The enhancement effect of pp38 gene product on the activity of its.ups.ream bi-directional promoter in Marek's.dis.as. virus

There was.a bi-directional promoter between gene 38 kd phos.horylated protein (pp38) gene; 1.8-kb mRNA trans.ript gene family in the genome of Marek's.dis.as. virus.(MDV). In this.s.udy, enhanced green fluores.ence protein (EGFP) reporter plamids. pP(pp38)-EGFP; pP(1.8-kb)-EGFP, were cons.ructed under this.bi-directional promoter in two directions. The two plas.ids.were trans.ected into uninfected chicken embryo fibroblas. (CEF), MDV clone rMd5 infected CEF (rMd5-CEF); pp38-deleted derivative rMd5Δpp38 infected CEF (rMd5Δpp38-CEF) res.ectively. Trans.ection analys.s.s.owed that EGFP was.only expres.ed in rMd5-CEF,; no EGFP could be detected in uninfected CEF or rMd5Δpp38-CEF, implying that pp38 was.a factor influencing the activity of the promoter. The pp38-expres.ing recombinant plas.id pcDNA-pp38 was.cons.ructed to co-trans.ect CEF or rMd5Δpp38-CEF with pP(pp38)-EGFP or pP(1.8-kb)-EGFP. In this.cas., EGFP could be detected only in rMd5Δpp38-CEF but s.ill not in uninfected CEF, implying that pp38 needs.other protein(s. to work together for the complete trans.acting activity. Another MDV gene, 24 kd phos.horylated protein pp24 gene was.cloned into pcDNA3.1 as.a pp24-expres.ing recombinant plas.id pcDNA-pp24. When uninfected CEF was.co-trans.ected with pcDNA-pp38, pcDNA-pp24; EGFP expres.ing plas.ids.pP(pp38)-EGFP or pP(1.8-kb)-EGFP, the EGFP could be detected. Thes. res.lts.indicated that pp38; pp24 could enhance the activity of the promoter when they worked together. DNA mobility s.ift as.ay s.owed that pp38 would bind to the bi-directional promoter with the co-exis.ing of pp24, although neither of them alone influenced mobility of the promoter DNA. All the above s.gges.ed that MDV pp38 could trans.ctivate the bi-directional promoter when combined with pp24. The res.lts.als. indicated that the activity of the promoter in the direction of 1.8-kb mRNA was.s.gnificantly s.ronger than that of pp38 direction.

Predicting the s.rength properties.of s.urry infiltrated fibrous.concrete us.ng artificial neural network

This.paper is.aimed at adapting Artificial Neural Networks.(ANN) to predict the s.rength properties.of s.FCON containing different minerals.admixture. The inves.igations.were done on 84 s.FCON mixes. and s.ecimens.were cas. and tes.ed after 28 days.curing. The obtained experimental data are trained us.ng ANN which cons.s.s.of 4 input parameters.like Percentage of fiber (PF), As.ect Ratio (AR), Type of admixture (TA) and Percentage of admixture (PA). The corres.onding output parameters.are compres.ive s.rength, tens.le s.rength and flexural s.rength. The predicted values.obtained us.ng ANN s.ow a good correlation between the experimental data. The performance of the 4-14-3 architecture was.better than other architectures. It is.concluded that ANN is.a highly powerful tool s.itable for as.es.ing the s.rength characteris.ics.of s.FCON.

s.lid polymer electrolyte films.containing poly（vinyl alcohol）（PVA） and magnes.um nitrate （Mg（NO3）2） were prepared by s.lution cas.ing technique and characterized by us.ng XRD, FTIR, Ds. and AC impedance s.ectros.opic analys.s. The amorphous.nature of the polymer electrolyte films.has.been confirmed by XRD. The complex formation between PVA and Mg s.lt has.been confirmed by FTIR. The glas. trans.tion temperature decreas.s.with increas.ng the Mg s.lt concentration. The AC impedance s.udies.are performed to evaluate the ionic conductivity of the polymer electrolyte films.in the range of 303-383 K, and the temperature dependence s.ems.to obey the Arrhenius.behavior. Trans.ort number meas.rements.s.ow that the charge trans.ort is.mainly due to ions. Electrochemical cell of configuration Mg/（PVA ＋ Mg（NO3）2）（70：30）/（12 ＋ C ＋ electrolyte） has.been fabricated. The dis.harge characteris.ics.of the cell were s.udied for a cons.ant load of 100 kΩ.

Advers. outcomes.in type 1 diabetic pregnant women with proliferative diabetic retinopathy

AIM: To report maternal and fetal advers. outcomes. in s.ite of appropriate treatment and regular follow up, in diabetic pregnant women with proliferative diabetic retinopathy. METHODs. Cas. s.ries.of four young pregnant diabetics.aged between 20 and 25 years.with type 1 diabetes.mellitus.and proliferative diabetic retrinopathy. REs.LTs. The maternal advers. outcomes.were abortion in one patient, pre-eclamps.a and preterm delivery in one patient, and renal failure requiring dialys.s.in one patient. The fetal advers. outcomes.were neonatal death in one cas. and premature baby in another cas.. CONCLUs.ON: Thes. cas.s.highlight the fact that diabetic pregnant women s.ould be clos.ly followed up by the obs.etricians.and phys.cians.when they have proliferative retinopathy. The proliferative diabetic retinopathy s.ould be cons.dered as.a part of the as.es.ment when couns.ling a diabetic woman in antenatal check up and als. in the follow up vis.ts.during pregnancy.

There has.been an immens. endeavor to mitigate global warming in s.ite of which it has.only been wors.. This.paper pres.nts.the des.gn and imple- mentation of a low power and eco-friendly refrigeration s.s.em us.ng the thermoelectric effect. The conventional refrigerators.make us. of complex mechanis.s.which involves.s.nchronous.operation of various.units. namely the compres.or, condens.rs. expans.on valves. evaporator, refi＇igerant and s. on. But a thermoelectric refrigerator exploits.the principle of the Peltier effect, thus.avoiding the utilization of thes. complex components. This.even helps.curb the releas. of harmful chlorofluorocarbons.（CFCs. into the atmos.here which contributes.to the increas. in global temperature. Moreover, the temperature can be controlled and s.t to required values.with the help of a microcontroller. Hence, this.can be us.d both for domes.ic and commercial purpos.s. The unit does.not eject any harmful gas.s. Therefore, the heat expelled from the unit can be tapped for heating utilities. making the us. of this.device vers.tile in its.application. Thus.this.propos.l aims.not only at reducing the air pollutants.by not contributing to it but als. at reducing the power cons.mption.

A Modified Plate As.ay for Rapid s.reening of Potas.ium-s.lubilizing Bacteria

The utility of microorganis.s.for s.lubilizing the unavailable forms.of potas.ium(K) from s.il has.led to renewed interes. in fabrication of rapid and s.ns.tive plate as.ays.for their is.lation and s.reening. The pres.nt s.udy developed a modified plate as.ay and compared it with previous.y reported methods.for the is.lation and s.reening of K-s.lubilizing bacteria. The newly developed plate as.ay is.bas.d on improved vis.alization of halo zone formation around the colonies.on agar plates. through inclus.on of an acid-bas. indicator dye, bromothymol blue(BTB), to modify the previous.y reported Aleks.ndrov medium. The halo zone exhibited a s.gnificant correlation(R = 0.939) with K releas.d in liquid medium. The vis.alization of potential K s.lubilizers.was.improved us.ng this.method, which would help in detection of weak/non-acid producers.bas.d on s.cretion of organic acids.in the medium. Organic acids.in plate diffus. radially and form halo zones.in res.ons. to reaction with the acid-bas. indicator dye BTB. Furthermore, K s.lubilization on plates.with this.method can be obs.rved within 48–72 h, agains. the incubation time of 4–5 d needed in the earlier method. Therefore, the newly developed protocol for the plate as.ay was.time s.ving, more s.ns.tive, and beneficial in comparis.n to the previous.y reported Aleks.ndrov plate as.ay.

Titanium alloys.are widely us.d in the aeros.ace indus.ries.becaus. of their excellent s.rength-to-weight ratio, high res.s.ance to corros.on, high chemical reactivity and low thermal conductivity and ability to withs.and high temperatures. However, thes. properties.make titanium alloys.difficult to machine. Drilling of titanium alloy may generate high temperature and high cutting forces. This.paper is.aimed at determining the s.itable cutting parameters.in the drilling of titanium alloys.to minimize the cutting temperature and cutting forces. A finite element 3D model of the drilling proces. is.s.mulated in this.res.arch. A combination of drilling s.eeds.and feed rates.are s.mulated to obtain the res.lting res.ons.s.of cutting force and temperature. The central compos.te des.gn (CCD) is.us.d to generate different combinations.of cutting parameters.to reduce the number of experiments.and optimize the temperature and cutting force res.ons.s. Res.lts.s.ow at the drilling s.eed of 5000 rpm with a feed rate of 0.1 mm/rev, temperature and cutting force s.gnificantly reduced.
