The role of interleukin 25 （IL-25） in a number of human diseas.s.still has not been extensively studied, here we attempt to evaluate the role of recombinant IL-25 （rIL-25） in the development of dextran sulfate sodium （Dss）- induced experimental colitis. Acute colitis was induced in female C57BL/6 mice by oral administration of 2.5% Dss in drinking water ad libitum. At the same time as the start of Dss exposure, mice were injected intraperitoneally with 0.4 ＋tg of rIL-25 or PBs. Then disease activity index （DAI）, histological changes and survival rate were observed. The levels of IL-17, IL-23, and TGF-β1 in colon tissues were determined by ELIsA, and the production of IL-17 by CD4＋/CD8＋ T cells was detected by intracellular flow cytometry. In contrast to the Dss treated mice, Dss ＋ rIL-25 treated mice displayed a lower DAI, limited histological changes and prolonged survival. The levels of IL-23 and TGF-β1 were significantly elevated in the Dss ＋ rIL-25 treated mice compared to the Dss treated mice. There was no significant difference in the production of IL-17 in colon tissues and CD4＋/CD8＋ T cells between the Dss ＋ rIL-25 treated mice and Dss treated mice. Our findings.s.ggest the role of IL-25 in inhibiting development and progression of acute colitis in Dss-induced mouse colitis model. Cellular ＆ Molecular Immunology. 2008;5（6）：425-431.

Objective: To investigate the s.s.eptibility to some conventional and non-conventional insecticides in laboratory and field larval populations of the West Nile vector Culex pipiens L.(Cx. pipiens), the dominant species in Jeddah Province, saudi ***: The tested conventional insecticides were Actikil and Pesgard, while the nonconventional ones were Bacilod, Dudim and Baycidal. Probit analys.s.and photomicroscopical observations were carried out to shed light on acute toxicity in laboratory and field Cx. pipiens ***: Cx. pipiens were more s.s.eptible to Pesgard(LC50: 0.045 and 0.032 mg/L)than Actikil(0.052 and 0.038 mg/L) and Bacilod(0.129 and 0.104 mg/L), for the field and laboratory strains, respectively. Results.s.owed that treatments with the chitin synthes.s.inhibitor Dudim and Baycidal evoked morphological effects.s.milar to those induced by other insect growth regulators. According to IC50 values obtained(concentration which to inhibit the emergence of 50% of mosquito adults), the compound Dudim(0.000 3 and 0.000 1 mg/L) was more effective against Cx. pipiens L. mosquitoes than Baycidal(0.000 4 and 0.000 3 mg/L) for both the field and laboratory strains, ***: Our results provide baseline data to enhance control programs and orient public health decisions on the selection of pesticides against mosquito vectors in saudi Arabia.

制备脂质体包裹的Ag85A口服DNA疫苗,并观察小鼠口服后所诱生的抗体产生情况。用脂质体包裹重组质粒pcDNA3.1/myc-HisA-Ag85A制备口服DNA疫苗,并用脂质体包裹空质粒pcDNA3.1/myc-HisA作为对照。将C57BL/6小鼠随机分为3组,即生理盐水组、空质粒组和重组质粒DNA疫苗组。分别将生理盐水、空质粒和重组质粒DNA疫苗以灌胃方式投给各组小鼠,共免疫3次,每次间隔14d,末次免疫后14d处死小鼠,ELIsA法检测血清中Ag85A特异性抗体水平,放射免疫法测定肠组织中分泌型IgA(sIgA)含量。重组质粒组血清中Ag85A特异性抗体滴度为1160,空质粒组和生理盐水组血清中均未捡出Ag85A特异性抗体。重组质粒组肠组织中sIgA含量(0.3761±0.0456)μg/mL较空质粒组(0.2374±0.0414)μg/mL和生理盐水(0.1993±0.0899)μg/mL组显著增高(P0.05)。口服脂质体包裹Ag85ADNA疫苗可诱导外周特异性抗体的产生和肠道黏膜局部sIgA的水平的升高。

日本沼虾（Macrobrachium nipponense）,广泛分布在东亚地区包括中国、日本、韩国、越南、缅甸等地,是中国重要的淡水养殖品种。近年来,其年养殖产量超27万吨,年产值约200亿元人民币,具有很高的经济效益。因此开展日本沼虾遗传多样性和种群结构研究对于其种质资源保护和开发利用意义重大。遗传多样性是指生物的遗传变异程度,可以表现在分子、细胞、个体等不同层次上。自然界中,多样性增强了生物适应环境的能力,且绝大多数通过有性生殖繁殖的生物,其种群内的个体之间不存在完全一致的基因型,这些具有不同遗传结构的个体组成了种群,对种群的遗传多样性研究可以为物种改良和种质保护提供重要的理论依据。水产动物遗传多样性研究多采用DNA分子标记技术,常用的标记有RAPD、AFLP、ssR等。线粒体DNA（mtDNA）D-Loop序列由于不编码蛋白质而不受自然选择的影响,因而进化速率最快,积累了较多的突变,在种群遗传学分析中有普遍应用。本文采用线粒体DNA控制区（D-loop）,系统地分析了中国从南到北不同纬度8个日本沼虾群体的多样性和遗传结构特征。采集的群体包括:松花江群体（sHJ）、黄河群体（HHX）、长江群体（YZ）、珠江群体（ZJ）、微山湖群体（WsH）、太湖群体（TH）、博思腾湖群体（BH）和万泉河群体（WQH）。对上述8个群体168个样本的日本沼虾线粒体D-loop基因进行分析,分别提取个体总DNA,通过线粒体DNA D-Loop基因片段进行扩增,并将产物进行测序。共有144个样品测序成功。通过比对和人工校对后共有1130bp的序列用于进一步的分析。研究结果表明,在1130bp的D-loop环中,有239个突变位点,产生73个单倍型。结果显示A+T（79.04%）的含量高于C+G（20.95%）,表明线粒体基因组D-loop区域的A+T含量高于C+G。群体单倍型多样性排序为:WQH（0.9810）＞YZ（0.9571）＞WsH（0.9429）＞ZJ（0.8901）＞sHJ0.8791)＞HH0.8693)＞TH（0.8667）＞BH（0.3684）;核苷酸多样性排序为:WQH（0.0607）＞YZ（0.0299）＞sHJ（0.0236）＞WsH（0.0221）＞TH（0.0132）＞ZJ（0.0131）＞HH（0.0118）＞BH（0.0011）,表明仅有博思腾湖群体遗传多样性很低,其余7个群体遗传多样性均很丰富,特别是万泉河群体多样性很高。另外,8个群体的群体内遗传距离的范围为0到0.06,两两群体间的遗传距离为0.014到0.102。两两群体间遗传分化指数（FsT）值显示,WsH群体与TH群体、HH群体、ZJ群体分化不显著（P＞0.05）,其余两两群体间差异达到极显著水平（P＜0.01）,说明8个日本沼虾群体遗传分化十分明显（P＜0.01）。基因流（Nm）也证明,WsH群体与TH群体（11.057）、HH群体（7.226）、ZJ群体（6.255）基因交流比较丰富。本文对日本沼虾的系统发育和种群结构进行了分析。在研究中,通过构建ML系统发育树,结果显示73个单倍型分为不同的两大支系,每支存在不同分支。仅有博思腾湖群体（BH）的所有单倍型聚在一起,其余群体的单倍型均分散在不同支系,表明BH群体内遗传差异很低,而其他群体内差异较高。Network结果与系统发育树结果一致。错配分布分析显示所有8个群体呈现多峰分布。通过Fu’s Fs检验和Tajima’D检验发现,8个群体均为非显著性负值,推测日本沼虾群体种群可能没有经历种群的扩张。本研究系统地从南向北调查了中国日本沼虾种群的遗传多样性和种群遗传结构特征,研究结果可为日本沼虾种质资源保护和开发利提供有力的依据。

Development of a New Bio-Based Insulating Fluid from <i>Jatropha curcas</i>Oil for Power Transformers

The present work aims to develop a new vegetable insulating fluid for power transformers based on Jatropha curcas oil. Besides its technical benefits, Jatropha curcas oil has a socio-economic role by promoting income to rural families, contributing to the countryside development and avoiding rural exodus. Thus, the entire transformer oil production (extraction, processing, characterization and accelerated aging) was covered and a new process was developed. For oil extraction, the most suitable process was the solvent extraction (5 mL of hexane per gram of crushed non-peeled seeds during 30 minutes) with an oil yield of 32%. In raw oil processing stage, the degumming, with 0.4 g of phosphoric acid per 100 g of oil, at 70°C, was used to remove phosphatides. Then, free fatty acids were 96% neutralized with a sodium hydroxide solution (0.5% w/w) at room temperature. For the oil clarification, the combination of 5% w/woil of activated carbon and 1% w/woil of MgO resulted in a bright, odorless and clear oil with an acid number of 0.04 mgKOH·g﹣1. The oil drying in a vacuum rotary evaporator, at 70°C, for 2 hours reduced the water content to 177 ppm. The processed oil was characterized following AsTM D6871 methods. This oil presented higher dielectric breakdown voltage (55 kV) than commercial transformer fluids (BIOTEMP?, EnvirotempFR3?, and Bivolt?), which increas.s.transformer safety, capacity and lifetime. In addition, the processed oil has a lower viscosity than BIOTEMP? fluid, which can enhance the heat dissipation efficiency in the transformer. Moreover, the processed oil flash and fire points of 310°C and >340°C, respectively, confirm the great security of vegetable insulating fluids. The analyzed properties of the processed oil fulfill all the AsTM D6871, ABNT NBR 15422 and IEC 62770 specifications. Therefore, Jatropha curcas oil is a potential substitute formineral insulating fluids.

The present work aimed at the study of citric acid solvent extraction in order to establish the composition of the organic phase and to obtain thermodynamic and kinetic data for the chosen s.s.em. Discontinuous extraction experiments in a single stage were performed from a synthetic solution of citric acid, with the typical concentration (10% w/v) observed in industrial fermented mus.s. Exploratory experiments were carried out using different organic phas.s.in order to select the most suitable solvent phase to further continuous extraction tes.s.in a mechanically agitated column. The selected organic phase composition was: Alamine? 336, ExxalTM 13 tridecyl alcohol, and the aliphatic diluent EscaidTM 110. Next, the effects of the contact time and of the concentrations of extractant and modifier on the citric acid extraction were studied. Among the investigated conditions, the best one was 10 minutes of contact time, 30% w/v of Alamine? 336, and 10% w/v of ExxalTM 13 tridecyl alcohol. For this condition, the equilibrium isotherm (28°C ± 2°C) was determined, and the equilibrium constant was calculated (36.8 (mol·L-1)-1.5). It was considered that trioctylamine and citric acid complexation reaction occurs mainly with non-dissociated citric acid form, because the aqueous feed solutions’ pH is lower than the citric acid pKa1. It was found that 1.5 molecules of the extractant, on average, are required to react with one citric acid molecule, which can indicate that reactions with different extractant/citric acid ratios occur simultaneously. Next, the rate constants for the direct and inverse reactions, 2.10 (mol·L-1)-1.5·s-1 and 5.69 × 10-2 s-1, respectively, were calculated. Coefficients of determination (R2) values higher than 0.93 were found in these calculations, suggesting that the results obtained using a computer modeling would be very close to those results obtained experimentally.
