Bacterial-based antitumor immunity has become a promising strategy to activate the immune system for fighting ***,the potential application of bacterial therapy is hindered by the presence of instability and susceptibility to infections within bacterial ***,monotherapy is ineffective in completely eliminating complex cancer with multiple contributing *** this study,based on our discovery that spore shell(SS)of Bacillus coagulans exhibits excellent tumor-targeting ability and adjuvant activity,we develop a biomimetic spore nanoplatform to boost bacteria-mediated antitumor therapy,chemodynamic therapy and antitumor immunity for synergistic cancer *** detail,SS is separated from probiotic spores and then attached to the surface of liposome(Lipo)that was loaded with hemoglobin(Hb),glucose oxidase(GOx)and JQ1to construct SS@Lipo/Hb/GOx/*** tumor tissue,highly toxic hydroxyl radicals(·OH)are generated via sequential catalytic reactions:GOx catalyzing glucose into H_(2)O_(2)and Fe^(2+)in Hb decomposing H_(2)O_(2)into·*** combination of·OH and SS adjuvant can improve tumor immunogenicity and activate immune ***,JQ1-mediated down-regulation of PD-L1 and Hb-induced hypoxia alleviation synergistically reshape immunosuppressive tumor microenvironment and potentiate immune *** this manner,SS@Lipo/Hb/GOx/JQ1 significantly suppresses tumor growth and *** summarize,the nanoplatform represents an optimum strategy to potentiate bacteria-based cancer immunotherapy.

The convenient synthesis of the composite electrode with high supercapacitance performance plays an important role in practical application but is ***,the carbon nanotubes(CNTs)coupled with lowcrystalline sulfur and nitrogen co-doped Ni Co-LDH(denoted as SN-Ni Co-LDH)nanosheets array are grown on Ni Co foam(NCF)substrate by two convenient steps of metal induced self-assembly and corrosion engineering,which present the advantages of operating at roomtemperature and low preparation *** from the S–N co-doping and low-crystallinity of Ni Co-LDH,the prepared SN-Ni Co-LDH@CNTs@NCF electrode presents a topping charge capacity of 2470 C·g^(-1)(4.94 C·cm^(-2))at 5 m A·cm^(-2).Furthermore,the fabricated asymmetry supercapacitor(ASC)achieves an extraordinary energy density of 77 Wh·kg^(-1)(0.617 m Wh·cm^(-2))at a power density of 438 W·kg^(-1)(3.5 m W·cm^(-2))and outstanding stability(91%capacity retention after 5000 cycles at20 m A·cm^(-2)).Impressively,the structure evolution of Ni Co-LDH during the charge/discharge processes has been thoroughly elucidated by in-situ Raman ***,this work verifies a powerful strategy and practical value for preparing composite electrodes with high supercapacitance performance,and also provides guidance for the rational design of the smart electrodes.

Electroacupuncture Promotes Angiogenesis in Mice with Cerebral Ischemia by Inhibiting miR-7

Objective To observe the angiogenesis effect of electroacupuncture(EA)at Shuigou acupoint(GV 26)in the treatment of cerebral ischemia,and explore the value of miRNA-7(miR-7)in *** First,48 mice were randomly divided into sham operation,middle cerebral artery occlusion(MCAO)model,and EA treatment *** 9 mice were divided into carrier control group,miR-7 knockout group and miR-7 overexpression group(n=3 each group).Finally,20 mice were divided into model and carrier control group,model and miR-7 knockout group,EA treatment and carrier control group and EA treatment and miR-7 overexpression group,with 3–6 mice in each *** MCAO model was established in the MCAO and EA *** deficit score and 2,3,5-triphenyltetrazolium chloride(TTC)staining were used to evaluate the severity of cerebral ***-eosin staining was used to describe basic pathological *** was used to quantify cerebral microvessel ***-time PCR and Western blot were used to detect the expression of miR-7 and its downstream target genes Krüppel-like factor 4/vascular endothelial growth factor(KLF4/VEGF)and angiopoietin-2(ANG-2)in the ischemic cerebral *** After EA,neurological deficit scores and infarction volumes decreased,and the density of cerebral microvessels *** the MCAO group,miR-7 expression was higher than that in the sham group(P<0.01).After EA at GV 26,miR-7 expression decreased(P<0.01)and the expression of downstream target genes KLF4/VEGF and ANG-2 increased as compared with the MCAO group(P<0.01).After EA combined with overexpression of miR-7,the expression of downstream target genes KLF4/VEGF and ANG-2 decreased compared to the control EA group(P<0.01).After miR-7 knockdown,the expression of KLF4/VEGF and ANG-2 increased(P<0.05 or P<0.01).Conclusions EA could promote angiogenesis in MCAO mice likely by inhibiting the expression of miR-7 and relieving inhibition of downstream target genes KLF4/VEG

Polycystic ovarian syndrome （PCOS） is the most principal reason for infertility in reproductive women， but no versatile treatment is feasible. Although trans-10-Hydroxy-2-decenoic acid （10H2DA） exhibits the anti-inflammatory， anti-cancerous and immunomodulatory properties， its role in improving PCOS remains elusive. This present study examined the effects of 10H2DA on the estrous cycle， ovarian morphology， reproductive hormone， glucose and insulin tolerances of PCOS mice， analyzed the binding between 10H2DA and free fatty acid receptor 4 （FFAR4）， and explored the regulation of 10H2DA on the c-Jun N-terminal kinase （JNK）/JUN （Jun proto-oncogene） signaling， cytosolic calcium ion (Ca2+)， endoplasmic reticulum （ER） Ca2+release， mitochondrial Ca2+， mitochondrial function， autophagy， lysosomal acidification， mitochondrial fission， secretion function and apoptosis of ovarian granulosa cells （GCs） using enzyme-linked immunosorbent assay， flow cytometry. western blotting， molecular docking， various fluorescent probes or plasmid， etc. The results found that 10H2DA alleviated the symptom of PCOS mice through improving the secretion dysfunction of GCs and protecting GCs against apoptosis. After binding to FFAR4 and inactivation of JNK/JUN signaling， 10H2DA restricted the release of ER Ca2+through targeting ryanodine receptors （RYRs） which had been identified as the downstream targets of JUN， diminished cytosolic Ca2+accumulation and prevented Ca2+influx into mitochondria via adjusting the mitochondrial calcium uptake 1 （MICU1） and mitochondrial calcium uniporter （MCU）. Concurrently， 10H2DA kept the integrity of mitochondrial function through maintaining mitochondrial Ca2+homeostasis and suppressed the discharge of mitochondrial reactive oxygen species （mtROS） into the cytosol through blocking the mitochondrial permeability transition pore opening to avoid the lipid peroxidation and ameliorate the secretory function and apoptosis of GCs.
