A dose-escalation and expansion,phase 1/2 study(***,NCT04818333)was conducted to assess the novel antibodydrug conjugate SHR-A1811 in pretreated HER2-altered advanced non-small cell lung cancer(NSCLC).Here,we report results from the phase 1 *** who had previously failed or were intolerant to platinum-based chemotherapy were enrolled and received SHR-A1811 intravenously at doses of 3.2 to 8.0 mg/kg every 3 *** escalation followed a Bayesian logistic regression model that included overdose control,with subsequent selection of tolerable levels for dose ***,63 patients were enrolled,including 43 receiving a recommended dose for expansion of 4.8 mg/*** patients had HER2-mutant ***-limiting toxicity occurred in one patient in the 8.0 mg/kg dose ***≥3 treatment-related adverse events occurred in 29(46.0%)*** patient in the 6.4 mg/kg cohort died due to interstitial lung *** of April 11,2023,the 4.8 mg/kg cohort showed an objective response rate of 41.9%(95%Cl 27.0-57.9),and a disease control rate of 95.3%(95%Cl 84.2-99.4).The median duration of response was 13.7 months,with 13 of 18 responses *** median progression-free survival was 8.4 months(95%CI 7.1-15.0).SHR-A1811 demonstrated favourable safety and clinically meaningful efficacy in pretreated advanced HER2-mutantNSCLC.

The crystal and molecular structure of the complex containing cobalt-carbon and iron- sulfur cluster cores,(μ-ρ-CH_3C_6H_4C_2S)(μ-n-C_3H_7S)Fe_2(CO)_6Co_2(CO)_6,has been determined by X-ray diffraction *** crystals are triclinic,space group P,with a=9.139(2),b=9.610(1), c=17.183(2),α=84.36(1),β=89.45(1),γ=88.15(1)°,V=1501.0~3;Z=2,D_c=1.74 g/cm^3. R=0.072,Rw=*** results of the structure determination show a cobalt-carbon cluster core formed through the reaction of(μ-ρ-CH_3C_6H_4C_2S)(μ-n-C_3H_7S)Fe_2(CO)_6 with Co_2(CO)_*** the cobalt-carbon cluster core,the bond length of the original C≡C lengthened to 1.324 which is close to the typical value of carbon-carbon double *** groups connecting the carbons of the cluster core are in cis position and lie on the opposite side of cobalt *** this complex,the conformation of-SC_3H_7 is e-type,while that of-SC_2C_6H_4CH_3 is a-type.

Objective:To study the effects of Yishen Qutong granula on pain sensitization and bone destruction of rats with bone cancer ***:Walker256 cells were passaged in ascites and injected into the tibia of female Wistar rats to prepare the bone cancer pain *** the 14th day after model establishment,60 rats were randomly divided into model group,sham-operated group,Yishen Qutong granula low,middle,high dose group and tramadol hydrochloride positive control *** continuous administration for 14 days,the mechanical pain threshold,thermal threshold and weight-bearing difference of both hind limbs were ***:Compared with the model group,Yishen Qutong groups increased the mechanical pain threshold,thermal pain threshold and reduced the weight difference of both hind limbs(P0.05),and Yishen Qutong granula significantly reduced the weight-bearing difference of hind limbs in all groups(Plieve pain sensitization and alleviate bone damage in rats with bone cancer pain.

OBJECTIVE To investigate the available parameters in gynecological screening for cervical lesions by liquid-based cytology technology （ThinPrep Cytology Test, TCT） and The Bethesda System （TBS）, also with computer image analysis. METHODS With application of the image analysis system, all grades of cervical lesion cells were detected quantitatively and sorted in atypical squamous cells of undetermined significance （ASCUS）, atypical squamous cells-cannot exclude HSIL （ASC-H）, low-grade squamous intraepithelial lesion （LSIL）, high-grade squamous intraepithelial lesion （HSIL） and cervical squamous cell carcinoma （SCC） with the mean optical density （MOD）, average grey （AG）, positive units （PU）, and nucleus to cytoplasmic ratio （N： C）. Differences between each group of cells were compared and analyzed statistically. RESULTS Apart from four stereologic parameters in LSIL and HSIL groups there were no differences among them, in the other groups, there was statistically significant in differences between MOD, AG and PU values. Differences between them in the ratio of nucleus to cytoplasm were highly statistically significant. CONCLUSION Stereological indexes may serve as a screening tool for cervical lesions. The image analysis system is expected to become a new means of cytological assisted diagnosis.

Aerobic anoxygenic phototrophic bacteria(AAPB)are significant bacterial groups in aquatic ecosystems,known for their rapid growth and photoheterotrophic ***,the distribution and ecological assembly process of AAPB in low irradiation freshwater basins remain unclear,warranting further *** this study,we present the diversity,abundance,spatial variations,ecological process,and community interaction of AAPB in sediment of Three Gorges Reservoir(TGR)under low *** findings demonstrate the dominant genera of AAPB community that exist in the TGR area also are appeared in different waters,with some regional ***,the concentration of pufM gene,an indicator for AAPB,maintains a consistently high numerical level ranging from(2.21±0.44)×10^(4)to(9.98±0.30)×10^(7)gene copies/*** solar irradiation is suggested as the major factor affecting AAPB,it remains unclear whether and how AAPB differ between regions due to varying solar irradiation *** results show spatial differences between total bacteria and AAPB communities,with significant differences observed only in *** and environmental factor contributed less than 10%to the spatial difference of community,with sediment type and environmental factors being the key factors influencing microbial community *** stochastic process plays a dominant role in the aggregation and replacement of AAPB communities,among which the most contribution is dispersal *** AAPB network,Yoonia and Gemmobacter are the hubs for *** results valuable insights into the AAPB communities in TGR with low irradiation.

Accurate and efficient online parameter identification and state estimation are crucial for leveraging digital twin simulations to optimize the operation of near-carbon-free nuclear energy *** previous studies,we developed a reactor operation digital twin(RODT).However,non-differentiabilities and discontinuities arise when employing machine learning-based surrogate forward models,challenging traditional gradient-based inverse methods and their *** study investigated deterministic and metaheuristic algorithms and developed hybrid algorithms to address these *** efficient modular RODT software framework that incorporates these methods into its post-evaluation module is presented for comprehensive *** methods were rigorously assessed based on convergence profiles,stability with respect to noise,and computational *** numerical results show that the hybrid KNNLHS algorithm excels in real-time online applications,balancing accuracy and efficiency with a prediction error rate of only 1%and processing times of less than 0.1 ***,algorithms such as FSA,DE,and ADE,although slightly slower(approximately 1 s),demonstrated higher accuracy with a 0.3%relative L_2 error,which advances RODT methodologies to harness machine learning and system modeling for improved reactor monitoring,systematic diagnosis of off-normal events,and lifetime management *** developed modular software and novel optimization methods presented offer pathways to realize the full potential of RODT for transforming energy engineering practices.

Accurate and sensitive strategies for Concanavalin A(Con A)sensing are conducive to the better cognition of various important biological and physiological ***,by designing dextran-functionalized fluorescent microspheres(DxFMs)and boric acid-modified carbon dots(BCDs)as recognition unit and built-in signal reference respectively,a ratiometric fluorescent detection platform was proposed for Con A detection with high *** this protocol,the BCDs/DxFMs precipitation was formed due to the covalent interactions between cis-diol of DxFMs and boronic acid groups of BCDs,thus only fluorescence of BCDs could be detected in the *** Con A was presented,it could bind to DxFMs through its carbohydrate recognition ability and suppress the subsequent assembly between DxFMs and BCDs,leading to the simultaneous capture of DxFMs and BCDs fluorescence in the *** the BCDs content was superfluous,their fluorescence intensities were basically constant in all *** on the unchanged BCDs fluorescence signal and target-dependent DxFMs fluorescence signal in supernatant,the ratiometric detection of Con A was *** optimized conditions,this ratiometric fluorescent platform displayed a linear detection range from 0.125μg/mL to 12.5μg/mL with a detection limit of 0.089μg/***,satisfied analytical outcomes for Con A detection in serum samples were obtained,manifesting huge application potential of this ratiometric fluorescent platform in clinical diagnosis.
