cretaceous amber from Myanmar(～99 Ma Burmese amber) has become a valuable supplement to the traditional skeletal record of small theropod dinosaurs preserved in sedimentary rocks, particularly for coelurosaurs and enantiornithines. The specimens recovered from this deposit preserve skeletal material and soft tissues in unmatched detail. This provides opportunities to study three-dimensional preservation of soft tissues, microstructure, and pigmentation patterns that are seldom available elsewhere in the fossil record. Ultimately, this line of research provides insights into life stages that are difficult to preserve, the ecology and appearance of the groups involved, and the evolutionarydevelopment of structures such as feathers. Here we describe the most recent discovery from Burmese amber, an articulated skeleton of an enantiornithine bird. This individual has been sectioned along the coronal plane, providing a unique view inside multiple body regions. Osteological observations and plumage patterns support placement within the Enantiornithes, and suggest that the animal may have been a juvenile at the time of death. The specimen has a complex taphonomic history that includes exposure at the surface of a resin flow prior to encapsulation, and may include scavenging by some of the insects trapped within the same amber piece. The chemical composition observed along surface exposures and shallowly buried regions of the body indicate that the specimen has not undergone significant exchange with its surroundings. High iron concentrations are present in regions that preserve soft tissues as carbon films, and calcium distribution corresponds to regions where bones breach the surface of the amber.

Bisphenol A exposure alters release of immune and developmental modulators and expression of estrogen receptors in human fetal lung fibroblasts

Bisphenol A （BPA） has been shown to exert biological effects through estrogen receptor （ER）-dependent and ER-independent mechanisms. Recent studies suggest that prenatal exposure to BPA may increase the risk of childhood asthma. To investigate the underlying mechanisms in the actions of BPA, human fetal lung fibroblasts {hFLFs） were exposed to varying doses of BPA in culture for 24 hr. Effects of BPA on localization and uptake of BPA, cell viability, release of immune and developmental modulators, cellular localization and expression of ERa, ERβ and G-protein coupled estrogen receptor 30 （GPR30）, and effects of ERs antagonists on BPA-induced changes in endothelin-1 （ET-1） release were examined. BPA at 0.01-100 μmol/L caused no changes in cell viability after 24 hr of exposure, hFLFs expresses all three ERs. BPA had no effects on either cellular distribution or protein expression of ERa, however, at 100 μmol/L （or 23 μmol/L intracellular BPA） increased ERβ protein levels in the cytoplasmic fractions and GPR30 protein levels in the nuclear fractions. These paralleled with increased release of growth differentiation factor-15, decreased phosphorylation of nuclear factor kappa B p65 at serine 536, and decreased release of ET-1, interleukin-6, and interferon gamma-induced protein 10. ERs antagonists had no effects on BPA-induced decrease in ET-1 release. These data suggest that BPA at 100 μmol/L altered the release of immune and developmental modulators in hFLFs, which may negatively influence fetal lung development, maturation, and susceptibility to environmental stressors, although the role of BPA in childhood asthma remains to be confirmed in in viuo studies.

Targeting an oncogenic kinase/phosphatase signaling network for cancer therapy

Protein kinases and phosphatases signal by phosphorylation and dephosphorylation to precisely control the activities of their individual and common substrates for a coordinated cellular outcome. In many situations, a kinase/phosphatase complex signals dynamically in time and space through their reciprocal regulations and their cooperative actions on a substrate. This complex may be essential for malignant transformation and progression and can therefore be considered as a target for therapeutic intervention. p38γ is a unique MAPK family member that contains a PDZ motif at its c-terminus and interacts with a PDZ domain-containing protein tyrosine phosphatase PTPH1. This PDZcoupled binding is required for both PTPH1 dephosphorylation and inactivation of p38γ and for p38γ phosphorylation and activation of PTPH1. Moreover, the p38γ/PTPH1 complex can further regulate their substrates phosphorylation and dephosphorylation, which impacts Ras transformation, malignant growth and progression, and therapeutic response. This review will use the p38γ/PTPH1 signaling network as an example to discuss the potential of targeting the kinase/phosphatase signaling complex for development of novel targeted cancer therapy.

主要目标:比较两种剂量右美托咪啶在全身麻醉苏醒期:1.气管导管拔管期间的咳嗽反应。2.睁眼时间和气管导管拔管时间。3.拔管期间血流动力学反应次要目标:1.拔管30分钟后疼痛VAS评分,Ramsay镇静评分。***和谵妄等术后不良反应。3.术后24小时患者VAS疼痛评分和满意度评分。方法:选择(ASA)Ⅰ-Ⅱ级全麻下行择期腹腔镜胆囊切除术(Lc)患者(年龄:18-65岁)72例,随机分成A、B、c三组,每组24例。A组(对照组):舒芬太尼0.1μg/kg;B组:舒芬太尼0.1 μg/kg+右美托咪定0.25μg/kg;c组:舒芬太尼0.1μg/kg+右美托咪定0.5μg/kg。在胆囊切除后,各组药品用生理盐水稀释为10ml,泵注,速度1ml/***.。手术结束转送PAcU,记录并比较三组拔管前,即刻,拔管后30分钟内血压、心率;呛咳程度;睁眼时间;拔管时间;拔管后30分钟Ramsay镇静评分,VAS疼痛评分;以及术后喉痉挛,支气管痉挛,恶心呕吐等并发症;术后24 hr的VAS疼痛评分与围麻醉期舒适度评分。结果:与对照组A相比,B组和c组的咳嗽发生率明显降低,差异有统计学意义。(p=0.046,p=0.003)。睁眼和气管导管拔管时间 c 组(23.45±7.551,23.55±7.520)与 A 组和 B 组比较(p=0.004,p=0.004)差异有统计学意义,A组(16.73±4.300,16.73±4.300)和 B组(18.21±5.429)的睁眼和气管导管拔管时间相近,统计学差异不明显(p=0.398,p=0.334)。气管拔管期间A组HR和MAP明显高于c组差异有统计学意义(p=0.009,p=0.027)。与A组相比,c组的Ramsay镇静评分更高,差异有统计学意义(p=0.005)。术后VAS疼痛评分在拔管后30分钟,病房24小时,c组明显低于A组,差异有统计学意义(p=0.013,p=0.028)。术后24小时,B组和c组较对照组A满意度评分更高,差异有统计学意义(p=0.001,p=0.001)。结论:两种剂量右美托咪定0.25 mcg/kg,0.5 mcg/kg均能明显降低气管导管拔管期间的咳嗽发生率,提高患者的术后满意度。c组(0.5 mcg/kg)明显减轻气管导管拔管期间循环激惹,并具有更好的术后镇静和缓解疼痛。但明显延长了睁眼及气管导管的拔管时间。

Developmental and Feedforward control of the Expression of Folate Biosynthesis Genes in Tomato Fruit

Little is known about how plants regulate their folate content, including whether the expression of folate biosynthesis genes is orchestrated during development or modulated by folate levels. Nor is much known about how folate levels impact the expression of other genes. These points were addressed using wild-type tomato fruit and fruit engineered for high folate content. In wild-type fruit, the expression of genes specifying early steps in folate biosynthesis declined during development but that of other genes did not. In engineered fruit overexpressing foreign GTP cyclohy- drolase I and aminodeoxychorismate synthase genes, the expression of the respective endogenous genes did not change, but that of three downstream pathway genes--aminodeoxychorismate lyase, dihydroneopterin aldolase, and mitochondrial folylpolyglutamate synthase--respectively increased by up to 7.8-, 2.8-, and 1.7-fold, apparently in response to the build-up of specific folate pathway metabolites. These results indicate that, in fruit, certain folate pathway genes are developmentally regulated and that certain others are subject to feedforward control by pathway intermediates. Micro- array analysis showed that only 14 other transcripts （of 11 000 surveyed） increased in abundance by two-fold or more in high-folate fruit, demonstrating that the induction of folate pathway genes is relatively specific.

Ultrasound-Guided Greater Occipital Nerve Hydrodissection for Treatment of cervicogenic Headache: A case Report

We describe an innovative technique of ultrasound-guided greater occipital nerve (GON) hydrodissection for treatment of cervicogenic headache and occipital neuralgia. A 35-year-old female presented to the pain clinic with severe chronic cervicogenic headache impacting her sleep, work and activities of daily living. conservative management had failed to adequately resolve her pain. Ultrasound-guided suboccipital hydrodissection of the greater occipital nerve was performed with the patient in the prone position. After skin sterilization, the linear ultrasound transducer was oriented in a transverse orientation at the level of the c2-c3 vertebrae. The needle was advanced from medial to lateral “in-plane” under direct ultrasound visualization, until the needle was positioned at the c2 lamina. After confirming the needle tip position, 10 ml of hydrodissection fluid was injected with good visualization of distribution of the solution. The patient described immediate and significant improvement in her symptoms. She reported a sustained decrease in pain scores when followed up in the pain clinic at six and twelve weeks respectively. To the best of our knowledge this is the first application of ultrasound-guided hydrodissection of the GON for cervicogenic headache. It offers a novel, safe and effective technique to aid in the diagnosis and treatment of a common pain condition.

Simulating gamma-ray production from cosmic rays interacting with the solar atmosphere in the presence of coronal magnetic fields

cosmic rays can interact with the solar atmosphere and produce a slew of secondary messengers,making the Sun a bright gamma-ray source in the *** observations with Fermi-LAT have shown that these interactions must be strongly affected by solar magnetic fields in order to produce a wide range of observational features,such as a high flux and hard ***,the detailed mechanisms behind these features are still a *** this study,we tackle this problem by performing particle-interaction simulations in the solar atmosphere in the presence of coronal magnetic fields using the potential field source surface(PFSS)*** find that low-energy(~GeV)gamma-ray production is significantly enhanced by the coronal magnetic fields,but the enhancement decreases rapidly with *** enhancement directly correlates with the production of gamma rays with large deviation angles relative to the input cosmic-ray *** conclude that coronal magnetic fields are essential for correctly modeling solar disk gamma rays below 10 GeV,but above that,the effect of coronal magnetic fields *** magnetic field structures are needed to explain the high-energy disk emission.

Vasoactive intestinal polypeptide (VIP) corrects chronic inflammatory response syndrome (cIRS) acquired following exposure to water-damaged buildings

Exposure in water-damaged buildings (WDB) to airborne bioaerosols including metabolic products of toxigenic fungi, bacteria and actinomycetes;and inflammagens, can lead to a persistent innate immune inflammatory illness. This illness, termed a chronic inflammatory response syndrome (cIRS-WDB), is systemic with symptoms acquired from multiple organ systems. Treatment of cIRS-WDB has progressed rapidly as a better understanding of the inflammatory pathophysiology has led to targeted, sequential therapies. The fundamental basis of uncontrolled innate immune responses, the humoral deficiency of regulatory neuropeptides melanocyte stimulating hormone (MSH) or vasoactive intestinal polypeptide (VIP), seen in over 98% of pa tients, has not consistently responded to any treatment modality. Use of replacement VIP has been attempted anecdotally;VIP replacement therapies show promise in short term studies but longer therapies have not been attempted. Here we report an open label trial of 20 patients with refractory cIRS-WDB illness who took replacement VIP in a nasal spray for at least 18 months with confirmation of durable efficacy and absence of significant side effects. These 20 patients were similar in symptoms and lab find- ings to three previously published cohorts in- volving 1829 patients and 169 controls. Dosage of VIP was titrated downwards from four to zero doses a day to determine minimum effective dose, and retitrated upwards for maximum improvement over time. The trial showed that VIP therapy safely 1) reduced refractory symptoms to equal controls;2) corrected inflammatory parameters c4a, TGF beta-1, VEGF, MMP9;3) corrected estradiol, testosterone and 25-OH Vitamin D;4) returned pulmonary artery systolic pressure (PASP) during exercise to normal;and 5) enhanced quality of life in 100% of trial patients. Subsequent identification of correction of T-regulatory cell levels supports the potential role of VIP in both innate and adaptive immune function.

Background:The vocalizations of birds are dynamic traits that often vary in output with time of day and time of *** quantifying patterns of diel and seasonal variation in vocal output,we can gain insight into the ecology and evolution of birds and the function of their *** this investigation,we quantified diel and seasonal variation in song output by studying a breeding population of Savannah Sparrows (Passerculus sandwichensis).Methods:We used autonomous recorders to collect extensive recordings across the breeding season in a long-term, colour-marked study population of Savannah Sparrows in eastern *** described diel and seasonal variation across five different breeding stages based on recordings of more than 50,000 songs from 34 ***:During the pre-breeding stage, prior to female arrival, males sang a pronounced dawn chorus with a peak in song output during the early *** the breeding stage,in contrast,the dawn chorus was diminished and males instead exhibited a pronounced dusk chorus with a peak in song output during the *** the breeding season,the highest levels of song output occurred in late April and early May,soon after males arrived on the breeding grounds,and the lowest levels occurred in August,prior to the departure of birds for the wintering ***:These patterns suggest that Savannah Sparrows' early-season dawn choruses are directed at males and serve a territorial defense ***-season dusk choruses,in contrast, appear to play a role in attracting mates and possibly acoustic mate guarding.
