Nitrate reduction capacity of the oral microbiota is impaired in periodontitis:potential implications for systemic nitric oxide availability

The reduction of nitrate to nitrite by the oral microbiota has been proposed to be important for oral health and results in nitric oxide formation that can improve cardiometabolic conditions. Studies of bacterial composition in subgingival plaque suggest that nitrate-reducing bacteria are associated with periodontal health, but the impact of periodontitis on nitrate-reducing capacity(NRC)and, therefore, nitric oxide availability has not been evaluated. The current study aimed to evaluate how periodontitis affects the NRC of the oral microbiota. First, 16S rRNA sequencing data from five different countries were analyzed, revealing that nitratereducing bacteria were significantly lower in subgingival plaque of periodontitis patients compared with healthy individuals(P ll five datasets with n = 20–82 samples per dataset). Secondly, subgingival plaque, saliva, and plasma samples were obtained from 42 periodontitis patients before and after periodontal treatment. The oral NRC was determined in vitro by incubating saliva with 8 mmol/l nitrate(a concentration found in saliva after nitrate-rich vegetable intake) and compared with the NRC of 15healthy individuals. Salivary NRC was found to be diminished in periodontal patients before treatment(P lthy levels 90 days post-treatment. Additionally, the subgingival levels of nitrate-reducing bacteria increased after treatment and correlated negatively with periodontitis-associated bacteria(P l treatment on the baseline saliva and plasma nitrate and nitrite levels was found, indicating that differences in the NRC may only be revealed after nitrate intake. Our results suggest that an impaired NRC in periodontitis could limit dietary nitrate-derived nitric oxide levels, and the effect on systemic health should be explored in future studies.

Phytochemical analysis and antibacterial activities of Eleutherine bulbosa(Mill.) Urb. extract against Vibrio parahaemolyticus

Objective: To analyze compounds in Eleutherine bulbosa(E. bulbosa)(Mill.) Urb. extract and to determine its antibacterial capability against Vibrio parahaemolyticus(V. parahaemolyticus).Methods: E. bulbosa bulb extract was preprared using 96% ethanol by the maceration method. Phytochemical investigation of E. bulbosa extract was analyzed using GC-MS, spectrophotometry and titrimetry methods. The zone of inhibition was identified by the diffusion agar method. The minimum inhibitory concentration and minimum bactericidal concentration were determined using the plate count method. The inhibitory rate against V. parahaemolyticus was determined by the microdilution method. Cellular leakage was evaluated by spectrophotometry and cellular damage was observed by scanning electron microscopy. Results: GC-MS analysis showed the high compound of the E. bulbosa extract was securixanthone E(7-hydroxy-1,2-dimethoxyxanthone). The compound groups also included fatty acid esters, isoquinolines, naphthalenes, and phenolics. The total phenolic content was(2.50 ± 0.00) mg/g, flavonoid(6.61 ± 0.00) mg/g, and tannin(0.03 ± 0.00)%. The greatest zone of inhibition and inhibitory rate were(11.83 ± 0.06) mm and(91.32 ± 2.76)%, respectively, at 10 mg/m l. The minimum inhibitory concentration was 0.156 mg/m l, while the minimum bactericidal concentration was 10 mg/m l. The E. bulbosa extract caused leakage and cellular damage to V. parahaemolyticus. Conclusions: The E. bulbosa extract possesses inhibitory activities against V. parahaemolyticus and causes cellular leakage and damage.

Kinases: Understanding Their Role in HIV Infection

Antiviral drugs currently on the market primarily target proteins encoded by specific viruses. The drawback of these drugs is that they lack antiviral mechanisms that account for resistance or viral mutation. Thus, there is a pressing need for researchers to explore and investigate new therapeutic agents with other antiviral strategies. Viruses such as the human immunodeficiency virus (HIV) alter canonical signaling pathways to create a favorable biochemical environment for infectivity. We used Qiagen Ingenuity Pathway Analysis (IPA) software to review the function of several cellular kinases and the resulting perturbed signaling pathways during HIV infection such as NF-κB signaling. These host cellular kinases such as ADK, PKR, MAP3K11 are involved during HIV infection at various stages of the life cycle. Additionally IPA analysis indicated that these modified host cellular kinases are known to have interactions with each other especially AKT1, a serine/threonine kinase involved in multiple pathways. We present a list of cellular host kinases and other proteins that interact with these kinases. This approach to understanding the relationship between HIV infection and kinase activity may introduce new drug targets to arrest HIV infectivity.

Assessment of Water Quality and Microbial load of Dhaleshwari River Tangail, Bangladesh

The present study was undertaken to investigate some water quality parameters (pH, BOD, DO and water temperature), anion concentration (PO43- , SO42- and NO3- ) and the availability of some selected microorganisms consisted of total coliform, fecal coliform (Escherichia coli) and Vibrio cholerae in the Dhaleshwari river during the period from April to June 2015. The water samples were collected from three different stations of the river namely Belta (St-1), Kendua (St-2) and Charabari (St-3) under the Porabari union at Tangail Sadar Upazilla. Water temperature varied from 29.5 oC in June to 31.7 oC in May with a highest mean 31.17 ± 0.47 in May. The pH of water was slightly alkaline in nature and ranged between 7.25 in June and 8.45 in April. Mean concentrations of BOD 0.87 ± 0.19, 1.12 ± 0.6 and 0.97 ± 0.67 were recorded in April, May and June respectively. Highest PO42- value 4.4 mg/l was found in May and lowest value of PO42- 1.74 mg/l was found in June. Mean sulfate values at different months of Dhaleshwaririver were 6.61 ± 1.78 mg/l, 7.61 ± 2.16 mg/l and 3.68 ± 0.23 mg/l respectively. Nitrate concentration in water samples varied from 0.51 mg/l in June to 3.66 mg/l in April. Total coliforms bacteria were found high in all water samples and values ranged from 11.05 × 108 - 19 × 1023 cfu/ml. The highest load of total coliforms bacteria was recorded at St-2 in May and the lowest was at St-1 in April. The highest concentration of (Escherichia coli counted in water sample was 6 × 1013 cfu/ml at St-2 during June, while lowest concentration was 5 × 105 cfu/ml at St-1 during May. The highest concentration of Vibrio cholerae was 9.66 × 1014 cfu/ml at St-1 during June, while lowest concentration was 4.2 × 108 cfu/ml at St-3 during April. The high number of total coliform, Escherichia coli and Vibrio cholerae indicated significant level of microbial pollution in Dhaleshwari river. Biochemical Traits and Physico-Chemical Attributes of Aromatic-Fine Rice in Relation to Yield Potential

Aromatic rice has great potential to attract rice consumer for its taste and deliciousness, and high price to boost up the economic condition of the rice grower in the developing countries. A field experiment was conducted to evaluate the morpho-physiological and biochemical attributes in relation to yield potentials of fine aromatic rice varieties in Bangladesh. Two aromatic fine rice cultivars, Ukunmadhu and Kataribough (local land races), one hybridized variety BRRIdhan-32 and one mutant variety Binasail were used in the experiment. The results revealed that rice varieties BRRIdhan-32 and Binasail showed the greater number of branches of panicle, grain size, grain yield and harvest index as compared to aromatic varieties. On the other hand, aromatic rice showed taller plant stature, more tiller, plant biomass and larger panicle but lower number of primary and secondary branches, grain size, yield and harvest index. The biochemical attributes namely chlorophyll and soluble protein content were higher at vegetative stage as compared to panicle initiation and flowering stages. The aromatic rice Kataribhough and Ukonmondhu contained higher amount of total free amino acid as compared to that of Binasail and BRRIdhan32. Other biochemical parameters namely nitrate reductase activity (NRA), total sugar and free amino acid contents were higher at flowering stage compared to those contained at tillering and panicle initiation stages. Physico-chemical properties of grain viz. length, breadth, l/B ratio, nitrogen (%) and protein (%) were significantly higher in fine rice as compared to aromatic rice. From the results it may be concluded that the high yielding varieties were more efficient in transfer of photosynthate to the grain i.e. economic yield and that is why BRRIdhan32 recorded the highest yield. Poor yield in Ukonmodhu and Kataribhough was due to low harvest index indicating minimum translocation of assimilates to the grains.

In this study we transfected CHO cells with human β2- AR （wild type （WT） or truncated at amino acids 344 or 349） and measured glucose uptake and cAMP accumulation, In CHO cells expressing WT β2- AR, glucose uptake was increased by isoprenaline, insulin and 8 - bromo - cAMP, In cells expressing truncated receptors （349 or 344） insulin and 8 - bromo - cAMP increased glucose uptake to the same degree as in the wild type β2 - AR - CHO cells, but isoprenaline - stimulated glucose uptake was significantly reduced, suggesting that C - terminal PKA and GRK phosphorylation sites are important for β2 - AR stimulated glucose uptake, cAMP responses to forskolin or isoprenaline were not significantly changed between the wild type and truncated receptors.

Hypertrophic cardiomyopathy (HCM) is a myocardial disorder, with complications including heart failure, thromboemboli and sudden death. Human and feline HCM (fHCM) are clinically comparable, thus fHCM may serve as a spontaneous animal model. fHCM in Maine Coon (MC) cats is associated with the p.A31P mutation in the cMyBP-C protein. The mutation is located in the cMyBP-C C0-domain which is known to interact with actin. The presence and levels of the wild type and mutated protein in heart tissue from mutant and wild type MC cats were examined by SDS-PAGE and mass spectrometry (MS). Quantitative yeast-2-hybrid (Y2H) protein-protein interaction analysis was used to assess the effect of the mutation on C0C1/actin interaction. The NMR-based structure of the C0 domain was used to calculate the energetic consequence of replacing alanine with a proline residue. In the homozygous MC cat, the mutated cMyBP-C protein was present, and cMyBPC-C levels were not reduced compared to that of the wild type cat. However, the interaction of actin with mutant cMyBP-C C0C1 was reduced compared to that of wild type. This may be because the substitution of the alanine with proline in position 31 was energetically highly unfavorable and resulted in only one hydrogen bond within the anti-parallel beta-strand compared to two hydrogen-bonds for alanine, possibly destabilizing the structure of the actin-interacting domain. The p.A31P mutation is present in cardiac tissue and the most likely pathogenic mechanism is interference with contractility by reducing binding of the C0C1 domain of cMyBP-C to actin.

目的:确定CO2超临界萃取细叶杜香的最佳工艺条件。方法:采用均匀设计,以萃取率为考察指标,对影响CO2超临界萃取因素中的萃取压力、萃取温度和萃取时间3个主要因素进行考察。结果:CO2超临界萃取细叶杜香的最佳工艺条件:萃取压力33PM,萃取温度42℃,萃取时间2.5h,在此条件下,细叶杜香挥发油的萃取率为7.90%。结论:该工艺具有萃取率高、速度快、操作简便等优点,可用于CO2超临界萃取细叶杜香。

Wild dogma： An examination of recent ＂evidence＂ for dingo regulation of invasive mesopredator release in Australia

There is growing interest in the role that apex predators play in shaping terrestrial ecosystems and maintaining trophic cascades. In line with the mesopredator release hypothesis, Australian dingoes (Canis lupus dingoand hybrids) are assumed by many to regulate the abundance of invasive mesopredators, such as red foxesVulpes vulpesand feral catsFelis catus, thereby providing indirect benefits to various threatened vertebrates. Several recent papers have claimed to provide evidence for the biodiversity benefits of dingoes in this way. Nevertheless, in this paper we highlight several critical weaknesses in the methodological approaches used in many of these reports, including lack of consideration for seasonal and habitat differences in activity, the complication of simple track-based indices by incorporating difficult-to-meet assumptions, and a reduction in sensitivity for assessing populations by using binary measures rather than potentially continuous measures. Of the 20 studies reviewed, 15 of them (75%) contained serious methodological flaws, which may partly explain the inconclusive nature of the literature investigating interactions between invasive Australian predators. We therefore assert that most of the “growing body of evidence” for meso-predator release is merely an inconclusive growing body of literature only. We encourage those interested in studying the ecological roles of dingoes relative to invasive mesopredators and native prey species to account for the factors we identify, and caution the value of studies that have not done so.
