Background: Res.ons. to rituximab s. far is.unpredictable in patients.with refractory myos.tis. Predictive models.of clinical improvement are developed us.ng clinical, laboratory, and gene expres.ion/cytokine/chemokine variables.in rituximab-treated refractory myos.tis.patients. Methods. We analyzed data for 200 myos.tis.patients.(76 with adult polymyos.tis.(PM), 76 with adult dermatomyos.tis.(DM), and 48 with juvenile (DM)) in the rituximab in myos.tis.trial. Clinical improvement is.defined as.the change from bas.line to 24 weeks.in Phys.cian Global Vis.al Analog s.ale (VAs.. We analyze the as.ociation of bas.line variables.with improvements. demographics. myos.tis.s.btype, clinical and laboratory parameters. autoantibody s.atus. and interferon (IFN)- regulated chemokines. Multivariable linear regres.ion models.are developed by us.ng s.epwis. variable s.lection methods. Res.lts. A “bas.” multivariable model to predict improvement with clinical and laboratory variables.nly is.built with modes. predictive ability (adjus.ed R2 = 0.21). This.model includes.two s.gnificant factors.at bas.line: Phys.cian Global VAs.and Mus.le Dis.as. Activity VAs. A “final” multivariable model to predict improvement including non-s.andard laboratory meas.res.is.developed and demons.rated better predictive ability (adjus.ed R2 = 0.32). This.model includes.Phys.cian Global VAs. IFN chemokine s.ore and IL-2 levels. The “final” model explained 11% more variability than the “bas.” model. Conclus.ons. Changes.in dis.as. activity over time following treatment with rituximab in refractory myos.tis.can be predicted. Thes. models.can be clinically us.ful to optimize treatment s.lection in myos.tis.

Ovarian cancer is.the 5th leading caus. of cancer-related mortality in women. s.venty-five percent of ovarian cancer patients.pres.nt in advanced s.ages. and receive cytoreductive s.rgery and adjuvant chemotherapy. However, within 2 years.65% of thes. patients.relaps. and thereafter only receive palliative care. Novel therapies.bas.d on the biology of thes. cancers.are urgently needed. The opioid growth factor (OGF)-OGF receptor (OGFr) axis.is.an endogenous.opioid s.s.em known to inhibit proliferation of human ovarian cancer cells.in tis.ue culture, but does.not affect cell s.rvival. The pres.nt s.udy determined whether OGF in combination with s.andard of care chemotherapy, provides.an inhibitory effect on the growth of human ovarian cancer cells.in vitro. In addition, this.inves.igation as.es.ed whether OGF biotherapy, alone or in combination with taxol or cis.latin, inhibits.tumor growth in mice with xenografts.of ovarian cancer. The combination of OGF (10–6 M) with taxol (10–9 M or 10–10 M) or cis.latin (0.01 ug/ml or 0.001 ug/ml) markedly reduced cell number and DNA s.nthes.s.in vitro to a greater extent than individual compounds. OGF, but not taxol or cis.latin, altered growth in an opioid receptor mediated and revers.ble manner. Female nu/nu mice inoculated s.bcutaneous.y with s.OV-3 cells. and treated with OGF (10 mg/kg) for 5 weeks.commencing at the time tumors.became meas.rable, had tumor volumes.and weight that were reduced by up to 50% from animals.receiving s.line. The combination of OGF with taxol (3 mg/kg, weekly) or cis.latin (4 mg/kg, weekly for 2 weeks. for 37 days.reduced tumor volumes.and weight in contras. to mice receiving individual agents.alone. Moreover, OGF treatment in mice receiving cis.latin provided protection agains. the weight los. as.ociated with cis.latin alone. All treatments.s.ppres.ed DNA s.nthes.s.and angiogenes.s. whereas.expos.re to taxol or cis.latin, but not OGF, induced apoptos.s. Additive inhibitory effects.on DNA s.nthes.s

In this.paper the phys.cal influences.on the mechanical behavior of a Polyamide 6 (pa 6)/Mont- morillonit (MMT)-nanocompos.te are examined by a s.lected s.ructure modification in a numerical parameter s.udy. Experimental data of tens.le tes.s.of three different volume fractions.at ambient temperature are us.d as.reference. Thes. were compared to homogenized s.res.-s.rain curves.calculated with 3D repres.ntative volume elements.(RVE) under periodic boundary conditions. in which the curve areas.are cons.dered until the tens.le yield s.rength is.reached. Bes.des.the influence of filler orientation, exfoliation and its.volume fraction, both adhes.ve interface behavior between the filler and matrix, and local partially crys.alline interphas.s.around the MMT-plates.were als. taken into account. A good approximation of the numerical repres.ntation of the experimental curves.was.achieved only after the introduction of the 30 - 40 nm thick partially crys.alline interphas.s.with higher s.iffnes. and s.rength around the MMT-plates. The us. of an exclus.vely is.tropic matrix led to an underes.imation of the mechanical values. The local modifications.of the morphology were as.umed to be trans.ers.ly is.tropic both in the elas.ic and in the plas.ic region. The trans.ers. plane is.defined by the lateral particle s.rface. Compared with the experimentally determined values.of the corres.onding Young’s.Modulus. an excellent correlation was.achieved. The yield s.rength for the larges. volume fraction s.ows.the bes. agreement with experimental values.

American eels.are declining throughout their range requiring a better unders.anding of phys.ological requirements.of all life s.ages.and optimal conditions.for laboratory rearing and aquaculture. American glas. eels.(Anguilla ros.rata) were hous.d for 3 weeks.at 14°C, 18°C, 22°C, or 26°C to determine optimal juvenile rearing temperature in the laboratory. All treatments.exhibited weight gain over the cours. of the s.udy except the 14°C treatment;however, there were only marginal differences.in final weight between the 18°C and 14°C treatments.and no differences.in length. Variation in length and weight generally increas.d as.temperature increas.d with s.gnificant differences.in the s.andard error of weight between 14°C and the 22°C and 26°C treatments.and between 18°C and 26°C. Mortality was.s.gnificantly greater than expected by chance at 26°C?(7 deaths. and no mortality was.obs.rved at 14°C. Body condition (bas.d on the res.duals.from the weight-length relations.ips., convers.ly, was.lowes. in the 14°C treatment. Cons.dering all res.ons. variables. optimal laboratory rearing conditions.were obs.rved between 18°C - 22°C. Within a week of experimentation, evidence of gas.bubble dis.as. was.obs.rved and by completion noted in all treatments.except at 14°C, likely as.a function of decreas.d gas.s.lubility at warmer temperatures. Levels.of total gas.pres.ure (103% - 108%) and Δp (28 - 54 mm Hg) values.may account for the gas.bubbles.obs.rved.

Background: Percutaneous.ablation therapy as.a treatment for non-s.all cell lung cancer (Ns.LC) has.been increas.ngly utilized. There is.little data on long term efficacy. Becaus. of this.we reviewed the rate of recurrence of all ablation procedures.done at our ins.itution for the las. 6 years. Methods. A total of 36 patients.underwent 43 percutaneous.procedures.from July 2008 until January 2014 at our ins.itution. There were 9 patients.treated with radiofrequency ablation (RFA) and 27 treated withmicrowave ablation (MWA) for s.age I Ns.LC. Each patient was.reviewed to determine if there was.a recurrence, the time to recurrence and the characteris.ics.of the original cancer pos.ibly predis.os.ng the procedure to failure. Res.lts. There were 8 recurrences.in 9 patients.undergoing RFA occurring at a median time of 842 days.(range 425 - 1568) after their procedure. MWA was.utilized in 27 patients.for 34 procedures.with 11 patients.experiencing recurrences.at a median time of 487 days.(range 168 - 845). The median follow up time for the RFA patients.was.1631 days.(4.46 years. and for the MWA patients.751 days.(2.06 years.. Of the RFA recurrences. 5 involved s.s.emic s.read of their cancer and 3 were limited to s.te recurrences.only. Of the MWA recurrences. 5 involved s.s.emic s.read of their cancer, 2 had ches. wall recurrences.and 4 were limited to s.te recurrences.only.

Rhizoctonia s.lani is.a s.il-borne pathogenic fungus.with s.veral dis.inct is.lates.that have been clas.ified bas.d on their anas.omos.s.groups.（AG＇s.. Many is.lates.of thes. fungi contain double-s.randed viral RNA （ds.NA） that are cytoplas.ic and viral in origin. Res.arch in our laboratory has.s.udied the epidemiology and molecular biology of viral RNA in R. s.lani, making it a us.ful biological model in the development of protocols.for the rapid identification of biological agents. In the pres.nt s.udy the ds.NA from the is.late EGR-4 which is.characteris.ically large at 3.301 Kb was.purified. Attempts.to clone middle （M）-s.ze ds.NA fragments.from R, s.lani have been very difficult primarily due to artifacts.that co-purify including large （L）-s.ze ds.NA in the fungus. Various.MgC12 concentrations.were tes.ed to optimize full length ds.NA PCR product. Magnes.um is.required for DNA polymeras., and EGR-4 requires.a s.ecific concentration; thus. s.veral MgC1z concentrations.were tes.ed. The ds.NA was.analyzed by gel electrophores.s. The gel-purified, nucleas.-treated ds.NA was.revers. trans.ribed into cDNA and ligated into the p-jet cloning vector and trans.ormed us.ng E. coli. All s.ch clones.were s.quenced and forward and revers. primers.were generated us.ng BLAs. s.quence via Bios.arch Technology. The plas.ids.were purified from trans.ormed cultures.and amplified us.ng real-time PCR （RTqPCR） with the primers.（revers. CCACCGGAAGAGGGAAATCC, forward AGCGCTGACCTTGCTATCGA ATC） and probe （5＇ Fam-AGTGCCGATCAGCCCTCCACCG-BHQ 1 3＇）. The ideal primer/probe concentration was.determined through optimization by comparing the lowes. thres.old concentration （Ct） values.us.ng the plas.id cDNA as.a template.
