Comparative Experimental and Numerical study of Wave Loads on A Monopile structure Using Different Turbulence Models

This study numerically and experimentally investigates the effects of wave loads on a monopile-type offshore wind turbine placed on a 1:25 slope at different water depths as well as the effect of choosing different turbulence models on the efficiency of the numerical *** numerical model adopts a two-phase flow by solving Unsteady Reynolds-Averaged Navier−stokes(URANs)equations using the Volume Of Fluid(VOF)method and three differentk-􀀀ωturbulence *** environmental conditions from the East China sea are *** wave run-up and the wave loads applied on the monopile are investigated and compared with relevant experimental data as well as with mathematical predictions based on relevant *** numerical model is well validated against the experimental data at model *** use of different turbulence models results in different predictions on the wave height but less differences on the wave *** baseline k-􀀀ωturbulence model and shear-stress Transport(ssT)k-􀀀ωturbulence model exhibit better performance on the prediction of hydrodynamic load,at a model-scale water depth of 0.42 m,while the laminar model provides better results for large water *** ssT turbulence model performs better in predicting wave run-up for water depth 0.42 m,while the laminar model and standard k-􀀀ωmodel perform better at water depth 0.52 m and 0.62 m,respectively.

Normalized urban heat island (UHI) indicators: Classifying the temporal variation of UHI for building energy simulation (BEs) applications

Despite known effects of urban heat island(UHI)on building energy consumption such as increased cooling energy demand,typical building energy simulation(BEs)practices lack a standardized approach to incorporate UHI into building energy *** seasonal and diurnal variation of UHI makes the task of incorporating UHI into BEs an especially challenging task,often limited by the availability of detailed hourly temperature data at building *** paper addresses the temporal variation of UHI by deriving four normalized UHI indicators that can successfully capture the seasonal and diurnal variation of *** accuracy of these indicators was established across four climate types including hot and humid(Miami,FL),hot and dry(Los Angeles,CA),cold and dry(Denver,CO),and cold and humid(Chicago,IL),and three building types including office,hospital,and *** four indicators are mean summer daytime UHI,mean summer nighttime UHI,mean winter daytime UHI,and mean winter nighttime UHI,which can accurately predict cooling,heating,and annual energy consumption with mean relative error of less than 1%.Not only do these indicators simplify the application of UHI to BEs but also,they provide a new paradigm for UHI data collection,storage,and usage,specifically for the purpose of BEs.

Molecular approaches to study the insect gut symbiotic microbiota at the ＇omics＇ age

Insect gut symbiotic microbiota play essential roles in the growth, development, pathogenesis and environmental adaptation of host insects. The molecular and systems level analysis of insect gut symbiotic microbial community will allow us to discover novel biocatalysts for biomass deconstruction and to develop innovative strategies for pest management. We hereby review the various molecular biology techniques as applied to insect gut symbiont analysis. This review aims to serve as an informative resource for experimental design and research strategy development in the field. We first discuss various strategies for sample preparation and their pros and cons. The traditional molecular techniques like DGGE, RFLP and FIsH are covered with respect to how they are applied to study the composition, diversity and dynamics of insect gut symbiotic microbiota. We then focus on the various ＇ omics＇ techniques. The metagenome analysis together with the recent advancements in next-generation sequencing will provide enormous sequencing information, allowing in-depth microbial diversity analysis and modeling of pathways for biological processes such as biomass degradation. The metagenome sequencing will also enable the study of system dynamics and gene expression with metatranscriptome and metaproteome methods. The integration of different ＇omics＇ level data will allow us to understand how insect gut works as a system to carry out its functions. The molecular and systems-level understanding will also guide the reverse design of next-generation biorefinery.

Prostatic diseases such as prostate cancer and benign prostatic hyperplasia are highly prevalent among men. The number of studies focused on the abundance and roles of intrinsically disordered proteins in prostate cancer is rather limited. The goal of this study is to analyze the prevalence and degree of disorder in proteins that were previously associated with the prostate cancer pathogenesis and to compare these proteins to the entire human proteome. The analysis of these datasets provides means for drawing conclusions on the roles of disordered proteins in this common male disease. We also hope that the results of our analysis can potentially lead to future experimental studies of these proteins to find novel pathways associated with this disease.

Multiple small bowel ruptures due to ischemic enteritis: A case report

A rare case of multiple small bowel ruptures due to ischemic enteritis （IsE） is reported. The patient was admitted to the hospital with acute abdominal pain followed by bloody diarrhoeas. Preoperative colonoscopic findings were similar to those presented in Crohn＇s disease. Intraoperatively, ischemic lesions and multiple ruptures were localized at the jejunum and the proximal ileum. Histopathological examination of the resected bowel segment established the diagnosis of IsE. Although IsE is not common, concurred multiple ruptures of the small bowel is a rare but actual complication.

Identification of stable normalization genes for quantitative real-time PCR in porcine articular cartilage

Background： Expression levels for genes of interest must be normalized with an appropriate reference, or housekeeping gene, to make accurate comparisons of quantitative real-time PCR results. The purpose of this study was to identify the most stable housekeeping genes in porcine articular cartilage subjected to a mechanical injury from a panel of 10 candidate genes. Results： Ten candidate housekeeping genes were evaluated in three different treatment groups of mechanically impacted porcine articular cartilage. The genes evaluated were： beta actin, beta-2-microglobulin, glyceraldehyde-3-phosphate dehydrogenase, hydroxymethylbilane synthase, hypoxanthine phosphoribosyl transferase, peptidylprolyl isomerase A （cyclophilin A）, ribosomal protein L4, succinate dehydrogenase flavoprotein subunit A, TATA box binding protein, and tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein--zeta polypeptide The stability of the genes was measured using geNorm, BestKeeper, and NormFinder software. The four most stable genes measured via geNorm were （most to least stable） succinate dehydrogenase flavoprotein, subunit A, peptidylprolyl isomerase A, glyceraldehyde-3-phosphate dehydrogenase, beta actin; the four most stable genes measured via BestKeeper were glyceraldehyde-3-phosphate dehydrogenase, peptidylprolyl isomerase A, beta actin, succinate dehydrogenase flavoprotein, subunit A; and the four most stable genes measured via NormFinder were peptidylprolyl isomerase A, sucdnate dehydrogenase flavoprotein, subunit A, glyceraldehyde-3-phosphate dehydrogenase, beta actin. Conclusions： BestKeeper, geNorm, and NormFinder all generated similar results for the most stable genes in porcine articular cartilage. The use of these appropriate reference genes will facilitate accurate gene expression studies of porcine articular cartilage and suggest appropriate housekeeping genes for articular cartilage studies in other species.
