While the adverse effects of glucocorticoids on bone are well described, positive effects of glucocorticoids on the differentiation of osteoblasts are also observed. These paradoxical effects of glucocorticoids are dose dependent. At both physiologicaland supraphysiological levels of glucocorticoids, osteoblasts and osteocytes are the major glucocorticoid target cells. However, the response of the osteoblasts to each of these is quite distinct. At physiology levels, glucocorticoids direct mesenchymal progenitor cells to differentiate towards osteoblasts and thus increase bone formation in a positive way. In contrast with ageing, the excess production of glucocorticoids, at both systemic and intracellular levels, appear to impact on osteoblast and osteocytes in a negative way in a similar fashion to that seen with therapeutic glucocorticoids. This review will focus on therole of glucocorticoids in normal bone physiology, with particular emphasis on the mechanism by which endogenous glucocorticoids impact on bone and its constituent cells.

We report here a method for the use of poly-L-lysine （PLL） to markedly improve the adenoviral transduction efficiency of primary murine osteoblasts and bone marrow stromal cells （BMsCs） in culture and in situ, which are typically difficult to transduce. We show by fluorescence microscopy and flow cytometry that the addition of PLL to the viral-containing medium significantly increases the number of green fluorescence protein （GFP）-positive osteoblasts and BMsCs transduced with an enhanced GFP-expressing adenovirus. We also demonstrate that PLL can greatly enhance the adenoviral transduction of osteoblasts and osteocytes in situ in ex vivo tibia and calvaria, as well as in long bone fragments. In addition, we validate that PLL can improve routine adenoviral transduction studies by permitting the use of low multiplicities of infection to obtain the desired biologic effect. Ultimately, the use of PLL to facilitate adenoviral gene transfer in osteogenic cells can provide a cost-effective means of performing efficient gene transfer studies in the context of bone research.

Interaction between the intracluster medium and the interstellar media of galaxies via ram-pressure stripping (RPs) has ample support from both observations and simulations of galaxies in clusters. some, but not all of the observations and simulations show a phase of increased star formation compared to normal spirals. Examples of galaxies undergoing RPs induced star formation in clusters experiencing a merger have been identified in high resolution optical images supporting the existence of a star formation phase. We have selected Abell 3266 to search for ram-pressure induced star formation as a global property of a merging cluster. Abell 3266 (z = 0.0594) is a high mass cluster that features a high velocity dispersion, an infalling subcluster near to the line of sight, and a strong shock front. These phenomena should all contribute to making Abell 3266 an optimum cluster to see the global effects of RPs induced star formation. Using archival X-ray observations and published optical data, we cross-correlate optical spectral properties ([OII, Hβ]), indicative of starburst and post-starburst, respectively with ram-pressure, ρvsup>2sup>, calculated from the X-ray and optical data. We find that post-starburst galaxies, classified as E + A, occur at a higher frequency in this merging cluster than in the Coma cluster and at a comparable rate to intermediate redshift clusters. This is consistent with increased star formation due to the merger. However, both starburst and post-starburst galaxies are equally likely to be in a low or high ram pressure environment. From this result, we infer that the duration of the starburst phase must be very brief so that: 1) at any time only a small fraction of the galaxies in a high ram pressure environment show this effect, and 2) most post-starburst galaxies are in an environment of low ram pressure due to their continued orbital motion in the cluster.

It is believed that human ancestors evolved the ability to run bipedally approximately 2 million years ago. This form of locomotion may have been important to our survival and likely has influenced the evolution of our body form. As our bodies have adapted to run, it seems unusual that up to 79% of modern day runners are injured annually. The etiology of these injuries is clearly multifactorial. However, 1 aspect of running that has significantly changed over the past 50 years is the footwear we use. Modern running shoes have become increasingly cushioned and supportive, and have changed the way we run. In particular, they have altered our footstrike pattern from a predominantly forefoot strike(FFs) landing to a predominantly rearfoot strike(RFs) landing. This change alters the way in which the body is loaded and may be contributing to the high rate of injuries runners experience while engaged in an activity for which they were adapted. In this paper, we will examine the benefits of barefoot running(typically an FFs pattern),and compare the lower extremity mechanics between FFs and RFs. The implications of these mechanical differences, in terms of injury, will be discussed. We will then provide evidence to support our contention that FFs provides an optimal mechanical environment for specific foot and ankle structures, such as the heel pad, the plantar fascia, and the Achilles tendon. The importance of footwear will then be addressed, highlighting its interaction with strike pattern on mechanics. This analysis will underscore why footwear matters when assessing mechanics. Finally, proper preparation and safe transition to an FFs pattern in minimal shoes will be emphasized. Through the discussion of the current literature, we will develop a justification for returning to running in the way for which we were adapted to reduce running-related injuries.

Piezoelectric resonant de-icing systems are attracting great *** paper aims to assess the implementation of these systems at the aircraft *** article begins with the model to compute the power requirement of a piezoelectric resonant de-icing system sized from the prototype detailed in Part 1/2 of this *** the mass,drag,and fuel consumption of this system and the subcomponents needed for its implementation are *** features of a piezoelectric resonant de-icing system are finally computed for aircraft similar to Airbus A320 aircraft and aircraft of different categories(Boeing 787,ATR 72 and TBM 900)and compared with the existing thermal and mechanical ice protection systems.A sensitivity analysis of the main key sizing parameters of the piezoelectric de-icing system is also performed to identify the main axes of improvement for this *** study shows the potential of such ice protection *** particular,for the realistic input parameters chosen in this work,the electro-mechanical solution can provide a 54% reduction in terms of mass and a 92% reduction in terms of power consumption for an A320 aircraft architecture,leading to a 74% decrease in the associated fuel consumption compared to the actual air bleed system.
