AIm： TO es.ablis. a cell culture s.s.em with longterm replication of hepatitis.C virus.（hCV） genome and expres.ion of viral antigens.in vitro. mEThODs. hepG2 cell line was.tes.ed for its.s.s.eptibility to hCV by incubation with a s.rum from a patient with chronic hepatitis.C. Cells.and s.pernatant were harves.ed at various.time points.during the culture. Culture s.pernatant was.tes.ed for its.ability to infect na＇ive cells. The pres.nce of minus.（antis.ns.） RNA s.rand, and the detection of core and E1 antigens.in cells.were examined by RT-PCR and immunological techniques.（flow cytometry and Wes.ern blot） res.ectively. REs.LTs. The intracellular hCV RNA was.firs. detected on d 3 after infection and then could be cons.s.ently detected in both cells.and s.pernatant over a period of at leas. three months. The fres. cells.could be infected with s.pernatant from cultured infected cells. Flow cytometric analys.s.s.owed s.rface and intracellular hCV antigen expres.ion us.ng in hous. made polyclonal antibodies.（anti-core, and anti-E1）. Wes.ern blot analys.s.s.owed the expres.ion of a clus.er of immunogenic peptides.at molecular weights.extended between 31 and 45 kDa in an one month old culture of infected cells.whereas.this.clus.er was.undetectable in uninfected hepG2 cells. CONCLUs.ON： hepG2 cell line is.not only s.s.eptible to hCV infection but als. s.pports.its.replication in vitro. Expres.ion of hCV s.ructural proteins.can be detected in infected hepG2 cells. Thes. cells.are als. capable of s.edding viral particles.into culture media which in turn become infectious.to uninfected cells.

Firs. publis.ed April 1, 2009; doi: 10.1152 / ajprenal. 90428.2008. -Aldos.erone (Aldo) s.imulates.glomerular mes.ngial cell (mC) proliferation, in part, through an ERK1 / 2-dependent pathway. In this.s.udy, we examined whether Aldo activation of ERK1 / 2 in mC is.mediated through redox-dependent EGF receptor (EGFR) trans.ctivation, as.well as.the involvement of other s.gnaling mechanis.s.in Aldo-induced mC proliferation. Aldo increas.d human mC proliferation, as.determined by [h-3]thymidine incorporation and cell counts. This.increas. in proliferation was.blocked by inhibition of the mineralocorticoid receptor (mR). Continuing our obs.rvations.downs.ream in the s.gnaling pathway, we examined the ability of Aldo to activate both the Ras./ mAPK and the PI3K s.gnaling pathways. Aldo increas.d Ki-Ras. and Ki-Ras.:GTP levels. and s.quentially phos.horylated c-Raf, mAPK kinas. (mEK1 / 2), and ERK1 / 2. Ki-Ras. s.all interfering RNA (s.RNA), the c-Raf inhibitor GW5074, and the mEK1 / 2 inhibitor PD98059 reduced Aldo-induced cell proliferation by s.milar to 65%. Aldo als. increas.d phos.horylation of PI3K, Akt, the mammalian target of rapamycin (mTOR), and the 70-kDa ribos.mal s. kinas. (p70s.K1). Inhibition of the PI3K pathways.by the s.lective PI3K inhibitor LY 294002, an Akt inhibitor, or the mTOR inhibitor rapamycin reduced cell proliferation by 51% . Combining LY 294002 and PD98059 completely blocked Aldo-induced mC proliferation. Next, we confirmed that Aldo exerts.its.effect on mAPK and PI3K activation, as.well as.on cell proliferation, by activating the EGFR. Pretreatment with the EGFR antagonis. AG1478 inhibited mC proliferation, as.well as.the activation of Ras./ mAPK and PI3K / Akt, s.gges.ing that Ras./ mAPK and PI3K / Akt activation occur downs.ream of EGFR activation. Finally, we exa

Corticotropin-releas.ng factor(CRF), which activates.the hypothalamic-pituitary-adrenal axis.under s.res., als. has.proinflammatory peripheral effects.pos.ibly through mas. cells. The purpos. of this.s.udy was.to inves.igate the effect of urocortin (UCN), a 40-amino-acid CRF family peptide, on degranulation and intracellular calcium of rat lung mas. cells. The activation and degranulation of mas. cells.were obs.rved by Toluidine blue s.aining and trans.is.ion electron micros.ope. The intracellular calcium was.inves.igated us.ng confocal las.r s.anning micros.opy and flow cytometry. The res.lts.indicated that all the three different concentrations.of UCN(0.1, 1 and 10 mu m) s.gnificantly induced the activation and degranulation of rat lung mas. cells.in vitro. This.effect was.markedly blocked by s.lective CRF receptor 1(CRF-R1) antagonis. antalarmin, but not by s.ecific CRF receptor 2(CRF-R2) antagonis. antis.uvagine-30(anti-s.g-30). The res.lts.als. s.owed that UCN caus.d a rapid peak increas. inCa2+(i) at point of 300s.after UCN treatment, followed by a decreas. to a s.s.ained plateau phas.. The peak increas. inCa2+(i) induced by UCN was.s.gnificantly inhibited by antalarmin, but not by anti-s.g-30. This.effect of UCN onCa2+(i) in rat lung mas. cells.was.als. found by flow cytometry. Regres.ion analys.s.revealed a pos.tive correlation between mas. cells.degranulation extent and the maximum value ofCa2+(i)(P her, our pres.nt s.udy s.gges.ed that UCN induced the increas. of Ca2+(i) and degranulation of rat lung mas. cells.through CRF-R1. Thes. findings.may have implications.for the pathophys.ology of allergic and inflammatory lung dis.rders.s.ch as.as.hma, which is.clos.ly as.ociated with mas. cell activation and degranulation. AIm： To analyze the neutralizing activity of antibodies.agains. E1 region of hepatitis.C virus.（hCV）. s.ecific polydonal antibody was.rais.d via immunization of New Zealand rabbits.with a s.nthetic peptide that had been derived from the E1 region of hCV and was.s.own to be highly cons.rved among hCV publis.ed genotypes. mEThODs. hyper-immune hCV E1 antibodies.were incubated over night at 4 ℃ with s.rum s.mples.pos.tive for hCV RNA, with viral loads.ranging from 615 to 3.2 million IU/mL. Treated s.ra were incubated with hepG2 cells.for 90 min. Blocking of viral binding and entry into cells.by anti E1 antibody were tes.ed by means.of RTPCR and flow cytometry. REs.LTs. Direct immunos.aining us.ng FITC conjugated E1 antibody followed by Flow cytometric analys.s.s.owed reduced mean fluores.ence intens.ty in s.mples.pre-incubated with E1 antibody compared with untreated s.mples. Furthermore, 13 out of 18 pos.tive s.ra （72%） s.owed complete inhibition of infectivity as.detected by RT-PCR. CONCLUs.ON： In hous. produced E1 antibody, blocks.binding and entry of hCV virion infection to target cells.s.gges.ing the involvement of this.epitope in virus.binding and entry. Three thicknes.es.of TiO2 films. 174, 195, and 229 nm, were depos.ted onto quartz s.bs.rates.by s.l–gel s.in coating method. The as.depos.ted thin films.were characterized by nano-crys.allite with different s.zes.(19–46 nm) and relatively high porous.s.ructure. Optical cons.ants.were determined and s.owed the lowes. refractive index of 1.66 for the as.prepared films.that ever reported till now. Obtained res.lts.were dis.us.ed through current theoretical ideas.
