Cloning and Characterization of a Putative CTR1 Gene from Wheat

CTR1 is a key negative regulator in ethylene signal transduction. A salt-induced CTR1 like gene （TaCTR1） was cloned from wheat, its expression under abiotic stresses, subcellular localization and the effect of overexpression of TaCTR1 on salt tolerance in tobacco was studied. A putative CTR1 gene was cloned and characterized from wheat via rapid amplification of cDNA ends （RACE） and RT-PCR. TaCTR1 expression under stresses was analyzed using semi-quantitative RT-PCR and the effect of overexpression of TaCTR1 on salt tolerance was conducted in tobacco. The full-length cDNA of TaCTR1 is 2 635 bp which codes for a polypeptide of 759 amino acids. There is a conserved serine/threonine protein kinase domain at the carboxyl terminus containing an ATP-binding site. Southern blot analysis revealed that TaCTR1 consisted of a gene family in wheat. The amino acid homologies of CTR1 among different organisms share higher similarities. Expression analysis revealed that TaCTR1 was induced by NaC1 and drought stress but inhibited by ABA treatment. Transient expression of TaCTR1-GFP in the onion epidermal cells indicated that TaCTR1 was probably targeted to the plasma membrane. Overexpression of TaCTR1 decreased salt tolerance in transgenic tobacco （Nicotiana tabacum L.） plants compared with the control. To our knowledge, TaCTR1 is the first CTR1 gene cloned in wheat and may be involved in various abiotic stresses. Overexpression of TaCTR1 decreased the salt tolerance in tobacco suggested that TaCTR1 may act as a negative regulator of salt stress in plants.

Four-Channel 1.55μm DFB Laser Array Monolithically Integrated with a 4× 1 Multimode-Interference Combiner Based on Nanoimprint Lithography

A monolithically integrated four-channel λ/4 phase shift distributed feedback （DFB） laser array with a 4 × 1 multimode interference combiner is designed and fabricated using nanoimprint lithography. The threshold currents of the lasers are less than 10mA, which are as good as a discrete DFB laser. Moreover, the side-mode suppression ratio is also better than 50 dB. The channel space is about 200 GHz for dense wavelength division multiplexing applications in an 1.55μm system.

The directional structure of Ni60/high-aluminum bronze composite coating was formed using induction remelting and forced *** microstructural evolution and the characteristics of interface growth were *** results showed that the remelted coating formed metallurgical bonding with the *** micros tructures changed from plane crystal to dendrite,cellular dendrite,fine cellular dendrite,and then to dendrite again with the increase in the cooling *** crystal grew along the heat flow direction and had(111) and(200)preferred orientations when the cooling rate was 1.886 ml-min^(-1).mm^(-2).The plane crystal,dendrite and cellular dendrite were mainly composed of compounds and solid solutions with Ni,Fe and Cu,and they were surrounded by strengthening phases composed of Cr,C and *** grain boundary of directional structure coatings showed the characteristic of regular eutectic growth,but grain boundary of remelted coating presented characteristic of divorced eutectic *** wear resistance of directional structure coatings is better than that of remelted coating.

The blasting experiment of the cement mortar model reveals that joint inclination affects blasted rock size distribution, which is an important index to evaluate the blasting effect. According to the fractal theory, the relationship is built up between fractal dimension and mass fraction of blasted fragments screened, that is, y x=100(x/x m ) 3-D . On the basis of the experiment results, the fractal dimension describing fragment size distribution is calculated.

Genetic alterations in hepatocellular carcinoma: An update

Hepatocellular carcinoma(HCC) is one of the leading causes of cancer-related deaths worldwide. Although recent advances in therapeutic approaches for treating HCC have improved the prognoses of patients with HCC, this cancer is still associated with a poor survival rate mainly due to late diagnosis. Therefore, a diagnosis must be made sufficiently early to perform curative and effective treatments. There is a need for a deeper understanding of the molecular mechanisms underlying the initiation and progression of HCC because these mechanisms are critical for making early diagnoses and developing novel therapeutic strategies. Over the past decade, much progress has been made in elucidating the molecular mechanisms underlying hepatocarcinogenesis. In particular, recent advances in next-generation sequencing technologies have revealed numerous genetic alterations, including recurrently mutated genes and dysregulated signaling pathways in HCC. A better understanding of the genetic alterations in HCC could contribute to identifying potential driver mutations and discovering novel therapeutic targets in the future. In this article, we summarize the current advances in research on the genetic alterations, including genomic instability, single-nucleotide polymorphisms, somatic mutations and deregulated signaling pathways, implicated in the initiation and progression of HCC. We also attempt to elucidate some of the genetic mechanisms that contribute to making early diagnoses of and developing molecularly targeted therapies for HCC.

Silver dressing in the management of an infant's urachal anomaly infected with methicillin-resistant Staphylococcus aureus:A case report

BACKGROUND Symptomatic urachal anomalies are rare *** management of urachal remnants has historically been surgical excision because of the connection between urachal remnants and risk of malignancy development later in ***,recent literature suggests that urachal anomalies that do not extend to the bladder can be treated with conservative *** this case,we report a newborn with an infected urachal remnant who was treated with a combination of antibiotics and a silver-based dressing and finally recovered *** SUMMARY Female baby A,weighing 2.88 kg at 38^(+5) wk of gestational age,was referred to the hospital because of a red,swollen umbilicus approximately 2 cm×2 cm in size with yellow purulent *** physical and ultrasound examination,the baby was finally diagnosed with a urachal *** first used oxacillin to prevent infection for 3 *** the 4^(th) day,microbiology testing of the umbilical exudate revealed the presence of methicillin-resistant Staphylococcus aureus(MRSA).We changed the treatment with oxacillin to vancomycin for systemic infection and treated the umbilical inflammation with a silver sulfate *** 5 d,the symptoms of the umbilicus disappeared,and we discontinued silver dressing *** the 12^(th) day,umbilical exudate testing was negative for *** the 14^(th) day,the baby's blood testing showed a white blood cell count of 14.7×10^(9)/L,neutrophil percentage of 27.8%,and C-reactive protein level of 1.0 mg/L,suggesting that the infection had been *** stopped treatment,and the baby was discharged with no *** this case,the infected urachal anomaly was cured with silver dressing and antibiotic application instead of surgical methods,which was a different course from that of some other urachal remnant *** Anomalies that do not connect with the bladder can be treated with nonoperative management,including application of conservative antibiotics and local i

Long non-coding RNAs in hepatocellular carcinoma: Potential roles and clinical implications

Long non-coding RNAs(lnc RNAs) are a subgroup of non-coding RNA transcripts greater than 200 nucleotides in length with little or no protein-coding potential. Emerging evidence indicates that lnc RNAs may play important regulatory roles in the pathogenesis and progression of human cancers, including hepatocellular carcinoma(HCC). Certain lnc RNAs may be used as diagnostic or prognostic markers for HCC, a serious malignancy with increasing morbidity and high mortality rates worldwide. Therefore, elucidating the functional roles of lnc RNAs in tumors can contribute to a better understanding of the molecular mechanisms of HCC and may help in developing novel therapeutic targets. In this review, we summarize the recent progress regarding the functional roles of lnc RNAs in HCC and explore their clinical implications as diagnostic or prognostic biomarkers and molecular therapeutic targets for HCC.

For better performances of Ni-based catalysts at low temperatures,Ni/SiC catalyst doped with a little amount of additive La was successfully *** catalytic CO methanation activity tests showed that 3%La-Ni/SiC catalyst was excellent at a low reaction temperature(95.9%CO conversion and 85.1%CH4 selectivity at250℃)with a superior stability compared with Ni/SiC(3.4%CO conversion and 0%CH4 selectivity at 250℃).This can be attributed to that the addition of La can markedly improve the dispersibility of active metal Ni and reduce the particle sizes of Ni nanoparticles or clusters,and can also regulate the interaction between active components and ***,the high thermal conductivity and thermal stability could avoid the generation of hot spots in the catalyst *** results will promote the development of highly active Ni-based catalysts for the low-temperature methanation reaction.

血栓形成是增加新冠肺炎患者死亡率和导致长新冠后遗症的关键因素。由活血化瘀中药丹参、川芎组成的冠心宁片(GXNT)具有显著的抗血栓活性,但其抗常规血栓与由新冠病毒引起微血栓机制之间的异同仍不清楚。本文运用体外抗血小板实验、网络药理学分析、分子对接技术和分子生物学实验,初步揭示了GXNT治疗血栓及新冠病毒引起的微血栓的主要活性成分、潜在靶点和作用机制。体外血小板聚集和黏附实验结果表明,GXNT具有显著的抗血小板聚集和黏附的活性,且呈一定的剂量依赖性。网络药理学分析发现,GXNT中的丹酚酸B、丹参酮IA、咖啡酸和川芎嗪能够通过关键靶点高迁移率族蛋白B1(highmobilitygroupbox1protein,HMGB1)、肿瘤坏死因子(tumor necrosis factor,TNF)、白细胞介素6(interleukin-6,IL6)和丝氨酸/苏氨酸激酶1(AKT serine/threonine kinase1,AKT1)来抗血栓及由新冠病毒引起的微血栓。其中,HMGB1信号通路为其关键的共同机制之一。Western blot实验结果也表明,GXNT对血小板中HMGB1蛋白的表达水平有显著抑制作用。综上,本文探究的GXNT抗常规血栓与由新冠病毒引起微血栓机制之间的异同为临床防治长新冠后遗症提供用药参考和理论依据。该动物实验已通过天津中医药大学实验动物伦理委员会审核批准(编号:TCM-LAEC2023187g1549)。
