Response surface methodology-based optimization of ultrasound-assisted extraction of β-sitosterol and lupeol from Astragalus atropilosus(roots) and validation by HPTLC method

Objective:To optimize the ultrasonication method for efficient extraction ofβ-sitosterol and lupeol from the roots of Astragalus atropilosus using Box-Behnken design of response surface methodology(RsM),and its validation by high performance thin layer chromatography(HPTLC)***:ultrasonication method was used to extractβ-sitosterol and lupeol from Astragalus atropilosus(roots).RsM was used to optimize the different extraction parameters *** to solid ratio(10–14 m L/g),temperature(60-80℃)and time(40–60 min)to maximize the yield ofβ-sitosterol and *** quantitative estimation ofβ-sitosterol and lupeol was done in chloroform extract of Astragalus atropilosus by validated HPTLC method on 10 cm×20 cm glass-backed silica gel 60 F254 plate using hexane and ethyl acetate(8:2,v/v)as mobile ***:A quadratic polynomial model was found to be most appropriate with regard to R1(yield of total extraction;R2/%CV=0.9948/0.28),R2(β-sitosterol yield;R2/%CV=0.9923/0.39)and R3(lupeol yield;R2/%CV=0.9942/0.97).The values of adjusted R2/predicted R2/signal to noise ratio for R1,R2,and R3 were 0.9782/0.9551/48.77,0.9904/0.9110/31.33,and 0.9927/0.9401/36.08,respectively,indicating a high degree of correlation and adequate *** linear correlation plot between the predicted and experimental values for R1,R2,and R3 showed high values of R2 ranging from 0.9905-0.9973.β-sitosterol and lupeol in chloroform extract of Astragalus atropilosus were detected at Rf values of 0.22 and 0.34,respectively,atλmax=518 *** optimized ultrasonic extraction produced 8.462%w/w of R1,0.451%w/w of R2 and 0.172%w/w of R3 at 13.5 m L/g liquid to solid ratio,78℃of temperature and 60 min of ***:The experimental findings of RsM optimized extraction and HPTLC analysis can be further applied for the efficient extraction ofβ-sitosterol and lupeol in other species of Astragalus.

BACKGROuND Vaccine hesitancy is a major challenge in the fight against the coronavirus disease 2019(COVID-19)*** the sociodemographic factors associated with vaccine acceptance among Nigerians is crucial for improving vaccine *** To assess the acceptance rate of COVID-19 vaccine and its related determinants among *** An online cross-sectional survey(observational study)was conducted between February 2021 and May 2021,using a questionnaire hosted on *** invitation to take part in the poll was sent out to participants through social networking platforms.A logistic regression was used to determine which sociodemographic factors were associated with vaccine acceptance *** A total of 1800 persons responded to the survey,a larger proportion of whom were males(53.9%)and within the age group of 21-30 years(29.4%)and earned an average income of less than$500 per month(43.3%).Only 0.56%of participants had a high perceived risk of COVID-19 infection,while only 1.11%had a perceived risk of dying from *** perception rate of the COVID-19 vaccine among participants was 51.1%,while the acceptance rate was 63.9%.There was no significant association between the COVID-19 vaccine acceptance rate and related determinants assessed,particularly age(χ²=3.049,P=0.550),sex(χ²=0.102,P=0.749),average income(χ²=3.802,P=0.875),and religion(χ²=2.819,P=0.420).Participants with chronic conditions demonstrated a higher acceptance rate compared to the general *** Despite the positive perception observed and substantial vaccine acceptance rate among the study participants,more public health interventions are still needed to enhance vaccine acceptability in Nigeria.

In this paper, we explore the spanning simplicial complex of wheel graph Wn on vertex set [n]. Combinatorial properties of the spanning simplicial complex of wheel graph are discussed, which are then used to compute the f-vector and Hilbert series of face ring k[Δs(Wn)] for the spanning simplicial complex Δs(Wn). Moreover, the associated primes of the facet ideal IF(Δs(Wn)) are also computed.

Inflow and outflow permeability tests in a very soft clay under low stresses

Influence of previous plane of nutrition on molecular mechanisms regulating the expression of urea and water metabolism related genes in the rumen and kidney of finishing crossbred Angus steers

This study aimed to understand how molecular mechanisms controlling water and urea metabolism at the finishing phase can be affected by previous plane of nutrition of crossbred Angus beef steers. Twentyfour(n = 24) animals were randomly distributed into either a moderate(MP) or high plane of nutrition during the background phase for 85 d. Animals were then blocked by their previous plane and were moved onto a 105-d finishing phase in a 2 × 2 factorial arrangement. The forage-finished group received only high-quality alfalfa hay, whereas the grain-fed group received a high grain diet(80% whole corn and20% alfalfa hay). By the end of the finishing phase, animals were harvested and tissue samples from the rumen and kidney were collected. Changes in gene expression of aquaporins(AQP)-2,-3,-4,-7, ATP1A1,ATP1B1, sGK1, CLIC1(kidney and rumen), uT-A1(kidney only) and uT-B(rumen only), were assayed via real-time q PCR; 18s r RNA was used as an endogenous control. One-way ANOVA followed by Tukey's post hoc analysis was conducted. When animals were from MP, forage-finishing increased the relative abundance of AQP3(P ≤ 0.05), AQP7(P ≤ 0.05), ATP1B1(P ≤ 0.05), and sGK1(P ≤ 0.05) in the kidney when compared to grain-fed animals. In the rumen, for the MP group, AQP7 was differentially expressed in both treatments at the finishing phase(P ≤ 0.01), with forage-finished steers having the highest expression of AQP7. For the MP group, uT-B had a tendency of presenting a higher expression on grainfed animals(P = 0.075). Overall, these results suggest that previous plane can impact expression of genes associated with water and urea metabolism during the finishing phase, namely AQP3, AQP7, ATP1B1, and sGK1 in the kidney, and AQP7 and uT-B in the rumen. The greatest impact observed on gene expression changes of investigated genes at the finishing phase was reflective of animals backgrounded on the moderate previous plane.

This paper considers a nonsmooth semi-infinite minimax fractional programming problem（sIMFP） involving locally Lipschitz invex functions. The authors establish necessary optimality conditions for sIMFP. The authors establish the relationship between an optimal solution of sIMFP and saddle point of scalar Lagrange function for sIMFP. Further, the authors study saddle point criteria of a vector Lagrange function defined for sIMFP.

Nursery practice on seed germination and seedling growth of Dalbergia sissoo using beneficial microbial inoculants

Nursery practice using microbial inoculants was performed to find out the efficacy of the inoculants on seed germination and seedling growth of sissoo（Dalbergia sissoo Roxb.）.Microbial inoculants or effective microorganisms（EM） are a mixture of many different beneficial microorganisms in a *** seedlings were grown in a mixture of sandy soil and cowdung（3：1） kept in polybags with pouring EM solution at different concentrations（0.1%,0.5%,1%,2%,5% and 10%） before and after a week of sowing the *** germination rate and growth parameters of seedlings were measured,such as,shoot and root length,vigor index,fresh and dry weight of shoot and root and total biomass *** nodulation status influenced by EM was also observed along with the measurement of pigment contents in *** highest germination rate（69%） was observed in 2% EM treatment,followed by 67% and 65% in 1% and 5% *** highest shoot length（33.2 cm） was in 2% EM,whereas highest root length（26.3 cm） was in 1% *** fresh and dry weights from shoot and root,were maximum（4.16 g and 1.57 g;2.12 g and 0.83 g respectively） in 2% EM and were significantly（p ≤ 0.05） different from *** index was highest（4071） in 2% EM,which was significantly（p ≤ 0.05） different from *** dry weight increment was highest in 2% EM treatment,followed by 1% and 5% concentrations of *** number was higher at very low（0.1%） concentration of EM solution but it normally decreased with the increase of EM *** contents of chlorophyll a,chlorophyll b and carotenoid were highest（60.11,17.05 and 42.48 mg·L1respectively） in 2% EM treatment and lowest（39.35,13.55 and 27.29 mg·L-1 respectively） in control ***,low concentration of EM（up to 2%） can be used for getting maximum seed germination rate and seedling development of Dalbergia sissoo Roxb.

Hybrid Active Contour Mammographic Mass segmentation and Classification

This research implements a novel segmentation of mammographic *** methods are proposed,namely,segmentation of mass based on iterative active contour,automatic region growing,and fully automatic mask selectionbased active contour *** the first method,iterative threshold is performed for manual cropped preprocessed image,and active contour is applied *** overcome manual cropping in the second method,an automatic seed selection followed by region growing is *** that the result is only a few images owing to over segmentation,the third method uses a fully automatic active *** of the segmentation techniques are compared with the manual markup by experts,specifically by taking the difference in their mean ***,the difference in the mean value of the third method is 1.0853,which indicates the closeness of the ***,the proposed method is compared with the existing fuzzy C means and level set *** automatic mass segmentation based on active contour technique results in segmentation with high *** using adaptive neuro fuzzy inference system,classification is done and results in a sensitivity of 94.73%,accuracy of 93.93%,and Mathew’s correlation coefficient(MCC)of 0.876.
