Correlation of Various Foods Intakes and Plasma Levels of Omega Fatty Acids in Healthy Japanese Old Men

Background: Trans fatty acids are considered to impair health and some ω fatty acids are protective against atherosclerosis and diabetes mellitus. Trans fatty acids are said to be formed by the partial hydrogenation of vegetable oils. Some amounts are produced in digestive organs of ruminants and present in dairy products or meat. It is important how much these intaken fatty acids influence their plasma levels. Methods: Plasma levels of fatty acids including transforms of healthy old men are measured by gas chromatography and correlations between various foods intakes and plasma levels of trans fatty acids, and ω fatty acids are examined. Results: Intake of fish resulted in increase in plasma levels of docosahexaenoic acid (DHA) and eicosapentaenoic acid (EPA) but intake of meat such as beef, cow and chicken meat did not increase plasma levels of arachidonic acid (AA). Intakes of oils increased plasma levels of dihomo-g-linolenic acid significantly and AA to some extent. Conclusion: Plasma levels of EPA and DHA increased upon intakes of fish in Japanese old men. Oil intake but not meat intake increased DGLA significantly. These results may explain low incidence of cardiovascular diseases in Japanese people compared with American people whose plasma levels of DHA and EPA are lower.

A Carbon Monoxide Sensing Film Based on Hemoglobin Allostery

Carbon monoxide (CO) is a gaseous mediator, which is generated via anenzymatic reaction of heme oxygenase, and it plays physiological roles to regulate cellular respiration and blood flow in the liver. The concentration and distribution of CO molecules in the living body is unknown owing to a lack of a suitable technique for measuring them in vivo. A needle-type CO sensor has been used for bioinstrumentation, but it is inappropriate for implantation in vivo and long-term monitoring. We developed a CO sensor sheet based on hemoglobin (Hb) allostery, as Hb undergoes a conformational change on CO binding. Hb was extracted from mice blood and mixed with agarose gel with a reducer to stabilize deoxy-Hb in the gel. CO-releasing molecules (CORM) were used to mimic CO-generating tissue, and the sensitivity of the Hb gel could be regulated by Hb concentration. We defined the CO–Hb index, an absorbance ratio at 539 and 557 nm, to estimate the accumulation of captured CO in the gel. It correlatively increased with CORM dose, indicating that gel-embedded Hb underwent a conformational change on CO binding, thereby acting as a CO sensor. We subsequently used the Hb-sensor sheet for two-dimensional imaging of CO distribution. CORM-containing gels with different sizes and doses were layered on this sheet. Size- and dose-dependent CO distribution was visualized by scanning the CO-Hb index in the sheet. Our Hb-based CO sensor sheet is composed of biocompatible materials and can be applied to detect low-level CO sources in the living body.

Portfolio Diversification Using Information Theory Applied to Brazilian Stocks

Using the concepts from information theory, it is possible to improve the traditional methodologies of asset allocation. In this paper, it was studied and extended the two existent approaches： the first is based on the Shannon entropy concept and the second on the Kullback-Leibler distance. In modem portfolio theory, the investor has two basic procedures： the choice of a portfolio that maximizes its risk-adjusted excess return or the mixed allocation between the maximum Sharpe portfolio and the risk-free asset. The first procedure was already addressed in the related literature. One important contribution of this paper is the consideration of the second procedure in the information theory context. The performance of these approaches was compared with the three traditional asset allocation methodologies： the Markowitz＇s mean-variance, the resampled mean-variance and the equally weighted portfolio. It was used simulated and real data from Brazilian stocks. The information theory-based methodologies were verified to be more robust when dealing with the estimation errors.

Is biliary bile acid a good predictor for acute cellular rejection in living donor liver transplantation?

BACKGROUND:In liver transplantation,acute cellular rejection(ACR)is still a major complication that can lead to *** secretion has been considered as a marker of early graft function. METHODS:The study included 41 adults who received living donor liver transplantation(LDLT)at Kyoto University Hospital between April 2007 and February 2008. The patients were stratified according to the presence or absence of *** samples were collected from donors once and from recipients every other day for the first 2 weeks after *** bile acid(BA)and taurine-conjugated bile acid(TCBA)in bile were measured by magnetic resonance *** recipient/donor (R/D)BA ratio and R/D TCBA ratio were calculated. RESULTS:The ACR group(n=12)showed a greater decrease in BA post-transplantation than the non-ACR group,but this difference was not statistically significant. On both day 7 and day 9 post-transplantation the R/D TCBA was significantly different between the two groups (P=0.038 on day 7 and P=0.036 on day 9).The R/D TCBA ratio≥0.5 on days 7 and 9,and≥0.38 on day 11 post- transplantation were associated with better ACR-free survival. CONCLUSION:The recipient/donor TCBA ratio can be a predictor for ACR after LDLT as early as post- transplantation day 7.

用转染EBV DNA Bam HI K片段后稳定表达EBNA-1的K_4细胞作为靶细胞,检测50份鼻咽癌病人和38份健康对照者血清中的IgG/EBNA-1抗体;阳性率分别为100％和92％。前者的平均几何滴度为89.4,后者为18.3,前者约为后者的5倍。同一批被检血清经SPA吸收去除IgG竞争性抑制后,鼻咽癌病人的IgA/EBNA-1抗体阳性率达78％(GMT 20.9),健康人的IgA/EBNA-1抗体阳性率仅5.3％,效价亦很低(GMT 5.2)。表明IgA/EBNA-1抗体对鼻咽癌是比较特异的,可考虑作为鼻咽癌血清学诊断的指标之一。

Background: It is not well analyzed whether there are differences in plasma levels of tryptophan (TRP) metabolites between healthy control people (HC) and patients of type II bipolar depression (BDII). Methods: Ultra high-speed liquid chromatography/mass spectrometry has been used for the simultaneous determination of plasma levels of tryptophan metabolites in depressive patients. Results: Plasma levels of TRP are not different between HC and patients of BDII. Serotonin (5-HT) levels are higher in BDII than HC. Plasma levels of 5-HIAA of HC are higher than those of old women of BDII, but lower in young women of BDII. Plasma levels of kynurenine (KYN) of HC are not different from those of patients of BDII. Conclusion: Plasma levels of 5-HT are higher in patients of BDII than those of HC, which may suggest that use of drugs inhibiting the 5-HT transportation and lower transporter biding may increase plasma levels of 5-HT in patients of BD.

Proposal of a new and simple staging system of colorectal liver metastasis

AIM： To create a new, simple and useful staging system for colorectal liver metastasis analogous to the Tumor Node Metastasis classification system of International Union Against Cancer. METHODS： A retrospective review was undertaken of 81 consecutive patients who underwent partial hepatectomy for colorectal liver metastases （group 1）. Clinical and pathological features of both primary and metastatic liver cancers were entered into a multivariate analysis to determine independent variables helpful in accurately predicting long-term prognosis after hepatectomy. Using selected variables, we created a new staging system like TNM classification. The usefulness of the new staging system was examined in a series of 92 patients from another hospital （group 2）. RESULTS： Multivariate analysis showed that 81 patients in group 1 had significant multiple hepatic tumors with the largest tumor being more than 5 cm in diameter, resectable extrahepatic distant metastases, and independent prognostic factors for poor survival after hepatectomy. Using these three variables, we created a new staging system to classify patients with colorectal liver metastases. Finally, our new staging system classified the patients both in group 1 and in group 2. CONCLUSION： Our new staging system of colorecta liver metastasis is simple and useful for staging patients.

Methods: Ultra high-speed liquid chromatography/mass spectrometry has been used for the simultaneous determination of plasma levels of tryptophan metabolites in depressive patients. Results: There are no significant differences between plasma levels of TRP between HC and MMD. Plasma levels of TRP of HC are higher in young men, young women, old men, and old women in this order. Serotonin (5-HT) levels are higher in MMD than HC. Plasma levels of 5-HIAA of HC are also higher than those of patients of MMD. Plasma levels of kynurenine (KYN) of healthy old men and old women are higher than those of young men and old women. Plasma levels of KYN are higher in old women and young men of MMD than those of HC. Conclusion: Plasma levels of 5-HT are higher in patients of MMD than those of HC, which may suggest that use of drugs inhibiting the 5-HT transportation may increase plasma levels of 5-HT in MMD.

System to Control Upright Posture with Alcoholic Intake

Equilibrium function in the cerebellum （vestibulo-cerebellar system） can deteriorate under the influence of alcohol. In the Romberg posture, the center of gravity, which was measured every 50 ms by stabilometry, appeared to shift with alcohol ingestion. In the previous study, a locus in the center of gravity （stabilogram） was converted to values of statistical indices such as area of sway, total locus length, sparse density, and locus length per unit area, although these indices could not always distinguish between the stabilograms sampled from seven healthy young males in sober and intoxicated states. This measurement was made with an AMTI force plate. In this study, ＂translation error＂ was estimated in a d-dimensional embedding space in order to compare stabilograms recorded before and after the ingestion of doubly diluted brandy in 30 s （1 ≤ d ≤ 10）. The authors succeeded in validating stochastic differential equations as a mathematical model of the body sway. Although the postural control system shows different patterns in the lateral and anterior-posterior directions, the randomness in the model was preserved after alcohol intake and significantly increased in the medial/lateral direction. Visual information referred by the postural conlrol system when standing might be interfered by the effects of intoxication, which was regarded as disturbance. The possibility of detecting alcohol-ingestion-induced reduction of the equilibrium function and its recovery process are also suggested in this paper. This method is considered to be useful to diagnose the disorders of the vestibulo-cerebellar system.

Deterioration of Dry Skin in Arthritis Model Mice via Stress-Induced Changes in Immune Cells in the Thymus and Spleen

Objective: Skin dryness is a characteristic of rheumatoid arthritis model mice. A previous study reported that the stress hormone glucocorticoid (i.e., corticosterone) is related to the induction of dry skin in arthritic mice. However, the mechanism through which stress induces dry skin in these mice is still unclear. Therefore, in this study, we examined the relationship between stress and induction of dry skin in arthritic mice. Methods: Physical stress load in mice with DBA/1JJmsSlc collagen-induced arthritis was treated with water immersion stress, and transepidermal water loss and the expression of markers associated with allergic reactions and inflammation was evaluated. Results: Deterioration of skin dryness was observed in stressed arthritic mice compared with that in unstressed arthritic mice. Moreover, plasma levels of interleukin-6 and corticosterone were increased in stressed arthritic mice compared with those in unstressed arthritic mice. We also observed decreased regulatory T cell numbers and increased T helper type 2 cell numbers in the thymus of stressed arthritic mice compared with those in unstressed arthritic mice. Conclusion: These results suggested that abnormalities in the immune system were related to deterioration of dry skin in stressed arthritic mice. Thus, reduction of stress may prevent deterioration of dry skin in mice with arthritis.
