Micromolar sodium fluoride mediates anti-osteoclastogenesis in Porphyromonas gingivalis-induced alveolar bone loss

Osteoclasts are bone-specific multinucleated cells generated by the differentiation of monocyte/macrophage lineage precursors. Regulation of osteoclast differentiation is considered an effective therapeutic approach to the treatment of bone-lytic diseases. Periodontitis is an inflammatory disease characterized by extensive bone resorption. In this study, we investigated the effects of sodium fluoride （NaF） on osteoclastogenesis induced by Porphyromonas gingivalis, an important colonizer of the oral cavity that has been implicated in periodontitis. NaF strongly inhibited the P. gingivalis-induced alveolar bone loss. That effect was accompanied by decreased levels of cathepsin K, interleukin （IL）-1β, matrix metalloproteinase 9 （MMP9）, and tartrate-resistant acid phosphatase, which were up-regulated during P. gingivalis-induced osteoclastogenesis. Consistent with the in vivo anti-osteoclastogenic effect, NaF inhibited osteoclast formation caused by the differentiation factor RANKL （receptor activator of nuclear factor KB ligand） and macrophage colony-stimulating factor （M-CSF）. The RANKL-stimulated induction of the transcription factor nuclear factor of activated T cells （NFAT） cl was also abrogated by NaF. Taken together, our data demonstrate that NaF inhibits RANKL-induced osteoclastogenesis by reducing the induction of NFATcl, ultimately leading to the suppressed expression of cathepsin K and MMP9. The in vivo effect of NaF on the inhibition of P. gingivalis-induced osteoclastogenesis strengthens the potential usefulness of NaF for treating periodontal diseases.

Rat embryonic stem cells create new era in development of genetically manipulated rat models

Embryonic stem(ES) cells are isolated from theinner cell mass of a blastocyst, and are used for the generation of gene-modified animals. In mice, the transplantation of gene-modified ES cells into recipient blastocysts leads to the creation of gene-targeted mice such as knock-in and knock-out mice; these gene-targeted mice contribute greatly to scientific development. Although the rat is considered a useful laboratory animal alongside the mouse, fewer genemodified rats have been produced due to the lack of robust establishment methods for rat ES cells. A new method for establishing rat ES cells using signaling inhibitors was reported in 2008. By considering the characteristics of rat ES cells, recent research has made progress in improving conditions for the stable culture of rat ES cells in order to generate gene-modified rats efficiently. In this review, we summarize several advanced methods to maintain rat ES cells and generate gene-targeted rats.

Measurements of Characteristics of Electromagnetic Radiation Caused by Electrostatic Discharges in Metal Sphere Gap at Voltages Less than 1 kV

In this study,the characteristics of Electromagnetic(EM) radiation caused by Electrostatic Discharges(ESDs) from metal spheres charged to voltages less than 1 kV are examined *** experimental system consists of a pair of spherical electrodes of different diameters,a 1-18 GHz-bandwidth horn antenna and a 20-GHz-bandwidth digitizing ***,waveform duration and peaks of antenna-received voltages from the EM field radiation are measured in order to clarify the EM radiation *** ratio of the received voltages between the antenna arrangements of the field polarization parallel and perpendicular to the spark pass is 18 to 20 *** polarities of the antenna-received voltages are the same as those of the charge voltages across the ***,the waveform duration and the first peaks increase with an increase in the diameters of the spherical ***,we find that the polarization,waveform duration and first peaks of the EM field radiation can be explained by a dipole antenna structure,which makes the spark part of the spherical electrodes a feeding point on the straight line passing through the centres of the two spheres.

We are dealing with the problem of maximizing the weighted number of pseudo just-in-time jobs on single machine model. The concept of pseudo just-in-time is a relaxation of just-in-time. In our formulation, each job is associated with a time-window instead of due date, and the lengths of all such time-windows are bounded above by a certain value given in advanced. A job is called pseudo just-in-time if it is completed within its time-window. We show that the problem is polynomial time solvable, and moreover, we show some results on computational complexity for more general settings.

Histological Findings of Bone Marrow Lesions on Magnetic Resonance Images in Patients with Varus Knee Osteoarthritis

Bone marrow lesions (BMLs) on magnetic resonance (MR) images in knee osteoarthritis patients are considered to predict the severity and progression of the disease. We evaluated the histological findings of BMLs on MR images of the subchondral area of the medial femoral condyle in varus osteoarthritic knees. In 24 patients with varus knee osteoarthritis who underwent total knee arthroplasty (TKA), sagittal T1- and T2-weighted MR images of the affected knee were acquired before TKA. During TKA, resected bone pieces from the distal medial femoral condyle were obtained. Sagittal specimens obtained from the center of the bone pieces were histologically examined. Twenty patients had BMLs. Histological findings of BMLs in the subchondral area showed various features, such as fibrovascular tissue, cyst formation, active bone remodeling with bone formation and bone resorption, and hyaline cartilage. BMLs were not found in four patients;histological findings of these patients showed normal bone marrow tissue with normal-thickness trabeculae. Subchondral bony end plate in knees with BMLs was usually thin or destroyed, while that without BMLs was thick or normal. The condition of the subchondral bony end plate would explain the differences in the severity and progression between patients with or without BMLs.

This paper presents an application of variable infinite impulse response （IIR） digital filters based on the Constantinides frequency transformation to adaptive filtering. We propose a new unified framework for adaptive detection of narrowband signals using variable band-stop IIR digital filters with arbitrary transfer functions. It is also shown that our proposed method includes the all-pass-based adaptive notch filters as a special case. A simulation example demonstrates the utility of our proposed method from the viewpoint of the signalto-noise ratio （SNR） improvement. This result shows that the Constantinides frequency transformation plays significant roles in not only the filter design, but also the adaptive signal processing.

Conscious Sedation and Awake Fiberoptic Intubation in a Patient with Difficult Mask Ventilation—A Case Report

Since maxillofacial malignancy is a common cause of facial defects and disfigurement of the face that may make fitting of a mask difficult and cause air leakage from the side, thus making mask ventilation difficult. In addition, distorted anatomy of the airway and base of the skull in such patients may cause difficult intubation (DI). We experienced a case with a huge facial defect due to maxillary carcinoma, in which difficult mask ventilation (DMV) and DI were predicted. After evaluation by three-dimensional airway computed tomography, the airway was secured with conscious sedation using dexmedetomidine, and awake fiberoptic intubation was safely performed. Three-dimensional airway computed tomography seems to be a good tool for successful intubation when DMV and DI are predicted.

An Unexpected Complication by Misplacement of an External Ventricular Drain;Transient Oculomotor Palsy Due to Direct Compression by a Drain—A Case Report

Background: Insertion of an external ventricular drain (EVD) is an important neurosurgical technique. There is a minor risk of misplacement, and complications including infections or hemorrhages have been reported. Transient neurological complications are, however, very rare, especially when EVD is placed under endoscope assistance. We report a case of unilateral transient oculomotor palsy after an EVD procedure. Case presentation: An 11-year-old boy with past medical history of moyamoya disease suffered from intraventricular hemorrhage and acute hydrocephalus. Insertion of EVD and hematoma removal was performed with endoscope assistance. After the surgery, transient oculomotor palsy occurred by the direct compression of the brain stem by the drainage tube. Conclusion: Transient unilateral oculomotor palsy due to the direct compression of the midbrain by an EVD tube is a very rare, but possible complication, even under endoscopic assistance. Attention through the procedure is required until skin closure to avoid tube dislocation.
