Ellagic acid induces apoptosis through inhibition of nuclear factor κB in pancreatic cancer cells

AIM： To determine the effect of ellagic acid on apoptosis and proliferation in pancreatic cancer cells and to determine the mechanism of the pro-survival effects of ellagic acid.METHODs： The effect of ellagic acid on apoptosis was assessed by measuring Phosphatidylserine externalization, caspase activity, mitochondrial membrane potential and DNA fragmentation; and proliferation by measuring DNA thymidine incorporation. Mitochondrial membrane potential was measured in permeabilized cells, and in isolated mitochondria. Nuclear factor κB （NF-κB） activity was measured by electromobility shift assay （EMsA）. REsULTs： We show that ellagic acid, a polyphenolic compound in fruits and berries, at concentrations 10 to 50 mmol/L stimulates apoptosis in human pancreatic adenocarcinoma cells. Further, ellagic acid decreases proliferation by up to 20-fold at 50 mmol/L. Ellagic acid stimulates the mitochondrial pathway of apoptosis associated with mitochondrial depolarization, cytochrome C release, and the downstream caspase activation. Ellagic acid does not directly affect mitochondria. Ellagic acid dose-dependently decreased NF-κB binding activity. Furthermore, inhibition of NF-κB activity using IkB wild type plasmid prevented the effect of ellagic acid on apoptosis. CONCLUsION： Our data indicate that ellagic acid stimulates apoptosis through inhibition of the prosurvival transcription factor NF-κB.

Analysis of Oseltamivir Resistance substitutions in Influenza Virus Glycoprotein Neuraminidase using a Lentivirus-Based surrogate Assay system

Influenza A virus poses a great threat to global health, and oseltamivir (trade marked as Tamiflu), which targets influenza surface glycoprotein neuraminidase (NA), is used clinically as a major anti-influenza treatment. However, certain substitutions in NA can render an influenza virus resistant to this drug. In this study, using a lentiviral pseudotyping system, which alleviates the safety concerns of studying highly pathogenic influenza viruses such as avian influenza H5N1, that utilizes influenza surface glycoproteins (hemagglutinin or HA, and NA) and an HIV-core combined with a luciferase reporter gene as a surrogate assay, we first assessed the functionality of NA by measuring pseudovirion release in the absence or presence of oseltamivir. We demonstrated that oseltamivir displays a dose-dependent inhibition on NA activity. In contrast, a mutant NA (H274Y) is more resistant to oseltamivir treatment. In addition, the effects of several previously reported substitution NA mutants were examined as well. Our results demonstrate that this lentivirus-based pseudotyping system provides a quick, safe, and effective way to assess resistance to neuraminidase inhibitors. And we believe that as new mutations appear in influenza isolates, their impact on the effectiveness of current and future anti-NA can be quickly and reliably evaluated by this assay.

Incident frailty and cognitive impairment by heart failure status in older patients with atrial fibrillation: the sAGE-AF study

Background Atrial fibrillation(AF) and heart failure(HF) frequently co-occur in older individuals. Among patients with AF, HF increases risks for stroke and death, but the associations between HF and incident cognition and physical impairment remain unknown. We aimed to examine the cross-sectional and prospective associations between HF, cognition, and frailty among older patients with AF. Methods The sAGE-AF(systematic Assessment of Geriatric Elements in AF) study enrolled 1244 patients with AF(mean age 76 years, 48% women) from five practices in Massachusetts and Georgia. HF at baseline was identified from electronic health records using ICD-9/10 codes. At baseline and 1-year, frailty was assessed by Cardiovascular Health survey score and cognition was assessed by the Montreal Cognitive Assessment. Results Patients with prevalent HF(n = 463, 37.2%) were older, less likely to be non-Hispanic white, had less education, and had greater cardiovascular comorbidity burden and higher CHA2 Ds2 VAsC and HAs-BLED scores than patients without HF(all P's justed regression models, HF(present vs. absent) was associated with both prevalent frailty(adjusted odds ratio [a OR]: 2.38, 95% confidence interval [CI]: 1.64-3.46) and incident frailty at 1 year(a OR: 2.48, 95% CI: 1.37-4.51). HF was also independently associated with baseline cognitive impairment(a OR: 1.60, 95% CI: 1.22-2.11), but not with developing cognitive impairment at 1 year(a OR 1.04, 95%CI: 0.64-1.70). Conclusions Among ambulatory older patients with AF, the co-existence of HF identifies individuals with physical and cognitive impairments who are at higher short-term risk for becoming frail. Preventive strategies to this vulnerable subgroup merit consideration.

Key changes across valley floors in mountain areas affected by different levels of human impact: a case study of the Ochotnica river catchment in the Polish Carpathians

The goal of this paper was to present knowledge on changes in the morphodynamic structure of the Ochotnica River(Polish Carpathians)and transformations within its active river zone since the end of the 19th *** study used a set of archival and contemporary cartographic materials and the results of three-fold field mapping of the morphodynamic structure of the *** and indirect human interference with the natural environment of the catchment and the fluvial system was taken into account in the *** of changes in the morphodynamic structure of the Ochotnica and its active river zone since the end of the 19th century indicated significant changes in the development trend of the *** contemporary structure of the Ochotnica is more complex than in the *** has been a fragmentation of the riverbed into sections with different morphodynamic *** proportion of erosive sections has increased,which explains the reduced in the width of the active river zone of the *** average width of the active river zone of the river between 1861 and 2022 has changed from 80 m to 18 *** impact has been identified as the main reason for the changes in the morphodynamic structure of the riverbed and its active river ***,only the unregulated sect ion is characterised by free lateral migration of the riverbed and the widest active river zone(~28 m).

Characteristics of Nb/Al superconducting tunnel junctions fabricated using ozone gas

To improve the energy resolution（？E） of Nb/Al superconducting tunnel junctions（sTjs）, an ozone（O3） oxidation process has been developed to fabricate a thin defect-free tunnel barrier that simultaneously shows high critical current jC〉 1000 A/cm^2 and high normalized dynamic resistance RDA 〉 100 MΩ·μm^2, where A is the size of the sTj. The 50-μm^2 sTjs produced by O3 exposure of 0.26 Pa·min with an indirect spray of O3 gas, which is a much lower level of exposure than the O2 exposure used in a conventional O2 oxidation process, exhibit a maximum jC= 800 A/cm^2 and a high RDA = 372 MΩ ·μm^2. The 100-pixel array of the 100-μm^2sTjs produced using the same O3 oxidation conditions exhibits a constant leak current I leak= 14.9 ± 3.2 n A at a bias point around △ /e（where e is half the energy gap of an sTj）,and a high fabrication yield of 87%. Although the I leak values are slightly larger than those of sTjs produced using the conventional O2 oxidation process, the sTj produced using O3 oxidation shows a ？E = 10 eV for the C-Kα line, which is the best value of our Nb/Al sTj x-ray detectors.

The beginnings of laparoscopic liver resection(LLR)were at the start of the 1990s,with the initial reports being published in 1991 and *** were followed by reports of left lateral sectionectomy in *** the years following,the procedures of LLR were expanded to hemi-hepatectomy,sectionectomy,segmentectomy and partial resection of posterosuperior segments,as well as the parenchymal preserving limited anatomical resection and modified anatomical(extended and/or combining limited)resection *** expanded range of LLR procedures,mimicking the expansion of open liver resection in the past,was related to advances in both technology(instrumentation)and technical skill with conceptual *** this period of remarkable development,two international consensus conferences were held(2008 in Louisville,KY,United states,and 2014 in Morioka,japan),providing up-to-date summarizations of the status and perspective of *** advantages of LLR have become clear,and include reduced intraoperative bleeding,shorter hospital stay,and-especially for cirrhotic patients-lower incidence of complications(e.g.,postoperative ascites and liver failure).In this paper,we review and discuss the developments of LLR in operative procedures(extent and style of liver resections)during the first quarter century since its inception,from the aspect of relationships with technological/technical developments with conceptual changes.

Investigation of newly developed added damping and stiffness device with low yield strength steel

Energy dissipators, isolated-resistant and specific structural forms for earthquake resistance are popular topics in the research to improve shock-resistance. In this work, experimental methods were used to investigate the property of low yield strength steel. Carbon content in LYs material is lower than that in other steels; the ultimate stress is three times the yield stress. The ultimate elongation rate is about 62% and the ductility is 23 times that of A36 steel. In order to overcome some defects of ordinary use metallic dampers, the mechanical characteristic of low yield strength steel is used to develop added damping and stiffness for rhombic steel plate absorber. Test of the energy dissipation behavior for this newly de-veloped device indicated that LYs could stably dissipate or absorb the input energy of earthquake. Then, the analytical model for the hysteretic behavior of this new device is proposed. Comparison of experimental data and numerical simulation results showed that this analytical model is suitable for simulating the hysteretic energy behavior of this new device.

Development and Applications of Ruggedized VIs/NIR spectrometer system for Oilfield Wellbores

The development and applications of a ruggedized visible to near-infrared （VIs/NIR） spectrometer system capable of measuring fluid spectra in oilfield wellbores are presented. Real-time assessment of formation fluid properties penetrated by an oilfield wellbore is critically important for oilfield operating companies to make informed decisions to optimize the development plan of the well and hydrocarbon reservoir. A ruggedized VIs/NIR spectrometer was designed and built to measure and analyze hydrocarbon spectra reliably under the harsh conditions of the oilfield wellbore environment, including temperature up to 175 ~C, pressure up to 170MPa, and severe mechanical shocks and vibrations. The accuracy of hydrocarbon group composition analysis was compared well with gas chromatography results in the laboratory.
