Poplar plantations are often established on nitrogen-poor land,and poplar growth and wood formation are constrained by low nitrogen(LN)***,the molecular mechanisms by which specific genes regulate wood formation in acclimation to LN availability remain ***,we report a previously unrecognized module,basic region/leucine zipper 55(PtobZIP55)–PtoMYB170,which regulates the wood formation of Populus tomentosa in acclimation to LN ***55 was highly expressed in poplar wood and induced by *** wood anatomical properties and increased lignification were detected in PtobZIP55-overexpressing poplars,whereas the opposite results were detected in PtobZIP55-knockout *** and transgenic analyses revealed that PtobZIP55 directly binds to the promoter sequence of PtoMYB170to activate its *** phenotypes of PtoMYB170 transgenic poplars were similar to those of PtobZIP55 transgenic poplars under LN *** molecular analyses revealed that PtoMYB170 directly bound the promoter sequences of lignin biosynthetic genes to activate their transcription to increase lignin concentrations in LN-treated poplar *** results suggest that PtobZIP55 activates PtoMYB170 transcription,which in turn positively regulates lignin biosynthetic genes,increasing lignin deposition in the wood of *** in the context of acclimation to LN availability.

Impact of land use change on carbon storage in the middle reaches of the Yellow River,China

The implementation of long-term shelterbelt programs in the middle reaches of the Yellow River(MRYR),China not only has improved the overall ecological environment,but also has led to the changes of land use pattern,causing carbon storage ***,the relationship between carbon storage and land use change in the MRYR is not concerned,which results in the uncertainty in the simulation of carbon storage in this *** use changes directly affect the carbon storage capacity of ecosystems,and as an indicator reflecting the overall state of land use,land use degree has an important relationship with carbon *** this study,land use data and the integrated valuation of ecosystem services and trade-offs(InVEST)model were used to assess the trends in land use degree and carbon storage in the MRYR during *** potential impact index and the standard deviation ellipse(SDE)algorithm were applied to quantify and analyze the characteristics of the impact of land use changes on carbon ***,land use transitions that led to carbon storage variations and their spatial variations were *** results showed that:(1)the most significant periods of carbon storage changes and land use transitions were observed during 1990-1995 and 1995-2020,with the most changed areas locating in the east of Fenhe River and in northwestern Henan Province;(2)the positive impact of land use degree on carbon storage may be related to the environmental protection measures implemented along the Yellow River,while the negative impact may be associated with the expansion of construction land in plain areas;and(3)the conversion of other land use types to grassland was the primary factor affecting carbon storage changes during *** future land use planning,attention should be given to the direction of grassland conversion,and focus on reasonably limiting the development of construction *** enhance carbon storage,it will be crucial to increase the area

Drought-induced desiccation cracking in surface soil:Two-dimensional characterization by FEM-OFDR framework

Drought-induced desiccation cracking can trigger several weakening mechanisms in surface soils, potentially precipitating instability and failure of slopes and earthen structures. To investigate the potential application of distributed fibre optical sensing(DFOS) based on optical frequency domain reflectometry(OFDR) technology in characterizing the twodimensional(2D) desiccation cracking processes of surface soils, a comprehensive test device is utilized to conduct soil evaporation tests, continuously record water content changes, desiccation cracking evolution, and FO sensing strain status. A deep learning-based quantitative analysis method is employed to meticulously examine the relationship between 2D cracking geometric parameters and strain status. The comprehensive analysis not only reveals the mutual feedback response mechanism between the strain status and the soil evaporation-shrinkage-cracking processes, but also clarifies the early detection distance of OFDR technology for 2D desiccation cracking. Specifically, OFDR technology can detect the propagation of horizontal desiccation cracks up to 23 mm in advance with a strain measurement accuracy of 1 με. To address the spatial continuity issue in OFDR sensing strain data, an innovative high-resolution characterization framework is proposed by combining the finite element method(FEM) and OFDR technology, referred to as the FEM-OFDR framework. Comparative results indicate that the proposed FEM significantly surpasses both the kriging and radial basis function(RBF) methods in inferring missing OFDR sensing strain data. Notably, during the drying process, reaching a critical water content causes the local decoupling between the uncracked clods and the substrate, resulting in a decreasing trend in the sensing strain at the crack position. 2020年石家庄市89份乳品安全指标检测结果分析

目的了解2020年石家庄市市售乳品安全情况。方法2020年采集石家庄市12个县区4类乳品共89份样品,按照GB 4789.41—2016《食品安全国家标准食品微生物学检验肠杆菌科检验》、NY/T 1331—2007《乳与乳制品中嗜冷菌、需氧芽孢及嗜热需氧芽孢数的测定》分别对乳粉中肠杆菌科细菌以及生牛乳、巴氏杀菌乳和超高温灭菌乳中需氧芽孢和嗜热需氧芽孢进行检测;参考国家食品污染物和有害因素风险监测工作手册对生牛乳和巴氏杀菌乳中碱性磷酸酶进行检测;采用气相色谱-串联质谱法对乳粉中氯丙醇酯和缩水甘油酯进行检测。并根据现有国家限量标准对检测结果进行分析。结果89份乳品样品中有9份样品出现单指标或多指标阳性。乳粉、超高温杀菌乳、生牛乳和巴氏杀菌乳的合格率分别为100%、100%、16.67%和55.56%,其中生牛乳的检测指标阳性率由高到低分别为:碱性磷酸酶(100%)>需氧芽孢(83.33%)>嗜热需氧芽孢(16.67%);巴氏杀菌乳的检测指标阳性率由高到低分别为:需氧芽孢(44.44%)>嗜热需氧芽孢(11.11%)。结论石家庄市市售乳品中生牛乳和巴氏杀菌乳的污染问题较为突出,应当加大乳品原料的生产、加工、运输与贮存等环节监管力度,防控乳品污染风险,以确保乳品质量安全。

Mechanical Properties of Railway High-strength Manufactured Sand Concrete: Typical lithology, Stone Powder Content and Strength Grade

In order to achieve the large-scale application of manufactured sand in railway high-strength concrete structure,a series of high-strength manufactured sand concrete(HMC)are prepared by taking the manufactured sand lithology(tuff,limestone,basalt,granite),stone powder content(0,5%,10%,15%)and concrete strength grade(C60,C80,C100)as *** evolution of mechanical properties of HMC and the correlation between cubic compressive strength and other mechanical properties are *** to river sand,manufactured sand enhances the cubic compressive strength,axial compressive strength and elastic modulus of concrete,while its potential microcracks weaken the flexural strength and splitting tensile strength of *** powder content displays both positive and negative effects on mechanical properties of HMC,and the stone powder content is suggested to be less than 10%.The empirical formulas between cubic compressive strength and other mechanical properties are proposed.

Transient chemical-mediated epigenetic modulation confers unrestricted lineage potential on human primed pluripotent stem cells

Human primed pluripotent stem cells are capable of generating all the embryonic ***,their extraembryonic trophectoderm potentials are *** remains unclear how to expand their developmental potential to trophectoderm *** we show that transient treatment with a cocktail of small molecule epigenetic modulators imparts trophectoderm lineage potentials to human primed pluripotent stem cells while preserving their embryonic *** chemically treated cells can generate trophectoderm-like cells and downstream trophoblast stem cells,diverging into syncytiotrophoblast and extravillous trophoblast *** and CUT&Tag analyses reveal that these induced cells share transcriptional profiles with in vivo trophectoderm and cytotrophoblast,and exhibit reduced H3K27me3 modification at gene loci specific to trophoblast lineages compared with primed pluripotent *** exploration highlighted the critical roles of epigenetic modulators HDAC2,EZH1/2,and KDM5s in the activation of trophoblast lineage *** findings demonstrate that transient epigenetic resetting activates unrestricted lineage potential in human primed pluripotent stem cells,and offer new mechanistic insights into human trophoblast lineage specification and in vitro models for studying placental development and related disorders.

Exit options sustain altruistic punishment and decrease the second-order free-riders, but it is not a panacea

Why do humans punish free riders at their own expense? This behavior represents an evolutionary puzzle in human societies. This study explores the role of exit strategies in fostering altruistic punishment within evolutionary game theory. We extend the traditional prisoner's dilemma model by incorporating exiters, players who opt out for a small payoff while nullifying their opponent's payoff, and altruistic punishers who cooperate and punish non-cooperators. Our findings indicate that in well-mixed populations, exiters destabilize defection but do not promote altruistic punishment. In social networks, however,exiters enable altruistic punishment via cyclic dominance among altruistic punishers, defectors, and exiters. Notably, this cyclic dominance is sensitive to exit payoffs; adjustments can lead to cyclic dominance of non-punishing cooperators, defectors, and exiters, or to a bi-stable state between these two types of cyclic dominance. These results highlight the nuanced impact of exiters on altruistic punishment, emphasizing the need for careful incentivization of exit behavior. While exiters can support altruistic punishment in networked populations, their effectiveness is not a panacea and is highly sensitive to exit payoffs, indicating limits to the voluntary participation mechanism.

Critical new insights into the interactions of hexafluoropropylene oxide-dimer acid(GenX or HFPO-DA)with albumin at molecular and cellular levels

A key characteristic to be elucidated,to address the harmful health risks of environmental perfluorinated alkyl substances(PFAS),is their binding modes to serum albumin,the most abundant protein in *** oxide-dimer acid(GenX or HFPO-DA)is a new industrial replacement for the widespread linear long-chain ***,the detailed interaction of new-generation short-chain PFAS with albumin is still ***,the binding characteristics of bovine serum albumin(BSA)to GenX were explored at the molecular and cellular *** found that this branched short-chain GenX could bind to BSA with affinity lower than that of legacy linear long-chain perfluorooctanoic acid(PFOA).Site marker competitive study and molecular docking simulation revealed that GenX interacted with subdomain IIIA to form BSA-GenX *** with its weaker affinity to albumin protein,the cytotoxicity of branched short-chain GenX was less susceptible to BSA binding compared with that of the linear long-chain *** contrast to the significant effects of strong BSA-PFOA interaction,the weak affinity of BSA-GenX binding did not influence the structure of protein and the cytotoxicity of *** detailed characterization and direct comparisons of serum albumin interaction with new generation short-chain GenX will provide a better understanding for the toxicological properties of this new alternative.
