Microwave induces apoptosis in A549 human lung carcinoma cell line

Background Microwaves have other biological effects on cancer as well besides killing tumor cells by *** studies showed that microwaves may induce apoptosis in some tumor cells. The apoptotic effect of microwaves may help in clinic to remove residual malignant cells nearby the primary lesion and avoid relapse subsequently. However,there is little evidence on this subject from lung cancer. We studied the effect of microwaves on inducing apoptosis in the human lung carcinoma cell line A549 cells, aiming to identify its effect on *** A549 cells were radiated by various intensities and durations of microwaves. Apoptosis induction in A549 cells was analyzed by morphological observations, tetrazolium blue color method （MTT） assays, flow cytometery,immunohistochemistry, and image *** Morphological changes in A549 cells, including cell shrinking and nuclear pycnosis, were observed after microwave radiation. Microwaves significantly inhibited metabolic activities and induced apoptosis in A549 cells. The results of the MTT assay showed a significant decrease of cell activities in all the radiation groups compared with the normal control （P ＜0.01）. The low point of cell activities often appeared at 6-12 hours after radiation. Apoptosis was also confirmed by flow cytometery. The early stage apoptotic rate reached 6.10%-17.98% and the advanced stage apoptotic rate ＋ necrosis rate reached 8.04%-44.06% at 6 hours after microwave irradiation, in contrast to 2.32% and 4.10% in the respective control groups. Down-regulation of Bcl-2 expression and up-regulation of p53 expression were observed by immunohistochemistry after radiation. In most treated groups, the down-regulation of Bcl-2 expression reached its lowest level at 3-6 hours after radiation （integrated optical density （IOD）-6 hours： 2.13±0.08-5.14±0.13 vs. control： 5.79±0.10, P＜0.01 ） and the up-regulation of P53 expression peaked at about 3 hours （IOD-3 hours： 2.61±0.13-8.07±0.11 vs. c

Heuristic Expanding Disconnected Graph:A Rapid Path Planning Method for Mobile Robots

Existing mobile robots mostly use graph search algorithms for path planning,which suffer from relatively low planning efficiency owing to high redundancy and large computational *** to the limitations of the neighborhood search strategy,the robots could hardly obtain the most optimal global path.A global path planning algorithm,denoted as EDG*,is proposed by expanding nodes using a well-designed expanding disconnected graph operator(EDG)in this ***,all obstacles are marked and their corners are located through the map ***,the EDG operator is designed to find points in non-obstruction areas to complete the rapid expansion of disconnected ***,the EDG*heuristic iterative algorithm is *** selects the candidate node through a specific valuation function and realizes the node expansion while avoiding collision with a minimum *** planning experiments were conducted in a typical indoor environment and on the public dataset *** result shows that the proposed EDG*reduced the planning time by more than 90%and total length of paths reduced by more than 4.6%.Compared to A*,Dijkstra and JPS,EDG*does not show an exponential explosion effect in map *** EDG*showed better performance in terms of path smoothness,and collision *** shows that the EDG*algorithm proposed in this paper can improve the efficiency of path planning and enhance path quality.

Comparative analysis of DNA-SIP and magnetic-nanoparticle mediated isolation (MMI) on unraveling dimethoate degraders

Microorganisms are crucial in the bioremediation of organophosphorus pesticides. However, most functional microorganisms (> 99%) are yet to be cultivated. This study applied two cultivation-independent approaches, DNA-SIP and magnetic-nanoparticle mediated isolation (MMI), to identify the functional microorganisms in degrading dimethoate in agricultural soils. MMI identified five dimethoate degraders: Pseudomonas, Bacillus, Ramlibacter, Arthrobacter, and Rhodococcus, whereas DNA-SIP identified three dimethoate degraders: Ramlibacter, Arthrobacter, and Rhodococcus. Also, MMI showed higher resolution than DNA-SIP in identifying functional microorganisms. Two organic phosphohydrolase (OPH) genes: ophC2 and ophB, were involved in dimethoate metabolism, as revealed by DNA-SIP and MMI. The degradation products of dimethoate include omethoate, O,O,S-trimethyl thiophosphorothioate, N-methyl-2-sulfanylacetamide, O,O-diethyl S-hydrogen phosphorodithioate, O,O,O-trimethyl thiophosphate, O,O,S-trimethyl thiophosphorodithioate, and O,O,O-trimethyl phosphoric. This study emphasizes the feasibility of using SIP and MMI to explore the functional dimethoate degraders, expanding our knowledge of microbial resources with cultivation-independent approaches.

The tip-clearance flow in a pump-jet propulsor exerts great impacts on the fluctuating pressures and resultant unsteady forces,which are important sources of structural vibrations and radiated noise *** blade geometry close to the tip is an important factor determining the vortex strength in the tip-clearance *** the open-water condition,the effects of raking the rotor tips on the duct-surface fluctuating pressures and the resultant unsteady forces acting on different components of the propulsor are investigated via physical model experiments and the numerical solution of Reynolds-averaged Navier-Stokes(RANS)equations coupled with the SST k-ωturbulence *** measured and simulated results of hydrodynamic pressures are consistent to each other,and the simulated flows help better understand why the fluctuating pressures change with the tip *** strong fluctuations of duct-surface pressures are caused by intensive tip separation *** duct-surface pressure fluctuations are effectively reduced by using the rake distribution near the tip towards blade back side and,for the combination of the five-bladed rotor and the seven-bladed stator,the resultant unsteady horizontal(and vertical)forces acting on the duct and stator are also reduced;while increasing rake leads to negative effect on pressure fluctuations and unsteady horizontal(and vertical)forces acting on all the components of the propulsor.

Determination of ice jam thickness—A new approach

In winter,rivers in cold regions often experience flood disasters resulted from ice jams or ice *** of the variation of ice jam thickness and water level during an ice jammed period are not only a practical need for ice prevention to avoid disaster and plan water resource,but also essential for the development of any mathematical model for predicting the evolution of ice *** far,some equations based on the energy equation have been proposed to describe the relationship between ice jam thickness and water ***,in the derivation of these equations,the local head loss coefficient at the ice jam head and the riverbed slope factor were ***,those reported equations cannot be used to preciously describe the flow energy equation with ice jams and accurately calculate the ice jam thickness and water *** the present study,a more comprehensive theoretical model for hydraulic calculation of ice jam thickness has been derived by considering important and essential factors including riverbed slope and local head loss coefficient at the ice jam ***,based on the data collected from laboratory experiments of ice jam accumulation,the local head loss coefficient at the ice jam head has been calculated,and the empirical equation for calculating the local head loss coefficient has been established by considering flow Froude number and the ratio of ice discharge to flow *** results of this study not only provide a new reference for calculating ice jam thickness and water level,but also present a theoretical basis for accurate CFD simulation of ice jams.

In vitro study of safety and co-efficiency of the transbronchial coagulation techniques

Background The usual transbronchial coagulation techniques include microwave, argon plasma coagulation （APC）, electrocautery and cryotherapy. However, there are serious clinical problems in the safety of each. By analyzing the experimental data and clinical observations, we observed the variable effects of different coagulation techniques via bronchofibroscopy, to look for an optimal interventional management of luminal bronchus diseases, and evaluate the safety and the equivalent point. Methods Four kinds of coagulation techniques under bronchoscopy were performed on the fresh bronchus of healthy sheep, and the pathologic changes in all groups were observed under the microscope. The different treatment parameters were as follows： microwave 60 Wxl second, 3 seconds, 5 seconds and 40 W×1 second, 3 seconds, 5 seconds; APC 40 W×1 second, 3 seconds, 5 seconds; electrocautery 40 W×1 second, 3 seconds, 5 seconds; cryotherapy 100 Ω×60 seconds, 120 seconds. Results After treatment, ovine bronchial mucosa in all groups showed pathologic changes such as local necrosis and amotio of the mucosa lining epithelium, local submucosa coagulative necrosis or tissue defects, while inflammation in the surrounding tissue was not obvious. Under the same output power and action time, different methods had different outcomes. The damage by APC was the most superficial, microwave was the second, and electrocautery caused the worst damage. The study also found that effects of electrocautery at 40 W×3 seconds, microwave at 40 W×5 seconds or 60 W×3 seconds, APC at 40 W×5 seconds and cryotherapy at 100 Ω×120 seconds were the equivalent point conditions. The appearance included mucosa absence, partial submucosa absence, and collagen fiber coagulation in treatment areas. Conclusions Each coagulation technique has its own characteristic. It is very important to choose the appropriate power and action time of the suitable method according to the therapy requirement.
