Perfluorinated iodine alkanes promote the differentiation of mouse embryonic stem cells by regulating estrogen receptor signaling

Investigating the development toxicity of perfuorinated iodine alkanes(PFIs)is critical,given their estrogenic effects through binding with estrogen receptors(ERs).In the present study,two PFIs,including dodecafuoro-1,6-diiodohexane(PFHx DI)and tridecafuorohexyl iodide(PFHx I),with binding preference to ERαand ERβ,respectively,were selected to evaluate their effects on proliferation and differentiation of the mouse embryonic stem cells(m EsCs).The results revealed that,similar to E_(2),50μmol/L PFHx DI accelerated the cell proliferation of the m *** PFI stimulation at the exposure concentrations of 2–50μmol/L promoted the differentiation of the m EsCs as characterized by the upregulation of differentiation-related biomarkers(i.e.,Otx2 and Dnmt3β)and downregulation of pluripotency genes(i.e.,Oct4,Nanog,sox2,Prdm14 and Rex1).Comparatively,PFHx DI exhibited higher induction effect on the differentiation of the m EsCs than did PFHx *** tests on ER signaling indicated that both PFI compounds induced exposure concentration-dependent expressions of ER signaling-related biomarkers(i.e.,ERα,ERβand Caveolin-1)in the m EsCs,and the downstream ER responsive genes(i.e.,c-fos,c-myc and c-jun)well responded to PFHx I *** role of ER in PFI-induced effects on the m EsCs was further validated by the antagonistic experiments using an ER inhibitor(ICI).The findings demonstrated that PFIs triggered ER signaling,and perturbed the differentiation program of the m EsCs,causing the potential health risk during early stage of development.

Tensile properties and deformation behavior of an extra-low interstitial fine-grained powder metallurgy near alpha titanium alloy by recycling coarse pre-alloyed powder

An extra-low interstitial near alpha alloy Ti-3Al-2Zr-2Mo(wt%) was fabricated by hydrogenation and thermomechanical consolidation(TMC) of the coarse and spherical pre-alloyed powder with particle sizes of 60 to 270 μm. The coarse powder is a byproduct of pre-alloyed powder produced for selective laser and electron beam additive manufacturing. The TMC process involves powder compaction, fast sintering,in-situ dehydrogenation and an immediate hot extrusion to form a fully dense and fine-grained martensitic microstructure. Further dehydrogenation in vaccum at 700 °C converted the martensitic microstructure into an interwoven α/β microstructure which exhibited an improved yield strength, apparent necking and premature cracking at grain boundary α(α_(GB)) ribbons. A further annealing of 880 ℃/1 h/AC led to the formation of a fine-grained α/β_(t)composite structure, which achieved an enhance ultimate tensile strength of 835 MPa and excellent tensile ductility of 16.0%. Analysis of the deformation behavior of the alloy in different states revealed that the α/β_(t)composite structures brought about an enhanced strain hardening capability by heterogeneous deformation effect of hard β_(t)and soft α-laths, which inhibited the formation of microcracks and consequently improved the coordinated deformation.

Intranasal administration of tetrabromobisphenol A bis(2-hydroxyethyl ether) induces neurobehavioral changes in neonatal sprague Dawley rats

Tetrabromobisphenol A（TBBPA） and its derivatives are now being highly concerned due to their emerging environmental occurrence and deleterious effects on non-target *** the potential neurotoxicity of TBBPA derivatives which has been demonstrated in vitro, what could happen in vivo is worthy of being studied. Tetrabromobisphenol A bis（2-hydroxyethyl ether）（TBBPA-BHEE）, a representative TBBPA derivative, was selected for a21-day exposure experiment on neonatal sprague Dawley（sD） rats through intranasal administration. The neurobehavioral, histopathological changes, and differentially expressed genes based on RNA microarray were investigated to evaluate the neurological effects of this chemical. The results indicated that TBBPA-BHEE exposure significantly compromised the motor co-ordination performance and the locomotor activities（p 〈 0.05）. The neurobehavioral phenotype could be attributed to the obvious histopathological changes in both cerebrum and cerebellum, such as neural cell swelling, microglial activation and proliferation. A total of 911 genes were up-regulated, whereas 433 genes were down-regulated. Gene set enrichment analysis showed multiple signaling pathways, including ubiquitin-mediated proteolysis and wingless-int（Wnt） signaling pathway etc. were involved due to TBBPA-BHEE exposure. The gene ontology enrichment analysis showed the basic cellular function and the neurological processes like synaptic transmission were influenced. The toxicological effects of TBBPA-BHEE observed in this study suggested the potential neuronal threaten from unintended exposure,which would be of great value in the biosafety evaluation of TBBPA derivatives.

Flow Pattern Identification of Fluidized Beds Using ECT

Electrical capacitance tomography(ECT) was applied in measuring solids distribution in square circulating fluidized beds. The fluidization conditions varied from bubbling fluidized bed to circulating fluidized bed. In the whole range of fluidization conditions, ECT was able to instantaneously provide the solids concentration and voids distributions in the fluidized beds. According to the acquired data from ECT and reconstructed image, different fluidization regimes can also be identified.

sediment loads response to climate change:A preliminary study of eight large Chinese rivers

Climate change characterized by increasing temperature is able to affect precipitation regime and thus surface ***,the manner in which river sediment loads respond to climate change is not well understood,and related assessment regarding the effect of climate change on sediment loads is *** present a quantitative estimate of changes in sediment loads(from 1.5 Gt yr-1pre-1990 to 0.6 Gt yr-1from 1991-2007) in response to climate change in eight large Chinese *** the past decades,precipitation change coupled with rising temperatures has played a significant role in influencing the sediment delivery dynamics,although human activities, such as reservoir construction,water diversion,sand mining and land cover change,are still the predominant forces. Lower precipitation coupled with rising temperatures has significantly reduced sediment loads delivered into the sea in semi-arid climates(4-61%).In contrast,increasingly warmer and wetter climates in subtropical zones has yielded more sediment(0.4-11%),although the increase was offset by human *** results indicate that,compared with mechanical retention by reservoirs,water reduction caused by climate change or human withdrawals has contributed more sediment reduction for the rivers with abundant sediment supply but limited transport capacity(e.g.,the Huanghe).Furthermore,our results indicate that every 1%change in precipitation has resulted in a 1.3%change in water discharge and a 2%change in sediment *** addition,every 1%change in water discharge caused by precipitation has led to a 1.6%change in sediment loads,but the same percentage of water discharge change caused largely by humans would only result in a 0.9%change in sediment *** figures can be used as a guideline for evaluating the responses of sediment loads to climate change in similar climate zones because future global warming will cause dramatic changes in water and sediment in river basins worldwide at rates previou
