminum content in daily necessities. The use of AIE-COF probes for aluminum detection provides a promising avenue for developing aluminum related AD clinical diagnosis and daily prevention tools.
