Emerging structures and dynamic mechanisms ofγ-secretase for Alzheimer’s disease

γ-secretase,called“the proteasome of the membrane,”is a membrane-embedded protease complex that cleaves 150+peptide substrates with central roles in biology and medicine,including amyloid precursor protein and the Notch family of cell-surface *** inγ-secretase and amyloid precursor protein lead to early-onset familial Alzheimer’s disease.γ-secretase has thus served as a critical drug target for treating familial Alzheimer’s disease and the more common late-onset Alzheimer’s disease as ***,critical gaps remain in understanding the mechanisms of processive proteolysis of substrates,the effects of familial Alzheimer’s disease mutations,and allosteric modulation of substrate cleavage byγ-*** this review,we focus on recent studies of structural dynamic mechanisms ofγ-*** mechanisms,including the“Fit-stay-Trim,”“sliding-Unwinding,”and“Tilting-Unwinding,”have been proposed for substrate proteolysis of amyloid precursor protein byγ-secretase based on all-atom molecular dynamics *** an incorrect registry of the Notch1 substrate was identified in the cryo-electron microscopy structure of Notch1-boundγ-secretase,molecular dynamics simulations on a resolved model of Notch1-boundγ-secretase that was reconstructed using the amyloid precursor protein-boundγ-secretase as a template successfully capturedγ-secretase activation for proper cleavages of both wildtype and mutant Notch,being consistent with biochemical experimental *** approach could be potentially applied to decipher the processing mechanisms of various substrates byγ-*** addition,controversy over the effects of familial Alzheimer’s disease mutations,particularly the issue of whether they stabilize or destabilizeγ-secretase-substrate complexes,is ***,an outlook is provided for future studies ofγ-secretase,including pathways of substrate binding and product release,effects of modulators on familial Alzheimer’s disease mutations

Unravelling CoVID-19 waves in Rio de Janeiro city:Qualitative insights from nonlinear dynamic analysis

since the CoVID-19 pandemic was first reported in 2019,it has rapidly spread around the *** countries implemented several measures to try to control the virus *** healthcare system and consequently the general quality of life population in the cities have all been significantly impacted by the Coronavirus *** different waves of contagious were responsible for the increase in the number of cases that,unfortunately,many times lead to *** this paper,we aim to characterize the dynamics of the six waves of cases and deaths caused by CoVID-19 in Rio de Janeiro city using techniques such as the Poincaréplot,approximate entropy,second-order difference plot,and central tendency *** results reveal that by examining the structure and patterns of the time series,using a set of non-linear techniques we can gain a better understanding of the role of multiple waves of CoVID-19,also,we can identify underlying dynamics of disease spreading and extract meaningful information about the dynamical behavior of epidemiological time *** findings can help to closely approximate the dynamics of virus spread and obtain a correlation between the different stages of the disease,allowing us to identify and categorize the stages due to different virus variants that are reflected in the time series.

Nonlinear study on the Mechanical Performance of Built-Up Cold-Formed steel Concrete-Filled Columns under Compression

Given their numerous functional and architectural benefits,such as improved bearing capacity and increased resistance to elastic instability modes,cold-formed steel(CFs)built-up sections have become increasingly developed and used in recent years,particularly in the construction *** paper presents an analytical and numerical study of assembled CFs two single channel-shaped columns with different slenderness and configurations(backto-back,face-to-face,and box).These columns were joined by double-row rivets for the back-to-back and box configurations,whereas they were welded together for the face-to-face *** built-up columns were filled with ordinary concrete of good *** element models were applied,using ABAQUs software,to assess mechanical performance and study the influence of assembly techniques on the behavior of cold-formed columns under axial *** approaches based on Eurocode 3 and Eurocode 4 recommendations for un-filled and concrete-filled columns respectively were followed for the numerical analysis,and concrete confinement effects were also considered per American Concrete Institute(ACI)standards for face-to-face and box *** obtained results indicated a good correlation between the numerical results and the proposed analytical methodology which did not exceed 8%.The failure modes showed that the columns failed due to instabilities such as local and global buckling.

Molecular mechanism of Xuebijing in treating pyogenic liver abscess complicated with sepsis

BACKGRoUND:Xuebijing(XBJ)can alleviate the inflammatory response,improve organ function,and shorten the intensive care unit(ICU)stay in patients with pyogenic liver abscess(PLA)complicated with sepsis,but the molecular mechanisms have not been *** study aimed to explore the molecular mechanism of XBJ in treating PLA complicated with sepsis using a network pharmacology ***:The active ingredients and targets of XBJ were retrieved from the ETCM *** targets related to PLA and sepsis were retrieved from the GeneCards,PharmGKB,DisGenet,online Mendelian Inheritance in Man(oMIM),Therapeutic Targets Database(TTD),and DrugBank *** targets of PLA complicated with sepsis were mapped to the targets of XBJ to identify potential treatment ***-protein interaction networks were analyzed using the sTRING *** treatment targets were imported into the Metascape platform for Gene ontology(Go)functional enrichment and Kyoto encyclopedia of Genes and Genomes(KEGG)pathway enrichment *** docking was performed to validate the interactions between active ingredients and core ***:XBJ was found to have 54 potential treatment targets for PLA complicated with ***-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor(TNF)were identifi ed as core *** enrichment analysis revealed important pathways,including the interleukin-17(IL-17)signaling pathway,the TNF signaling pathway,the nuclear factor-kappa B(NF-κB)signaling pathway,and the Toll-like receptor(TLR)signaling *** docking experiments indicated stable binding between XBJ active ingredients and core ***:XBJ may exert therapeutic eff ects on PLA complicated with sepsis by modulating signaling pathways,such as the IL-17,TNF,NF-κB,and TLR pathways,and targeting IL-1β,IL-6,and TNF.

We revisit the problem of adsorption of a single^(4)He layer on graphene,focusing on the commensurate(C_(1/3))crystalline phase,specifically on whether it may possess a nonzero superfluid response,and on the existence of superfluid phases,either(metastable)liquid or vacancy-doped *** make use of canonical quantum Monte Carlo simulations at zero and finite temperature,based on a realistic microscopic model of the *** results confirm the absence of any superfluid response in the commensurate crystal,and that no thermodynamically stable uniform phase exists at lower *** evidence of a possibly long-lived,metastable superfluid phase at C_(1/3)coverage is ***,the results of ground-state projection methods and finite-temperature simulations are entirely consistent.

FEA-based Geometrical Modification of switched Reluctance Motor for Radial Force Reduction

switched reluctance motors(sRMs)are becoming increasingly popular in the automotive sector owing to their robust ***,sRMs are preferred particularly for EV applications owing to their fault tolerance,magnet-free structure,and high power/torque *** main concerns of sRM compared to other machines include torque ripple and *** primary cause of vibration is the radial force created by the sRM.A geometry-based modification of the sRM to reduce the radial force without significantly changing the average torque produced is *** primary goal is to design a 4-phase,8/6 sRM with a lower radial *** possible geometrical alterations are proposed:one with square windows and the other with circular holes on the rotor *** windows are sized and positioned to avoid flux *** criteria are developed for the optimal window size and *** element analysis(FEA)modelling of the sRM is used to validate its *** FEA results are compared with the performance parameters obtained using the analytical *** the multiphysics design tool ANsYs to obtain the natural frequencies and associated deformations through modal *** to the conventional geometry,the radial force is significantly reduced by providing windows.
