Typing of pancreatic cancer-associated fibroblasts identifies different subpopulations

AIM To determine whether it is possible to identify different immune phenotypic subpopulations of cancer-associated fibroblasts (CAFs) in pancreatic cancer (PC). METHODs We defined four different stromal compartments in surgical specimens with PC: The juxtatumoural, peripheral, lobular and septal stroma. Tissue microarrays were produced containing all pre-defined PC compartments, and the expression of 37 fibroblast (FB) and 8 extracellular matrix (ECM) markers was evaluated by immunohistochemistry, immunofluorescence (IF), double-IF, and/or in situ hybridization. The compartment-specific mean labelling score was determined for each marker using a four-tiered scoring system. DOG1 gene expression was examined byquantitative reverse transcription PCR (qPCR). REsULTs CD10, CD271, cytoglobin, DOG1, miR-21, nestin, and tenascin C exhibited significant differences in expression profiles between the juxtatumoural and peripheral compartments. The expression of CD10, cytoglobin, DOG1, nestin, and miR-21 was moderate/strong in juxtatumoural CAFs (j-CAFs) and barely perceptible/weak in peripheral CAFs (p-CAFs). The upregulation of DOG1 gene expression in PC compared to normal pancreas was verified by qPCR. Tenascin C expression was strong in the juxtatumoural ECM and barely perceptible/weak in the peripheral ECM. CD271 expression was barely perceptible in j-CAFs but moderate in the other compartments. Galectin-1 was stronger expressed in j-CAFs vs septal fibroblasts, PDGFRβ, tissue transglutaminase 2, and hyaluronic acid were stronger expressed in lobular fibroblasts vs p-CAFs, and plectin-1 was stronger expressed in j-CAFs vs l-FBs. Effects of a novel bacterial phytase expressed in Aspergillus Oryzae on digestibility of calcium and phosphorus in diets fed to weanling or growing pigs

In 2 experiments, 48 weanling （initial BW： 13.5 ± 2.4 kg, Exp. 1） were used to determine effects of a novel bacterial 6-phytase and 24 growing pigs （initial BW： 36.2 ± 4.0 kg, Exp. 2） expressed in Aspergillus oryzae on the apparent total tract digestibility （ATTD） of phosphorus and calcium in corn-soybean meal diets fed to weanling and growing pigs. In Exp. 1 and 2, pigs were randomly allotted to 6 dietary treatments using a randomized complete block design and a balanced 2 period changeover design, respectively. In both experiments, 6 diets were formulated. The positive control diet was a corn-soybean meal diet with added inorganic phosphorus （Exp. 1：0.42 and 0.86% standardized total tract digestible phosphorus and total calcium, respectively; Exp. 2：0.32 and 0.79% standardized total tract digestible phosphorus and total calcium, respectively）. A negative control diet and 4 diets with the novel phytase （Ronozyme HiPhos, DsM Nutritional Products Inc., Parsippany, N J） added to the negative control diet at levels of 500, 1,000, 2,000, and 4,000 phytase units （FYT）/kg were also formulated. In Exp. 1, the ATTD of phosphorus was greater （P 〈 0.01） for the positive control diet （60.5%） than for the negative control diet （40.5%）, but increased （linear and quadratic, P 〈 0.01） as phytase was added to the negative control diet （40.5% vs. 61.6%, 65.1%, 68.7%, and 68.0%）. The breakpoint for the ATTD of phosphorus （68.4%） was reached at a phytase inclusion level of 1,016 FYT/kg. In Exp. 2, the ATTD of phosphorus was greater （P〈 0.01） for the positive control diet （59.4%） than for the negative control diet （39.8%） and increased （linear and quadratic, P〈 0.01） as phytase was added to the negative control diet （39.8% vs. 58.1%, 65.4%, 69.1%, and 72.8%）. The breakpoint for the ATTD of phosphorus （69.1%） was reached at a phytase inclusion level of 801 FYT/kg. in conclusion, the novel bacterial 6-phytase improved the ATTD of phosphorus and calcium in both weanling and growing pigs.

The pathogenesis of inflammatory bowel disease (IBD) is complex and largely unknown. Until recently, research has focused on the study of protein regulators in inflammation to reveal the cellular and molecular networks in the pathogenesis of IBD. However, in the last few years, new and promising insights have been generated from studies describing an association between an altered expression of a specific class of non-coding RNAs, called microRNAs (miRs or miRNAs) and IBD. The short (approximately 22 nucleotides), endogenous, single-stranded RNAs are evolutionary conserved inanimals and plants, and regulate specific target mRNAs at the post-transcriptional level. MiRNAs are involved in several biological processes, including development, cell differentiation, proliferation and apoptosis. Furthermore, it is estimated that miRNAs may be responsible for regulating the expression of nearly one-third of the genes in the human genome. Thus, miRNA deregulation often results in an impaired cellular function, and a disturbance of downstream gene regulation and signaling cascades, suggesting their implication in disease etiology. Despite the identification of more than 1900 mature human miRNAs, very little is known about their biological functions and functional targets. Recent studies have identified dysregulated miRNAs in tissue samples of IBD patients and have demonstrated similar differences in circulating miRNAs in the serum of IBD patients. Thus, there is great promise that miRNAs will aid in the early diagnosis of IBD, and in the development of personalized therapies. Here, we provide a short review of the current state-of-the-art of miRNAs in IBD pathogenesis, diagnostics and therapeutics.

A method of estimating the Martian neutral atmospheric density at 130 km, and comparison of its results with Mars Global surveyor and Mars Odyssey aerobraking observations based on the Mars Climate Database outputs

Profiles of the Martian dayside ionosphere can be used to derive the neutral atmospheric densities at 130 km,which can also be obtained from the Mars Climate Database(MCD)and spacecraft aerobraking *** this research,we explain the method used to calculate neutral densities at 130 km via ionosphere observations and three long-period 130-km neutral density data sets at northern high latitudes(latitudes>60°)acquired through ionospheric data measured by the Mars Global surveyor(MGs)Radio Occultation *** calculated 130-km neutral density data,along with 130-km density data from the aerobraking observations of the MGs and Mars Odyssey(ODY)in the northern high latitudes,were compared with MCD outputs at the same latitude,longitude,altitude,solar latitude,and local *** 130-km density data derived from both the ionospheric profiles and aerobraking observations were found to show seasonal variations similar to those in the MCD *** a negative shift of about 2×10^10 cm^−3,the corrected 130-km neutral densities derived from MCD v4.3 were consistent with those obtained from the two different *** result means that(1)the method used to derive the 130-km neutral densities with ionospheric profiles was effective,(2)the MCD v4.3 data sets generally overestimated the 130-km neutral densities at high latitudes,and(3)the neutral density observations from the MGs Radio science Experiment could be used to calibrate a new atmospheric model of Mars.

Climate change-driven shifts in plant–soil feedbacks:a meta-analysis

Background:Climate change is expected to affect plant–soil feedbacks(PsFs,i.e.,the effects of a plant on the growth of another plant or community grown in the same soil via changes in soil abiotic and biotic properties),influencing plant community dynamics and,through this,ecosystem ***,our knowledge of the effects of climate changes on the magnitude and direction of PsFs remains limited,with considerable variability between *** quantified PsFs associated with common climate change factors,specifically drought and warming,and their corresponding ambient(control)conditions using a meta-analytical *** investigated whether drought and warming effects on PsFs were consistent across functional groups,life histories(annual versus perennial)and species origin(native versus non-native),planting(monoculture,mixed culture)and experimental(field,greenhouse/laboratory)***:PsFs were negative(a mechanism that encourage species co-existence)under drought and neutral under corresponding ambient conditions,whereas PsFs were negative under both ambient and elevated temperatures,with no apparent difference in effect *** response to drought was largely driven by stronger negative PsFs in grasses,indicating that grasses are more likely to show stronger negative PsFs than other functional groups under ***,non-native species showed negative drought-induced PsFs while native species showed neutral PsFs under *** contrast,we found the opposite in pattern in response to warming for native and non-native *** herbs displayed stronger drought-induced negative PsFs than annual *** species communities displayed more negative PsFs than monocultures,independent of climate ***,warming and drought treatment PsF effect sizes were more negative in experiments performed in the field than under controlled ***:We provide evidence that drought and warming can induce context-specific

Association between Depression, Pressure Pain sensitivity, stress and Autonomous Nervous system Function in stable Ischemic Heart Disease: Impact of Beta-Adrenergic Receptor Blockade

Background: Depression and ischemic heart disease (IHD) are associated with persistent stress and autonomic nervous system (ANs) dysfunction. The former can be measured by pressure pain sensitivity (PPs) of the sternum, and the latter by the PPs and systolic blood pressure (sBP) response to a tilt table test (TTT). Beta-blocker treatment reduces the efferent beta-adrenergic ANs function, and thus, the physiological stress response. Objective: To test the effect of beta-blockers on changes in depression score in patients with IHD, as well as the influence on persistent stress and ANs dysfunction. Methods: Three months of non-pharmacological intervention aiming at reducing PPs and depression score in patients with stable IHD. Beta-blocker users (N = 102) were compared with non-users (N = 75), with respect to signs of depression measured by the Major Depressive Inventory questionnaire (MDI), resting PPs, and PPs and sBP response to TTT. Results: MDI score decreased 30% in non-users (p = 0.005) compared to 4% (p > 0.1) among users (between-group p = 0.003;effect size = 0.4). Resting PPs decreased in both the groups. Among most vulnerable patients with MDI ≥ 15, reductions in MDI score and resting PPs score correlated in non-users, only (r = 0.69, p = 0.007). Reduction in resting PPs correlated with an increase in PPs and sBP response to TTT. Conclusions: stress intervention in patients with IHD was anti-depressive in non-users, only. similarly, the association between the reduction in depression, reduction in persistent stress, and restoration of ANs dysfunction was only seen in non-users, suggesting a central role of beta-adrenergic receptors in the association between these factors.
