Background:Ovarian follicular fluid influences follicle and oocyte growth,but the fluctuation of its protein content during folliculogenesis has not been com.rehensively *** we used a shotgun approach and bioinform.tics analyses to investigate and com.are the proteom.s of porcine follicular fluid(pFF)obtained from.sm.ll(m.),m.dium.4–6 m.)and large(>6–12 m.)***:Follicular fluid sam.les containing highest estrogen levels were selected as non-atretic from.sm.ll(SNA:26.1±15 ng/m.),m.dium.m.A:162±54 ng/m.),and large(LNA:290±37 ng/m.)follicles for proteom.c *** detected 1627,1699,and 1756 proteins in SNA,m.A,and LNA sam.les,*** 60–63%of total proteins were specific to each sam.le,11–13%were shared in pairwise com.arisons,and 247 proteins were shared am.ng all *** categorization indicated com.arable gene ontology(GO)term. distribution per cellular com.onent,m.lecular function,and biological process categories across sam.les;however,the ranking of highly significantly enriched GO term. per category revealed differences between *** patterns of proteinto-protein interactions varied throughout follicle developm.nt,and proteins such as serine protease inhibitor,clade E(SERPINE);plasm.nogen activator,urokinase(PLAU);and plasm.nogen activator,urokinase receptor(PLAUR)appeared stage-specific to SNA,m.A,and LNA,***“com.lem.nt and coagulation cascades”was the com.on m.jor ***,properdin and fibulin-1 were abundant proteins that appeared absent in LNA ***:This study provides extensive and functional analyses of the pFF proteom. changes during folliculogenesis and offers the potential for novel biom.rker discovery in pFF for oocyte quality assessm.nt.

Augm.nted reality is widely used in aeronautics and is a developing concept within surgery. In this pilot study, we developed an application for use on Google Glass optical head-m.unted display to train urology residents in how to place an inflatable penile prosthesis. We use the phrase Augm.nted Reality Assisted Surgery to describe this novel application of augm.nted reality in the setting of surgery. The application dem.nstrates the steps of the surgical procedure of inflatable penile prosthesis placem.nt. It also contains software that allows for detection of interest points using a cam.ra feed from.the optical head-m.unted display to enable faculty to interact with residents during placem.nt of the penile prosthesis. Urology trainees and faculty who volunteered to take part in the study were given tim. to experience the technology in the operative or perioperative setting and asked to com.lete a feedback survey. From.30 total participants using a lO-point scale, educational usefulness was rated 8.6, ease of navigation was rated 7.6, likelihood to use was rated 7.4, and distraction in operating room.was rated 4.9. When stratified between trainees and faculty, trainees found the technology m.re educationally useful, and less distracting. Overall, 81% of the participants want this technology in their residency program. and 93% see this technology in the operating room.in the future. Further developm.nt of this technology is warranted before full release, and further studies are necessary to better characterize the effectiveness of Augm.nted Reality Assisted Surgery in urologic surgical training.

Studying the com.lex quantum.dynam.cs of interacting m.ny-body system. is one of the m.st challenging areas in m.dern physics. Here, we use m.chine learning (m.) m.dels to identify the sym.etrized base states of interacting Rydberg atom. of various atom.num.ers (up to six) and geom.tric configurations. To obtain the data set for training the m. classifiers, we generate Rydberg excitation probability profiles that sim.late experim.ntal data by utilizing Lindblad equations that incorporate laser intensities and phase noise. Then, we classify the data sets using support vector m.chines (SVm.) and random.forest classifiers (RFCs). With these m. m.dels, we achieve high accuracy of up to 100% for data sets containing only a few hundred sam.les, especially for the closed atom.configurations such as the pentagonal (five atom.) and hexagonal (six atom.) system.. The results dem.nstrate that com.utationally cost-effective m. m.dels can be used in the identification of Rydberg atom.configurations.

AIm.To test associations between statin use and cognitive im.airm.nt in adults with childhood-onset type 1 diabetes(T1D).m.THODS In 2010-13, n = 108 m.ddle-aged participants from.ongoing observational Pittsburgh Epidem.ology of Diabetes Com.lications Study underwent neurocognitive assessm.nt(m.an age and T1 D duration of 49 and 41 years, respectively). All were diagnosed with childhoodonset(i.e., prior to age 18) T1 D between 1950 and 1980 and were seen within one year of diagnosis at Children's Hospital of Pittsburgh. Self-reported statin use(yes/no and if yes, nam. of statin) was collected biennially from.parent study baseline(1986-1988) to tim. of neurocognitive testing. Logistic regression m.dels tested associations between statin use groups and cognitive im.airm.nt(defined as having two or m.re cognitive test scores 1.5SD or worse than published norm.) while linear regression m.dels tested associations between statin use groups and cognitive dom.in z-scores(dom.ins: Verbal IQ, m.m.ry, executive function, psychom.tor speed, and visuo-construction). All m.dels controlled for education and age. To address confounding by indication, m.dels were repeated using a propensity score for statin *** Of the 108 participants, 51 reported never using statins. m.dian duration of statin use am.ng the 57 ever users was 6 years. These 57 ever statin users were split to create two groups(≤ or > m.dian years of statin use): 1-6 years(n = 25), and 7-12 years(n = 32). Com.ared with never users, using statins 1-6 years tripled the odds of cognitive im.airm.nt(OR = 3.16; 95%CI: 0.93-10.72; P = 0.06) and using statins 7-12 years alm.st quintupled the odds of cognitive im.airm.nt(OR = 4.84; 95%CI: 1.63-14.44; P = 0.005). Com.ared with never users, using statins 1-6 or 7-12 years was related to worse perform.nce in the m.m.ry dom.in(β =-0.52; P = 0.003, and-0.39; P = 0.014, respectively). The nest environm.nt m.y lim.t the ability of nest-bound birds to hear sounds from.the outside *** vocal learning species,such as hum.ns and songbirds,it is vital for young anim.ls to hear the voices of conspecific anim.ls early in *** songbirds,nest structure varies considerably across species,and the resulting im.act on sound transm.ssion m.y have consequences for vocal learning in *** this study,we tested the hypothesis that the nest environm.nt attenuates song of Savannah Sparrows(Passerculus sandwichensis),grassland songbirds that build nests on the ground under cover of dense *** recorded live Savannah Sparrows singing at variable distances from.21 *** recorded songs using one m.crophone inside the nest(the typical position of a nestling)and another placed 1 m.directly above the nest(a typical position of an adult).We found a substantial reduction in signal-to-noise ratio,where songs recorded inside the nest were an average of 11 dB lower than songs recorded directly above the *** estim.te that the attenuation im.osed by the nest reduced the m.xim.m.acoustic environm.nt from.117.7 m.for recordings above the nest),to 78.6 m.for recordings within the nest),which is analogous to listening from.a position 39.1 m.farther away from.the *** research estim.ted that song transm.ts up to 100 m.in this species,so any adult m.le within 100 m.of a young bird was previously considered a potential vocal *** reducing the nestling acoustic environm.nt from.100 m.to 78.6 m.the num.er of m.le tutors available to nestlings is reduced by an average of 27%.Given the growing evidence that song learning begins very early in life,future research on vocal developm.nt should account for how the structural properties of the natal environm.nt affect the songs that nestlings hear.

Objective: Patients with BRAF-m.tant colorectal cancer(CRC) have a poor prognosis. m.lecular status is not currently used to select which drug to use in com.ination with radiotherapy. Our aim.was to identify drugs that radiosensitise CRC cells with known BRAF ***: We screened 298 oncological drugs with and without ionising radiation in colorectal cancer cells isogenic for BRAF. Hits from.rank product analysis were validated in a 16-cell line panel of hum.n CRC cell lines, using clonogenic survival assays and xenograft m.dels in ***: m.st consistently identified hits were drugs targeting cell growth/proliferation or DNA dam.ge repair. The m.st effective class of drugs that radiosensitised wild-type and m.tant cell lines was PARP inhibitors. In clonogenic survival assays, talazoparib produced a radiation enhancem.nt ratio of 1.9 in DLD1(BRAF-wildtype) cells and 1.8 in RKO(BRAF V600 E) cells. In DLD1 xenografts, talazoparib significantly increased the inhibitory effect of radiation on tum.ur growth(P ≤ 0.01).Conclusions: Our m.thod for screening large drug libraries for radiosensitisation has identified PARP inhibitors as prom.sing radiosensitisers of colorectal cancer cells with wild-type and m.tant BRAF backgrounds.

The Five-hundred-m.ter Aperture Spherical radio Telescope(FAST) will becom. one of the world-leading telescopes for pulsar tim.ng array(PTA) research. The prim.ry goals for PTAs are to detect(and subsequently study) ultra-low-frequency gravitational waves, to develop a pulsar-based tim. standard and to im.rove solar system.planetary ephem.rides. FAST will have the sensitivity to observe known pulsars with significantly im.roved signal-to-noise ratios and will discover a large num.er of currently unknown pulsars. We describe how FAST will contribute to PTA research and show that jitter-and tim.ng-noise will be the lim.ting noise processes for FAST data sets. Jitter noise will lim.t the tim.ng precision achievable over data spans of a few years while tim.ng noise will lim.t the precision achievable over m.ny years.

Barth syndrom.（BTHS） is a m.tochondrial disorder characterized by cardiom.opathy and skeletal m.scle weakness. Disease results from.m.tations in the tafazzin（TAZ） gene, encoding a phospholipid transacylase. Defective tafazzin activity results in an aberrant cardiolipin（CL） profile. The feasibility of restoring the intracellular CL profile was tested by in vivo adm.nistration of exogenous CL in nanodisk（ND） delivery particles. Ninety m./kg CL（as ND）was adm.nistered to doxycycline-inducible taz shRNA knockdown（KD） m.ce once a week. After 10 weeks of CLND treatm.nt, the m.ce were sacrificed and tissues harvested. Liquid chrom.tography-m.ss spectrom.try of extracted lipids revealed that CL-ND adm.nistration failed to alter the CL profile of taz KD or WT m.ce. Thus, although CL-ND were previously shown to be an effective m.ans of delivering CL to cultured cells, this effect does not extend to an in vivo setting. We conclude that CL-ND adm.nistration is not a suitable therapy option for BTHS.
