Relationship between Phenotypic Changes of Dendritic Cell Subsets and the Onset of Plateau Phase during Intermittent Interferon Therapy in Patients with CHB

Objective This study aimed to evaluate whether the onset of the plateau phase of slow hepatitis B surface antigen decline in patients with chronic hepatitis B treated with intermittent interferon therapy is related to the frequency of dendritic cell subsets and expression of the costimulatory molecules CD40,CD80,CD83,and CD86.Method This was a cross-sectional study in which patients were divided into a natural history group(namely NH group),a long-term oral nucleoside analogs treatment group(namely NA group),and a plateau-arriving group(namely P group).The percentage of plasmacytoid dendritic cell and myeloid dendritic cell subsets in peripheral blood lymphocytes and monocytes and the mean fluorescence intensity of their surface costimulatory molecules were detected using a flow cytometer.Results In total,143 patients were enrolled(NH group,n=49;NA group,n=47;P group,n=47).The results demonstrated that CD141/CD1c double negative myeloid dendritic cell(DNmDC)/lymphocytes and monocytes(%)in P group(0.041[0.024,0.069])was significantly lower than that in NH group(0.270[0.135,0.407])and NA group(0.273[0.150,0.443]),and CD86 mean fluorescence intensity of DNmDCs in P group(1832.0[1484.0,2793.0])was significantly lower than that in NH group(4316.0[2958.0,5169.0])and NA group(3299.0[2534.0,4371.0]),Adjusted P all<0.001.Conclusion Reduced DNmDCs and impaired maturation may be associated with the onset of the plateau phase during intermittent interferon therapy in patients with chronic hepatitis B.

Association of Cytokines with Clinical Indicators in Patients with Drug-Induced Liver Injury

Objective To explore characteristics of clinical parameters and cytokines in patients with drug-induced liver injury(DILI)caused by different drugs and their correlation with clinical indicators.Method The study was conducted on patients who were up to Review of Uncertainties in Confidence Assessment for Medical Tests(RUCAM)scoring criteria and clinically diagnosed with DILI.Based on Chinese herbal medicine,cardiovascular drugs,non-steroidal anti-inflammatory drugs(NSAIDs),antiinfective drugs,and other drugs,patients were divided into five groups.Cytokines were measured by Luminex technology.Baseline characteristics of clinical biochemical indicators and cytokines in DILI patients and their correlation were analyzed.Results 73 patients were enrolled.Age among five groups was statistically different(P=0.032).Alanine aminotransferase(ALT)(P=0.033)and aspartate aminotransferase(AST)(P=0.007)in NSAIDs group were higher than those in chinese herbal medicine group.Interleukin-6(IL-6)and tumor necrosis factor alpha(TNF-α)in patients with Chinese herbal medicine(IL-6:Pge may be more prone to DILI.Patients with NSAIDs have more severe liver damage in early stages of DILI,TNF-αand IL-6 may partake the inflammatory process of DILI.

Laboratory and field evaluation of a low-cost optical particle sizer

Low-cost sensors are widely used to collect high-spatial-resolution particulate matter data that traditional reference monitoring devices cannot.In addition to the mass concentration,the number concentration and size distribution are also fundamental in determining the origin and hazard level of particulate pollution.Therefore,low-cost optical sensors have been improved to establish optical particle sizers(OPSs).In this study,a low-cost OPS,the Nova SDS029,is introduced,and it is evaluated in comparison to two reference instruments—the GRIMM 11-D and the TSI 3330.We first tested the sizing accuracy using polystyrene latex spheres.Then,we assessed the mass and number size distribution accuracy in three application scenarios:indoor smoking,ambient air quality,and mobile monitoring.The evaluations suggest that the low-cost SDS029 rivals research-grade optical sizers in many aspects.For example,(1)the particle diameters obtained with the SDS029 are close to the reference instruments(usuallyge;(2)the number of particles and mass concentration are highly correlated(r≥0.99)with the values obtained with the reference instruments;and(3)the SDS029 slightly underestimates the number concentration,but the derived PM_(2.5)values are closer to monitoring station than the reference instruments.The successful application of the SDS029 in multiple scenarios suggests that a plausible particle size distribution can be obtained in an easy and cost-efficient way.We believe that low-cost OPSs will increasingly be used to map the sources and risk levels of particles at the city scale.

Asymmetrical Fault Current Calculation Method and Influencing Factors Analysis of Droop-Controlled Inverters

Since the fault dynamic of droop-controlled inverter is different from synchronous generators (SGs), protection devices may become invalid, and the fault overcurrent may damage power electronic devices and threaten the safety of the microgrid. Therefore, it is imperative to conduct a comprehensive fault analysis of the inverter to guide the design of protection schemes. However, due to the complexity of droop control strategy, existing literatures have simplified asymmetric fault analysis of droop-controlled inverters to varying degrees. Therefore, accurate fault analysis of a droop-controlled inverter is needed. In this paper, by analyzing the control system, an accurate fault model is established. Based on this, a calculation method for instantaneous asymmetrical fault current is proposed. In addition, the current components and current characteristics are analyzed. It was determined that fault currents are affected by control loops, fault types, fault distance and nonlinear limiters. In particular, the influences of limiters on the fault model, fault current calculation and fault current characteristics were analyzed. Through detailed analysis, it was found that dynamics of the control loop cannot be ignored, the fault type and fault distance determine fault current level, and part of the limiters will totally change the fault current trend. Finally, calculation and experimental results verify the correctness of the proposed method.

Exploring the correlation between temperature and crime:A case-crossover study of eight cities in America

Recent years have seen increasing academic interest in exploring the correlation between temperature and crime.However,it is uncertain whether similar long-term trends or seasonality(rather than causal effect)of temper-ature and crime is the major reason for the observed correlation between them.To explore whether there is still a correlation between temperature and crime when long-term trends and seasonal cycles are filtered out,we use the Kalman filter to decompose the time series of temperature and crimes,and then the fast Fourier transform is used to calculate the exact circle of their seasonality separately.Based on that,the box-plot method and linear regression are used to explore the correlation between temperature residuals and crime residuals.The results show that more than half of the crime types have similar seasonal cycles(approximately 1 year)to that of temperature.Moreover,the daily residual analyses show that temperature residuals have a positive correlation with assault and robbery residuals in all cities,whose average slopes are more than 0.1.The other four types of crimes vary greatly from case to case.The temperature residuals show a weak correlation with the residuals of some crime types.

Metal-free graphitic carbon nitride(g-C_(3)N_(4))has captured significant attention as a low-cost and efficient hydrogen production photocatalyst through.Effectively regulating the microstructure and accelerating the separation of photogenerated carriers remain crucial strategies for promoting the photocatalytic performance of this material.Herein,a novel sulfur–carbon co-doped g-C_(3)N_(4)(SCCN)hierarchical microtubules filled with abundant nanosheets inside by thermal polymerization is reported.Numerous nanosheets create abundant pores and cavities inside the SCCN microtubes,thereby increasing the specific surface area of g-C_(3)N_(4)and providing sufficient reactant attachment sites.Besides,the hierarchical structure of SCCN microtubules strengthens the reflection and scattering of light,and the utilization of visible light is favorably affected.More importantly,co-doping S and C has greatly improved the photocatalytic performance of graphitic carbon nitride,optimized the band gap structure and enhanced the photogenerated carrier splitting.Consequently,the SCCN exhibits a remarkable photocatalytic H_(2)evolution rate of 4868μmol/(g·h).This work demonstrates the potential of multi-nonmetal doped g-C_(3)N_(4)as the ideal photocatalyst for H2 evolution.

Coupling WO_(3-x) dots-encapsulated metal-organic frameworks and template-free branched polymerization for dual signal-amplified electrochemiluminescence biosensing

Developing accurate and sensitive DNA methyltransferase(MTase) analysis methods is essential for early clinical diagnosis and development of antimicrobial drug targets. In this work, by coupling WO_(3-x) dotsencapsulated metal-organic frameworks(MOFs) as co-reactants and terminal deoxynucleotidyl transferase(Td T)-mediated template-free branched polymerization, a dual signal-amplified electrochemiluminescent(ECL) biosensor was constructed to detect DNA adenine methylation(Dam) MTase. The employment of WO_(3-x) dots-encapsulated MOFs(i.e., NH_(2)-UIO66@WO_(3-x) ) was not only beneficial for biomolecule conjugation because of the abundant amino groups but also led to a 7-fold enhanced ECL response due to the increased loading of WO_(3-x). Moreover, Td T-mediated template-free branched polymerization promoted the capture of ECL emitters on the electrode surface, achieving 20-fold enhanced signal amplification. The presented ECL biosensor demonstrated a low detection limit of 2.4 × 10^(-4)U/m L, and displayed high reliability for the detection of Dam MTase in both spiked human serum and E. coli cell samples, and for the screening of potential inhibitors. This study opens a new avenue for designing a dual signal amplificationbased ECL bioassay for Dam MTase and screening inhibitors in the fields of clinical diagnosis and drug development.

Isolation and microbial transformation of tea sapogenin from seed pomace of Camellia oleifera with anti-inflammatory effects

In the current study,tea saponin,identified as the primary bioactive constituent in seed pomace of Camellia oleifera Abel.,was meticulously extracted and hydrolyzed to yield five known sapogenins:16-O-tiglogycamelliagnin B(a),camelliagnin A(b),16-O-angeloybarringtogenol C(c),theasapogenol E(d),theasapogenol F(e).Subsequent biotransformation of compound a facilitated the isolation of six novel metabolites(a1−a6).The anti-inflammatory potential of these compounds was assessed using pathogenassociated molecular patterns(PAMPs)and damage-associated molecular patterns molecules(DAMPs)-mediated cellular inflammation models.Notably,compounds b and a2 demonstrated significant inhibitory effects on both lipopolysaccharide(LPS)and high-mobility group box 1(HMGB1)-induced inflammation,surpassing the efficacy of the standard anti-inflammatory agent,carbenoxolone.Conversely,compounds d,a3,and a6 selectivity targeted endogenous HMGB1-induced inflammation,showcasing a pronounced specificity.These results underscore the therapeutic promise of C.oleifera seed pomace-derived compounds as potent agents for the management of inflammatory diseases triggered by infections and tissue damage.

Ultra-high extinction ratio optical pulse generation with a thin film lithium niobate modulator for distributed acoustic sensing

We experimentally demonstrate ultra-high extinction ratio(ER)optical pulse modulation with an electro-optical modulator(EOM)on thin film lithium niobate(TFLN)and its application for fiber optic distributed acoustic sensing(DAS).An interface carrier effect leading to a relaxation-tail response of TFLN EOM is discovered,which can be well addressed by a small compensation component following the main driving signal.An ultrahigh ER>50 dB is achieved by canceling out the tailed response during pulse modulation using the EOM based on a cascaded Mach–Zehnder interferometer(MZI)structure.The modulated optical_(√)pulses are then utilized as a probe light for a DAS system,showing a sensitivity up to-62.9 dB·rad∕Hz~2(7 pε/Hz)for 2-km single-mode sensing fiber.Spatial crosstalk suppression of 24.9 dB along the fiber is also obtained when the ER is improved from 20 dB to 50 dB,clearly revealing its importance to the sensing performance.

Effect of moxibustion on expression profile of miRNAs in Tripterygium glycoside-induced decreased ovarian reserve

OBJECTIVE: To explore the possible regulatory mechanism of microRNA(miRNA) in moxibustion treatment for decreased ovarian reserve(DOR). METHODS: The DOR model was constructed by intragastrical Tripterygium glycoside suspension administration, and moxibustion therapy was simultaneously given. The morphological ovarian changes were observed by hematoxylin and eosin staining. The miRNA expression profile was detected by RNA sequencing, and bioinformatics analysis was performed. Cytoscape software 3.6.1 was used to establish a regulatory network and differentially expressed miRNAs were verified by reverse transcription quantitative polymerase chain reaction(RT-qPCR). RESULTS: Decreased number of mature follicles, increased atresia follicles, and abnormal granulosa cell morphology were observed in the model group compared with the control group. The moxibustion group demonstrated increased mature follicles, decreased atretic follicles, and significantly decreased abnormal morphology of granulosa cells compared with the model group. Additionally, RNA sequencing results manifested significantly up-regulated miRNA expressions(miR-92b-3p, miR-26-5p＿R + 1＿1ss10TC, miR-206-3p, miR-9993b-3p＿1ss6GA, miR-7857-3p＿R-1, miR-219a-2-3p＿1ss10GC, miR-3968-p5＿1ss10AT, and PC-5p-6478＿1795) and down-regulated miR-664-2-5p＿R + 1 in the model group, compared with the control group, and the moxibustion group reversed abnormal disorder levels of these miRNAs. Moreover, these differentially expressed miRNAs were mainly involved in the phosphatidylinositol-3-kinase/protein kinase B signaling pathway and nuclear factor erythropoietin-2-related factor 2/heme oxygenase 1 signaling pathway. Finally, network and RT-qPCR verification revealed miR-9993b-3p＿1ss6GA as the most critical miRNA. CONCLUSION: This experiment proved the effectiveness of moxibustion in improving the ovarian reserve of rats by regulating miRNA expression, especially miR-9993b-3p＿1ss6GA.
