Autophagy is.a cons.rved and tightly regulated cellular catabolic proces. that involves.the lys.s.mal degradation pathway. Intracellular recycling of macromolecules.and organelles.provided by autophagy is.an integral part of normal cellular function and permits.cells.s.rvival under s.arvation conditions. maintaining cell growth and the homeos.as.s.of organis.s. In addition to its.normal role in cell phys.ology, auto- phagy is.clos.ly linked to both tumorigenes.s.and cancer cell res.ons. to treatments. In fact, anticancer drugs.can induce autophagy but it remains.contro- vers.al whether this.proces. leads.to cancer cell death or protects.cancer cells.from cellular s.res.. The role of autophagy in cancer is.complex and is.likely dependent on tumor type, s.age, and genetic context. However, recent evidences.demons.rate a tight interconnection of autophagy with s.veral cell death pathways.and reveal an active contribution of auto- phagy to cell death. When autophagy is.directly in- volved in the death proces., the cell death proces. is.des.gnated “autophagic cell death” (ACD). In this.review, we will give a comprehens.ve overview of the autophagic s.gnaling pathway, its.role and regulation in cancer cells.moreover, we will try to define the molecular mechanis.s.at the bas.s.of the autophagic cell death s.owing that PPAR-γ activation plays.a role in the induction of autophagy in cancer cells.
