Kinetic study of all-or-none hemolysis induced by cationic amphiphilic polymethacrylates with antimicrobial activity

To gain an understanding of the toxicity of antimicrobial polymers to human cells, their hemolytic action was investigated using human red blood cells （RBCs）. We examined the hemolysis induced by cationic amphiphilic methacrylate random copolymers, which have amino ethyl sidechains as cationic units and either butyl or methyl methacpjlate as hydrophobic units. The polymer with 30 tool% butyl sidechains （B3o） displayed higher hemolytic toxicity than the polymer with 59 tool% methyl sidechains （Ms9）. B3o also induced faster release of hemoglobin from RBCs than lVlsg. A new theoretical model is proposed based on two consecutive steps to form active polymer species on the RBC membranes, which are associated to RBC lysis. This model takes the all-or-none release of hemoglobin by the rupture of RBCs into account, providing new insight into the polymer-induced hemolysis regarding how individual or collective ceils respond to the polymers.

The aim of the present research was to establish a case study for the prediction of the unknown EDZ(Excavation Damaged Zone)distribution using a numerical analysis calibrated by replicating the trends in the EDZ observed from one of the representative underground research fields in Japan(Horonobe URL).In this study,a 2D numerical analysis using a damage model,which can determine rock deformation and fracturing simultaneously,is presented.It was calibrated to reproduce the excavation of the gallery at the Horonobe URL at a depth of 350 m.Simulated results show an excellent agreement with the extent of the measured EDZ and capture the failure modes of EDZ fractures suggested by the in-situ observations.Finally,the calibrated numerical analysis was used to realistically estimate the EDZ formation for the geological disposal of high-level radioactive waste(HLW)under the same environment as that of the above-mentioned galley at the Horonobe URL.Consequently,it was shown that the tensile/shear hybrid fractures dominantly constituted the EDZ and propagated to a maximum extent of about 0.3 m from the cavity wall during the cavity excavation for the HLW disposal.Overall,the calibrated numerical analysis and resulting estimations,targeted for the environment at the depth of 350 m at the Horonobe URL,where mudstone is located,should be useful for predicting the trends in the EDZ distribution expected in the implementation of HLW disposal projects under deep geological conditions,such as those that exist in Japan,which are dominated by sedimentary rocks,including mudstone。

Effect of permeants on fracture permeability in granite under hydrothermal conditions

Flow-through experiments were conducted with three permeants to determine the effect of pH,temperature,and cation concentrations on changes in permeability.Granite with a single fracture was used for each sample.Changes in the permeant concentrations due to pressure dissolution,free-face dissolution,and precipitation were identified by measuring the element concentrations before and after the experiments.In addition,the mineral transformation was analyzed by SEM-EDX.The results of the flow-through experiments showed a reduction in permeability in almost all the samples.This decrease in permeability may have been caused by the interaction between pressure dissolution and free-face dissolution,which occurred in the high pH water experiment,or between pressure dissolution and precipitation,which occurred in the saturated mineral water and simulated seawater experiments.When pressure dissolution and free-face dissolution occurred in the samples,the pH and temperature were seen to greatly affect the decrease in permeability,namely,the permeability decreased significantly with increasing pH and temperature.This remarkable decrease in permeability could have taken place because the dissolution rate constant of the mineral increased with the increasing pH and temperature.Moreover,when pressure dissolution and precipitation occurred in the samples,the cation concentrations and temperature were seen to greatly affect the changes in permeability,namely,the permeability decreased significantly with increasing cation concentrations and decreasing temperature.

DEAR EDITOR,Ancient DNA(a DNA) from mollusc shells is considered a potential archive of historical biodiversity and evolution.However, such information is currently lacking for mollusc shells from the deep ocean, especially those from acidic chemosynthetic environments theoretically unsuitable for longterm DNA preservation. Here, we report on the recovery of mitochondrial and nuclear gene markers by Illumina sequencing of a DNA from three shells of Archivesica nanshaensis – a hydrocarbon-seep vesicomyid clam previously known only from a pair of empty shells collected at a depth of 2626 m in the South China Sea.

Investigation on possible induction of adaptive response(AR)by high-liner energy transfer(LET)particle radiation for protection against low-LET photon radiation-induced detrimental effects has not yet been performed in utero.This study verified if an AR could be induced by high-LET particle radiation from accelerated heavy ions against low-LET X-ray radiation-induced detrimental effects on fetal mice.Total body irradiation of pregnant C57BL/6J mice were performed by delivering a priming dose ranging from 10 mGy to 320 mGy of particle radiation on gestation day 11 followed one day later by a challenge dose at 3500 mGy from X-ray radiation.The monoenergetic beams of carbon,silicon and iron with the LET values of about 15,55,and 200 KeV/μm,respectively,were examined.Significant suppression by the priming radiation of the detrimental effects(fetal death,malformation,or low body weight)was used as the endpoints for judgment of a successful AR induction on gestation day 18.Existence of AR was not observed.On the other hand,the priming dose of high-LET particle radiation,in some cases,even increased the detrimental effects induced by the challenge dose from low-LET X-ray radiation.Although existence of AR induced by high-LET radiation in cultured mammalian cells in vitro and in certain tissues of laboratory mice in vivo was demonstrated,the present study did not suggest that low dose of high-LET particle radiation could induce an AR in fetal mice in utero under the setup of our experimental system.

Lengthening periods of hospitalization, increasing numbers of patients with age-related complications, and a shortage of nursing staff have been of great concern in medical psychiatry in Japan. Under these circumstances, countries such as Japan that face a super-aging society and a decline in the working-age population, have been recommended for use of advanced information and communications technology (ICT) to improve the efficiency of medical treatment and care. This study aims to develop the “Monitoring Application for Inappropriate Expressions in Nursing Records” for using PsyNACS©, which will enable psychiatric nursing plans to harness the advantages of ICT. The functions considered necessary are as follows: 1) identification of users who enter information;2) a necessary database for lists of inappropriate expressions;3) development of a matching function to recommend proper writing input and a warning function;and 4) a management function for an inappropriate expression list database. A demonstration experiment for developing the application in this study was conducted at a specialized psychiatric hospital. To introduce them to the application, nurses and nurse managers were informed about the system developed in this study, and a survey regarding their opinions on the functions of the application was conducted with ten nurse managers. The results were evaluated in terms of usefulness for nursing care, documentation, and the education of nurses from an ethical perspective. This study suggests that matching their input with an inappropriate expression database will allow nurses to record more appropriate expressions. From an ethical perspective, the ability to use appropriate expressions makes records more likely to withstand disclosure requests from patients and their families.

Synchronous coexistence of liver metastases from cecal leiomyosarcoma and rectal adenocarcinoma: A case report

Multiple liver tumors represent a challenging condition for abdominal surgeons both in the selection of technique and the rarity of diagnosis. There are no case reports on co-existence of liver metastases from both intestinal leiomyosarcoma and adenocarcinoma. The patient described in this report successfully underwent resection of both primary lesions and liver metastases in combination with chemotherapy. As for the leiomyosarcoma, the primary cecal lesion was revealed more than three years after the patient's first visit. Peritoneal, lymph-node, and lung recurrences were observed afterward, and thus surgeries on those regions were performed. Pathologically, the peritoneal and lung recurrences comprised leiomyosarcoma and the lymphnode recurrence was diagnosed as adenocarcinoma. Despite newly discovered multiple lung recurrences and regional lymph-node metastases, the patient lived a normal life for 73 mo after the initial operation based on multidisciplinary therapy. He ultimately died of liver failure due to invasive lymph-node recurrence from the rectal adenocarcinoma, in addition to multiple lung recurrences from the leiomyosarcoma. Hepatic recurrence did not occur in this patient's case, which appears to be one reason for his long-term survival.

Explanatory Variables as Evidence for Precision in Intramuscular Long-Acting Injections of Patients with Schizophrenia

Intramuscular injections can cause damage to arteries, veins and nerves. Achieving maximum effects of medications through precise intramuscular (IM) injections must be assured and with certainty, particularly with long-acting injection drugs (LAI). The purpose of this study is to determine precision in IM LAIs of patients with schizophrenia. These evidences estimate “Distance from the Epidermis to the Under-Fascia” (DEUF) and “Distance from Epidermis to the Upper-Arm Bone” (DEB) of the deltoid muscle. Explanatory variables include body height, weight, body-mass index (BMI), deltoid-muscle circumference, fat percentage and muscle mass measured by body composition monitor. Sixty nine subjects are included based on the Diagnostic Statistical Manual of Mental Disorders (DSM-IV, 1994), and receive treatments of typical and atypical antipsychotic LAI. There are 46 males and 23 females with average age of 51.41 (Standard Deviation = 13.58) and ranging from 21 to 81 years who are all right-handed. Ultrasonographic data and those from explanatory variables are calculated using Spearman’s rank correlation coefficients. Multiple regression analysis (step-wise method/forced input method) is performed assuming DEUF or DEB as dependent variables. Significant correlations are found with highest adjusted R-square. Paired t-tests show differences in average values of actual ultrasonographic measurements assigned to DEUF or DEB, and the regression equations for accuracy. Ultrasonographic values are assigned multiple-regression equations as true values, and the calculated values are compared with those obtained by regression equations. There are no significant differences observed for either the right or left arm. The multiple regression equations for BMI and fat percentages (upper extremity estimation) of DEUF, and for BMI and injection site circumference of estimated DEB, successfully value the DEUF and DEB. In Japan, the shortage of personnel is a problem in long-term care nursing and rehabilitative care prevention. Nevertheless, Japan has taken measures to compensate for these shortages by promoting medical and nursing care activities using robotic technologies, and employing human resources from overseas. The purpose of this study was to determine potential legal issues and subsequent implications for care during prevention gymnastic exercises for the elderly using Pepper in long-term health facilities. The application program of Care-Prevention Gymnastics Exercises for Pepper (Pepper with CPGE) was made by the Xing Company Japan. Currently, care workers become intermediaries for the safe use of Pepper with CPGE. However, it was realized that some legal issues may arise if Pepper with CPGE alone will carry out these preventive care programs for the elderly without the presence of care workers as intermediaries. In this situation, using Pepper with CPGE alone to conduct care prevention gymnastics will require safety measures to prevent these possible practice issues and anticipate implications for care. In this regard, determining detailed target levels of rehabilitation exercise demands and environmental setting safety become essential factors. The use of humanoid robots in healthcare is expected to influence more practice protocols in contemporary and futurist rehabilitative human care. The identification of possible safety issues in performance and environmental situations, and implications for care are critical to ensure safe and valuable rehabilitative health care practices for the elderly population.
