The mechanism of action of clearing agents to improve optical imaging of mouse skin during reflectance-mode confocal microscopy was *** dermal side of excised dorsal mouse skin was exposed for one hour to saline,glycerin,or 80% DMSO,then the clearing agent was removed and the dermis placed against a glass cover slip through which a confocal microscope measured reflectance at 488 nm *** untreated control was also *** axial attenuation of reflectance signal,R(z_(f))versus increasing depth of focus zf behaved as R=ρexp(−μz_(f) 2G),where ρ is tissue reflectivity and μ is attenuation[cm^(−1)].The factor 2G accounts for the in/out path of photons,and the numerical aperture of the *** ρ,μ data were mapped to values of scattering coefficient(μ_(s)[cm^(−1)])and anisotropy of scattering(g).Images showed that glycerin significantly increased the g of dermis from about 0.7 to about 0.99,with little change in the μ_(s) of dermis at about 300cm^(−1).DMSO and saline had only slight and inconsistent effects on g and μ_(s).

This work was carried out with the aim of contributing to the treatment of cancer. Cancer is one of the most common causes of death. It constitutes a public health problem. Photodynamic therapy (PDT) is one treatment option. This study contributes to the search for photosensitizing molecules used in PDT. Makaluvamines have shown interesting properties in the treatment of several human cancer cell lines. The present study analyzes the ultraviolet and visible absorption spectroscopic properties of a few Makaluvamines. These have been listed in the literature and can be in neutral or charged states (protonated and methylated). The investigation is based on quantum chemical calculations. Molecular geometries and vibrational frequencies have been calculated at the B3lYP/6-311++G(d,p) level. Absorption properties in the visible and ultraviolet spectral range are measured on optimized structures using time-dependent density functional theory (TD-DFT). The absorption spectra are obtained using the “Chemissian” software. The results of our calculations have allowed us to determine the absorption zones of the molecules studied, the energy gaps of the frontier orbitals, the main transitions associated with the absorption process, and their lifetimes. They have also identified four Makaluvamines (E, G, M, and l) that absorb in the therapeutic domain and may have photosensitizer properties.

Introduction: Arbovirus diseases such as dengue and chikungunya threaten public health worldwide. Early and rapid diagnosis and surveillance of dengue virus (DENV) and chikungunya virus (CHIKV) infections are essential to the control of these diseases. In this study, we evaluate the diagnostic performance of our new in-house multiplex RT-qPCR method for detecting DENV serotypes and CHIKV in an external laboratory. Methodology: The evaluation study was conducted on 200 clinical samples of suspected patients for arbovirus disease infection, collected in Centre de Recherche Biomoléculaire Pietro Annigoni (CERBA), Ouagadougou, Burkina Faso. Our new multiplex RT-qPCR was compared to the commercial kit, the Zika, Dengue, and Chikungunya (ZDC) Real-Time PCR Assays kit (Bio-Rad, California, USA). Results and Conclusions: Among 200 samples, 21.5% (43/200) were DENV-positive by multiplex RT-qPCR, and 21.5% (43/200) were also DENV-positive by reference real-time RT-PCR. 157 (78.5%) samples tested negative for DENV by both tests (new mRT-qPCR and reference test). The sensitivity and specificity of mRT-qPCR were 100%. The DENV serotypes detected were DENV-1 60.5% (26/43) and DENV-3 39.5% (17/43). CHIKV was not detected in this study. Our new mRT-qPCR is sensitive, cost-effective, simple, and can be used in developing country laboratories.

At peak granulite-facies metamorphic conditions, lower continental crust is arguably fluxed by large amounts of two key low water activity fluids：（i） high-density CO2 and/or （ii） concentrated saline so- lutions. These fluids are either internally-derived, generated by mineral reactions or dehydration melting or, notably for CO2, externally-derived, issued from the underlying mantle. Postmetamorphic evolution results in complete disappearance of these fluids, except for minute remnants preserved in minerals as fluid inclusions. Two major processes are involved：（i） at peak conditions, granitoid magmas form, migrate upward, and crystallize as shallow intrusions in the upper crust （mineralized porphyry types or reduced intrusions）; （ii） during the rapid decompression which almost systematically follows a period of post-peak isobaric cooling, especially for ultrahigh-temperature granulites （anticlockwise P-T paths）, quartz-carbonate megashear zones are formed by repeated periods of seismic activity. Seismic activity may continue until all free fluids have disappeared, resulting in the ultramylonites and pseudotachylites that are found in many granulite domes. A great majority of vein-type Au deposits worldwide occur in the above-mentioned settings or nearby. We suggest that the Au has been scavenged by the granulite fluids, then redistributed and concentrated during the formation of veins and related phenomena.

This paper reviews the origin and evolution of fluid inclusions in ultramaflc xenoliths, providing a framework for interpreting the chemistry of mantle fluids in the different geodynamic settings. Fluid inclusion data show that in the shallow mantle, at depths below about 100 km, the dominant fluid phase is CO2 ± brines, changing to alkali-, carbonate-rich （silicate） melts at higher pressures. Major solutes in aqueous fluids are chlorides, silica and alkalis （saline brines; 5-50 wt.% NaCl eq.）. Fluid inclusions in peridotites record CO2 fluxing from reacting metasomatic carbonate-rich melts at high pressures, and suggest significant upper-mantle carbon outgassing over time. Mantle-derived CO2 （±brines） may eventually reach upper-crustal levels, including the atmosphere, independently from, and additionally to magma degassing in active volcanoes.
