Wogonin(WOG)has been demonstrated to have anti-cancer activity,but the mechanisms remain unclear.In this study,new targets of WOG were predicted for lung cancer,bladder cancer,and colon cancer by using bioinformatics methods.wOG might primarily suppress cancers via regulating arachidonic acid,Ras,MAPK,linoleic acid,PI3K Akt,and folate biosynthesis pathways.3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium(MTS)assay showed that WOG inhibited the proliferation of A549 cells.Real-time quantitative reverse transcription PCR(RT-qPCR)results indicated that anti-lung cancer effect of WOG was achieved by regulating the expression of 18 target genes,including AKRIBI0,AKRIC3,BDNF,CAVI,CXCL2,CYP2B6,CYP4F3,DAO,EGF,ENO3,IL6,PLA2GlB,PLA2G2F,PLA2G4A,PTGES,SLCOIBI,SLCOIB3,and TFAP2A.The Kaplan-Meier survival curves further confirmed that DAO,PLA2G1B,SLCO1B3 and TFAP2A were essential targets via which WOG affected lung cancer survival.Moreover,BDNF,FGF2,and PTGS1 were predicted to be the targets via which WOG alleviated cancer proliferation and invasion in bladder cancer.As for colon cancer,WOG might induce autophagy and inhibit proliferation by down-regulating NTF4 and TH.The study will provide clue for using wOG as a promising antineoplastic agent in basic and translational research,and bring light to the application of herbs containing WOG as food supplements.

Vascular endothelial growth factor and its mimic peptide KLTWQELYQLKYKGI(QK)are widely used as the most potent angiogenic factors for the treatment of multiple ischemic diseases.However,conventional topical drug delivery often results in a burst release of the drug,leading to transient retention(inefficacy)and undesirable diffusion(toxicity)in vivo.Therefore,a drug delivery system that responds to changes in the microenvironment of tissue regeneration and controls vascular endothelial growth factor release is crucial to improve the treatment of ischemic stroke.Matrix metalloproteinase-2(MMP-2)is gradually upregulated after cerebral ischemia.Herein,vascular endothelial growth factor mimic peptide QK was self-assembled with MMP-2-cleaved peptide PLGLAG(TIMP)and customizable peptide amphiphilic(PA)molecules to construct nanofiber hydrogel PA-TIMP-QK.PA-TIMP-QK was found to control the delivery of QK by MMP-2 upregulation after cerebral ischemia/reperfusion and had a similar biological activity with vascular endothelial growth factor in vitro.The results indicated that PA-TIMP-QK promoted neuronal survival,restored local blood circulation,reduced blood-brain barrier permeability,and restored motor function.These findings suggest that the self-assembling nanofiber hydrogel PA-TIMP-QK may provide an intelligent drug delivery system that responds to the microenvironment and promotes regeneration and repair after cerebral ischemia/reperfusion injury.

A precise and reliable analytical method of high performance liquid chromatography-tandem mass spectrometry(HPLCMS/MS)was developed to measure trace levels of enrofloxacin(ENR)and its major metabolite ciprofloxacin(CIP)in carp tissues.Optimized chromatographic separation was obtained on a Waters Xterra MS C_(18) reversed-phase column using gradient elution with methanol and 0.1%formic acid aqueous solution including 5mmolL^(-1) of ammonium acetate.The established method was applied to study the pharmacokinetics and distribution of ENR and CIP in tissues of carp following a single oral administration in feed at a dosage of 40mgkg^(-1) bw(body weight).Data were analyzed using DAS 2.0 dynamics software,and the experimental results suggest that ENR was rapidly absorbed and extensively distributed in carp tissues through systemic circulation,and the pharmacokinetic characteristics can be described with a two-compartment model.The elimination half-lives(t_(1/2β))from muscle,liver,gill,plasma and skin were 131,160,104,132 and 310 h,respectively.The areas under the drug concentration-time curves(AUC)for these tissues were 491,972,750,249 and 706hmgkg^(-1),respectively.The maximum concentration(C_(max))values were 13,29,37,9 and 5mgkg^(-1) with peak times(t_(max))of 8,4,4,2 and 4 h,respectively.Ciprofloxacin,the active metabolite of ENR,was also detected in carp tissues,indicating that only 1.54%of de-ethylation of ENR occurs in carp.At a water temperature of 18℃,the drug withdrawal time was determined to be no less than 24 d while the carp was fed at a single dosage of 40mgkg^(-1).

Molecular profiling of cell-surface proteins is a powerful strategy for precise cancer diagnosis.While mass cytometry(MC)enables synchronous detection of over 40 cellular parameters,its full potential in disease classification is challenged by the limited types of recognition probes currently available.In this work,we synthesize a panel of heavy isotopeconjugated aptamers to profile cancer-associated signatures on the surface of hematological malignancy(HM)cells.Based on 15 molecular signatures,we performed cell-surface profiling that allowed the precise classification of 8 HM cell lines.Combined with machine-learning technology,this aptamer-based MC platform also achieved multiclass identification of HM subtypes in clinical sampleswith 100%accuracy in the training cohort and 80%accuracy in the test cohort.Therefore,we report an effective and practical strategy for precise cancer classification at the singlecell level,paving the way for its clinical use in the near future.

采用水泥基复合材料(engineering cementitious composite,ECC)替代混凝土可以提高结构在偶然荷载作用下的抗连续倒塌性能,但在悬链线大变形阶段钢筋与ECC间可能会出现黏结滑移破坏。基于分离式霍普金森压杆(SHPB)试验装置,进行了钢筋与ECC的动态黏结滑移性能试验,分析了ECC强度等级、应变率、钢筋直径对极限黏结强度、刚度和滑移量的影响规律,获得了高应变率下钢筋与ECC的平均黏结滑移曲线,得到其黏结滑移破坏模式。并与静态黏结滑移试验进行对比,得到了动态黏结强度增强因子。进一步,通过钢筋开槽内贴应变片法,获得不同锚固位置的黏结应力及相对滑移的分布规律,提出黏结位置函数。最后,根据试验结果得到钢筋与ECC平均黏结滑移本构,进而乘以黏结位置函数得到考虑锚固位置影响的动态黏结滑移本构,为ECC结构构件设计及有限元分析提供试验依据和理论参考。

Nypa fruticans(Wurmb),a mangrove palm species with origins dating back to the Late Cretaceous period,is a unique species for investigating long-term adaptation strategies to intertidal environments and the early evolution of palms.Here,we present a chromosome-level genome sequence and assembly for N.fruticans.We integrated the genomes of N.fruticans and other palm family members for a comparative genomic analysis,which confirmed that the common ancestor of all palms experienced a whole-genome duplication event around 89 million years ago,shaping the distinctive characteristics observed in this clade.We also inferred a low mutation rate for the N.fruticans genome,which underwent strong purifying selection and evolved slowly,thus contributing to its stability over a long evolutionary period.Moreover,ancient duplicates were preferentially retained,with critical genes having experienced positive selection,enhancing waterlogging tolerance in N.fruticans.Furthermore,we discovered that the pseudogenization of Early Methionine-labelled 1(EM1)and EM6 in N.fruticans underly its crypto-vivipary characteristics,reflecting its intertidal adaptation.Our study provides valuable genomic insights into the evolutionary history,genome stability,and adaptive evolution of the mangrove palm.Our results also shed light on the long-term adaptation of this species and contribute to our understanding of the evolutionary dynamics in the palm family.

为开发既具有电热性能又满足对三维表面具有一定适应性的电加热织物,采用定制纤维铺放(TFP)工艺设计制备直线形、正弦波形、锯齿形和尖角形4种镍铬合金丝排布方式的电加热织物,研究电加热织物的电热性能和半球成型性能。结果表明:10 V直流电压下通电30 s,4种电加热织物表面最高平衡温度分别达到159.5、92.8、66.7和31.5℃,温度分布均匀;反作用力与镍铬合金丝排布方式密切相关;半球成型最终状态下,直线形电加热织物成型反作用力最小,表面褶皱和细观缺陷最少,且最大面内剪切角为32.26°;与未引入镍铬合金丝玻璃纤维基底织物相近,即直线形电加热织物半球成型性优于正弦波形、锯齿形和尖角形电加热织物。

基于种群生态学理论,将黄河流域971个国家级特色保护类村庄视作一个地理种群。综合GIS空间分析和数理统计方法,明晰这一种群的分布动态和生境选择特征,进而应用地理探测器对比揭示影响该种群分布的生态因子。结果表明:(1)黄河流域特色保护类村庄的分布密度随种群增长而大幅提高,分布模式随尺度变大由显著聚集转为不显著离散,分布格局随时间推移由多中心向单中心演化,但集聚中心一直位于河谷盆地;(2)黄河流域特色保护类村庄具有“聚中低山区、向阳坡斜坡、喜温暖湿润、近黄河干流”的偏好,趋向中等人口密度、经济欠发达地区分布,主要位于国道沿线30 km、省道沿线10 km、中心城市周边20~80 km、历史城镇周边60 km、旅游景区和文保单位周边20 km范围内;(3)影响特色保护类村庄种群分布的生态因子具有综合性、非等价性和限定性,在黄河流域以文保单位和海拔为主导。

为通过微胶囊化提高螺旋藻多酚的稳定性。利用玉米醇溶蛋白(Zein)和多孔淀粉(porous starch,PS)对螺旋藻多酚(polyphenol,Phe)进行包埋,制备了螺旋藻多酚-多孔淀粉(Phe-PS),螺旋藻多酚-多孔淀粉-玉米醇溶蛋白(Phe-PS-Zein)两种微胶囊,并对Phe,Phe-PS和Phe-PS-Zein进行了微观结构和稳定性、抗氧化性及体外模拟消化性质的比较。结果表明,Phe-PS微胶囊在芯壁比1:10、温度80℃、磁力搅拌时间120 min条件下,包埋率为60.29%;Phe-PS-Zein微胶囊在芯壁比1:10、壁材比1:1、搅拌时间90 min、温度60℃和pH7时包埋率最高,达86.93%。XRD和XPS图谱结果表明,添加Zein后,晶体衍射峰强度有所降低,主要与Zein的无定型结构有关。DSC分析结果表明,蛋白做壁材改善了微胶囊热稳定性。通过对抗氧化性、稳定性以及体外缓释效果的测定发现,与游离Phe和Phe-PS相比,Phe-PS-Zein对DPPH自由基清除力和Fe3+的还原力提高。综上,Phe-PSZein颗粒可以作为螺旋藻多酚的有效递送载体。本研究结果有望为Phe的高值化利用提供理论和实验基础。
