AIM:To examine the regulatory role of microRNA-204(miR-204)on silent information regulator 1(sIRT1)and vascular endothelial growth factor(VEGF)under highglucose-induced metabolic memory in human retinal pigment epithelial(hRPE)***:Cells were cultured with either normal(5 mmol/L)or high D-glucose(25 mmol/L)concentrations for 8d to establish control and high-glucose groups,*** induce metabolic memory,cells were cultured with 25 mmol/L D-glucose for 4d followed by culture with 5 mmol/L D-glucose for *** addition,exposed in 25 mmol/L D-glucose for 4d and then transfected with 100 nmol/L miR-204 control,miR-204 inhibitor or miR-204 mimic in 5 mmol/L D-glucose for *** reverse transcription-polymerase chain reaction(RT-qPCR)was used to detect miR-204 mRNA ***1 and VEGF protein levels were assessed by immunohistochemical and Western *** cytometry was used to investigate apoptosis ***:It was found that high glucose promoted miR-204 and VEGF expression,and inhibited sIRT1 activity,even after the return to normal glucose culture *** of miR-204 promoted apoptosis inhibiting sIRT1 and increasing VEGF ***,downregulation of miR-204 produced the opposite ***:The study identifies that miR-204 is the upstream target of sIRT1and VEGF,and that miR-204 can protect hRPE cells from the damage caused by metabolic memory through increasing sIRT1 and inhibiting VEGF expression.

BACkGROUND The nutritional status is closely related to the prognosis of liver transplant re-cipients,but few studies have reported the role of preoperative objective nutri-tional indices in predicting liver transplant *** To compare the predictive value of various preoperative objective nutritional indicators for determining 30-d mortality and complications following liver transplantation(LT).METHODs A retrospective analysis was conducted on 162 recipients who underwent LT at our institution from December 2019 to June *** This study identified several independent risk factors associated with 30-d mor-tality,including blood loss,the prognostic nutritional index(PNI),the nutritional risk index(NRI),and the control nutritional *** 30-d mortality rate was 8.6%.Blood loss,the NRI,and the PNI were found to be independent risk factors for the occurrence of severe postoperative *** NRI achieved the highest prediction values for 30-d mortality[area under the curve(AUC)=0.861,Psevere complications(AUC=0.643,P=0.011).Compared to those in the high NRI group,the low patients in the NRI group had lower preoperative body mass index and prealbumin and albumin levels,as well as higher alanine aminotransferase and total bilirubin levels,Model for End-stage Liver Disease scores and prothrombin time(Psignificantly greater incidences of intraabdominal bleeding,primary graft nonfunction,and *** The NRI has good predictive value for 30-d mortality and severe complications following *** NRI could be an effective tool for transplant surgeons to evaluate perioperative nutritional risk and develop relevant nutritional therapy.

Water contamination by tetracycline(TC)has emerged as an environmental concern owing to its widespread use and antibiotic *** of peracetic acid(PAA)in the water and wastewater treatment has recently been proposed and demonstrated to be effective for TC abatement,yet the underlying reaction kinetics between the PAA and TC are not yet *** explore the reaction kinetics,the effect of solution pH on TC abatement by PAA is studied and the species-specific rate constants are *** ability to donate and accept electrons for different species of TC and PAA is evaluated via density functional theory(DFT)*** pH-dependent apparent second-order rate constants of TC abatement by PAA exhibits the parabolic shape with the maximum at p H 8.5(9.75 L mol^(-1)s^(-1)).This phenomenon is closely related to the speciation of TC and PAA,in which the reaction between PAAH and TTC^(2-)possesses the highest species-specific rate constants according to the kinetic *** DFT calculations suggest that the HOMO of TTCH^(+),TTC,TTC^(-),TTC^(2-)and the LUMO of PAAH and PAA-are–6.40,–6.26,–5.10,–4.94 e V and–0.24,0.60 e V,*** to the DFT calculations,deprotonation of TC and PAA leads to an increase of the HOMO value of TC and the LUMO value of ***,the HOMOTC–LUMOPAAvalues is in good agreement with the trend of species-specific rate constants,which can be used to evaluate the reactivity between PAA and TC with different *** study provides the kinetic data and theoretical basis for the reaction of PAA and TC,which is critical for the application of PAA in the treatment of water and wastewater.

Artificial intelligence(AI)technology has become integral in the realm of medicine and healthcare,particularly in human activity recognition(HAR)applications such as fitness and rehabilitation *** study introduces a robust coupling analysis framework that integrates four AI-enabled models,combining both machine learning(ML)and deep learning(DL)approaches to evaluate their effectiveness in *** analytical dataset comprises 561 features sourced from the UCI-HAR database,forming the foundation for training the ***,the MHEALTH database is employed to replicate the modeling process for comparative purposes,while inclusion of the WIsDM database,renowned for its challenging features,supports the framework’s resilience and *** ML-based models employ the methodologies including adaptive neuro-fuzzy inference system(ANFIs),support vector machine(sVM),and random forest(RF),for data *** contrast,a DL-based model utilizes one-dimensional convolution neural network(1dCNN)to automate feature ***,the recursive feature elimination(RFE)algorithm,which drives an ML-based estimator to eliminate low-participation features,helps identify the optimal features for enhancing model *** best accuracies of the ANFIs,sVM,RF,and 1dCNN models with meticulous featuring process achieve around 90%,96%,91%,and 93%,*** analysis using the MHEALTH dataset showcases the 1dCNN model’s remarkable perfect accuracy(100%),while the RF,sVM,and ANFIs models equipped with selected features achieve accuracies of 99.8%,99.7%,and 96.5%,***,when applied to the WIsDM dataset,the DL-based and ML-based models attain accuracies of 91.4%and 87.3%,respectively,aligning with prior research *** conclusion,the proposed framework yields HAR models with commendable performance metrics,exhibiting its suitability for integration into the healthcare services system through AI-driven applications.
