Dear Editor,Severe fever with thrombocytopenia syndrome virus(SFTSV),an emerging pathogen,is a tick-borne bunyavirus belonging to the genus Bandavirus in the family Phenuiviridae(Kuhn et al.,2020).This pathogen was first identified in China during the heightened surveillance of acute febrile illness in 2009,and has been reported to cause several outbreaks in eastern Asia areas,including China,Japan,and Korea(Yu et al.,2011).

Monkeypox was declared a global health emergency by the World Health Organization,and as of March 2023,86,000 confirmed cases and 111 deaths across 110 countries have been reported.Its causal agent,monkeypox virus(MPV)belongs to a large family of double-stranded DNA viruses,Orthopoxviridae,that also includes vaccinia virus(VACV)and others.MPV produces two distinct forms of viral particles during its replication cycles:the enveloped viron(EV)that is released via exocytosis,and the mature viron(MV)that is discharged through lysis of host cells.This study was designed to develop multi-valent m RNA vaccines against monkeypox EV and MV surface proteins,and examine their efficacy and mechanism of action.Four m RNA vaccines were produced with different combinations of surface proteins from EV(A35R and B6R),MV(A29L,E8L,H3L and M1R),or EV and MV,and were administered in Balb/c mice to assess their immunogenicity potentials.A dynamic immune response was observed as soon as seven days after initial immunization,while a strong Ig G response to all immunogens was detected with ELISA after two vaccinations.The higher number of immunogens contributed to a more robust total Ig G response and correlating neutralizing activity against VACV,indicating the additive potential of each immunogen in generating immune response and nullifying VACV infection.Further,the m RNA vaccines elicited an antigen-specific CD4^(+)T cell response that is biased towards Th1.The m RNA vaccines with different combinations of EVand MV surface antigens protected a mouse model from a lethal dose VACV challenge,with the EV and MV antigens-combined vaccine offering the strongest protection.These findings provide insight into the protective mechanism of multi-valent m RNAvaccines against MPV,and also the foundation for further development of effective and safe m RNA vaccines for enhanced protection against monkeypox virus outbreak.

Dear Editor,The early diagnosis and prognosis of COVID-19 remain major challenges.At present,there are few studies focusing on the changes of sera proteome betAA/een active and recovered COVID-19 patients,and almost no biomarkers in sera are used to predict the function disorder of the injured tissues and organs in COVID-19 patients.In this study,we applied sera proteomics in the active and recovered COVID-19 patients,as well as healthy volunteers,combining with pathological staining of the patients'tissues to identify the potential prognosis biomarkers that can reflect specific organ damage.

Dear Editor,Almost one-half of COVID-19 patients have symptoms of liver injury.However,the molecular characteristics in the COVID-19 liver remain unknown.Therefore,the need for long-term medication for recovery make it imperative to study the infectivity and pathogenicity of SARS-CoV-2 in the liver.Here,we have identifed signifcant proteomic changes for the first time in the liver of COVID-19 patients.We detected differential expression levels of several proteins that mediate characteristic cellular host responses elicited by SARS-CoV-2 infection in the liver.

Although targeted therapy has been extensively investigated for breast cancers,a molecular target with broad application is currently unavailable due to the high heterogeneity of these cancers.Mammaglobin-A(Mam-A),which is overexpressed in most breast carcinomas,has been proposed as a promising target.However,the lack of specific targeting moieties due to uncertain binding epitopes hampers further translational study.Here,seven potential epitopes of Mam-A were disclosed,and a unique epitope was then identified in most types of breast cancers,despite the genotypic heterogeneity.With phage display technology,the epitope was determined to be N-terminal amino acids 42–51 of Mam-A(N42–51).Then,the N42–51 epitope-specific monoclonal antibody,mAb785,was conjugated to poly lactic-co-glycolic acid(PLGA)nanoparticles loaded with therapeutic agents,thereby enhancing the drug uptake and therapeutic efficacy in different genotypes of breast cancers.The computer simulation of the N42–51 epitope and the mAb785 structures,as well as their interactions,further revealed the specific targeting mechanism of the mAb785-conjugated nanoparticles to breast cancers.

以煤焦油中难转化的酚油为原料,采用Ni、W负载的MgO和酸碱耦合剂为催化剂,在高压反应釜内对酚油催化加氢制环烷烃反应性能进行研究。结果表明:P改性MgO(P质量分数为0.8%)与酸碱耦合剂均能有效降低加氢催化剂结晶度、优化孔道结构、提高产物环烷烃和芳烃选择性;在Ni-W活性金属负载量(w)为9.0%时,催化剂选择性加氢活性最高,环烷烃选择性达到57.5%。煤焦油加氢的最佳工艺条件为:氢初压8.0 MPa,温度380℃,反应时间60 min。

“产教融合、校企合作”是高职院校职业教育人才培养提升的重要方式,实现职教高素质和技术技能人才的有效途径。食品营养与检测专业通过校企共建高职人才培养模式,产教融合为特色的实训平台,实施专业课程项目化技能体系,项目化教学成果显著,学生职业技能同时得到提升。但是,在人才培养模式的实践下,课程不能与企业技术同步更新,缺乏有效的信息共享和沟通平台,顶岗实习较分散、难以管理等问题依然存在。因此,新时代背景下,“产教融合、校企合作”是当前职业教育改革和发展的重点,是解决当前职业教育问题的突破口和关键点。论证校企合作视域下高职食品营养检测专业校企合作人才培养模式改革的可行性,探索当前的校企合作人才培养模式构建与实施新路径。

The COVID-19 pandemic has emerged as a global health emergency due to its association with severe pneumonia and relative high mortality.However,the molecular characteristics and pathological features underlying COVID-19 pneumonia remain largely unknown.To characterize molecular mechanisms underlying COVID-19 pathogenesis in the lung tissue using a proteomic approach,fresh lung tissues were obtained from newly deceased patients with COVID-19 pneumonia.After virus inactivation,a quantitative proteomic approach combined with bioinformatics analysis was used to detect proteomic changes in the SARS-CoV-2-infected lung tissues.We identified significant differentially expressed proteins involved in a variety of fundamental biological processes including cellular metabolism,blood coagulation,immune response,angiogenesis,and cell microenvironment regulation.Several inflammatory factors were upregulated,which was possibly caused by the activation of NF-κB signaling.Extensive dysregulation of the lung proteome in response to SARS-CoV-2 infection was discovered.Our results systematically outlined the molecular pathological features in terms of the lung response to SARS-CoV-2 infection,and provided the scientific basis for the therapeutic target that is urgently needed to control the COVID-19 pandemic.

目的:观察一次离心运动后不同恢复时程大鼠骨骼肌程序性坏死的趋势变化,分析RIPK1/RIPK3途径在运动性骨骼肌损伤及修复中的可能作用。方法:70只雄性8周龄Sprague-Dawley(SD)大鼠据体重随机分为安静对照组(C,n=10)和运动组(E,n=60),其中E组又根据不同时程分为运动后即刻、运动后12 h、24 h、48 h、72 h、120 h共6组,每组10只。E组大鼠持续性下坡跑(-16°,16 m/min,90 min)。各组分别于相应时程点对比目鱼肌进行取材。透射电镜观察比目鱼肌超微结构变化,蛋白质印迹检测比目鱼肌程序性坏死蛋白RIPK1、p-RIPK1S166、RIPK3、p-RIPK3S232、MLKL、p-MLKLS358、HMGB1、IL-1α等表达。结果:1)一次离心运动后恢复期比目鱼肌超微结构发生改变,在恢复期12—48 h肌纤维排列紊乱,线粒体肿胀明显,肌膜下大量线粒体聚集;2)一次离心运动后恢复期比目鱼肌细胞膜形态改变,运动后48 h肌纤维细胞膜不完整、起皱受损明显,72 h与120 h逐渐恢复;3)MLKL表达在恢复期48 h达到峰值,在恢复期24 h(P<0.05)和48 h(P<0.01)明显升高;p-MLKLS358表达在恢复期48 h达到峰值,在恢复期48 h(P<0.01)和72 h(P<0.01)显著升高;p-MLKLS358/MLKL比值在恢复期72 h达到峰值,在恢复期12 h(P<0.05)和24 h(P<0.05)明显降低,72 h(P<0.05)显著升高;HMGB1表达在恢复期48 h达到峰值,在恢复期24 h(P<0.05)和48 h(P<0.01)显著升高;IL-1α表达在恢复期12 h达到峰值,在12 h(P<0.01)、24 h(P<0.05)和48 h(P<0.01)明显升高;4)RIPK1表达在恢复期48 h达到峰值,在恢复期运动后即刻(P<0.01)、12 h(P<0.01)、24 h(P<0.01)、48 h(P<0.01)、72 h(P<0.01)、120 h(P<0.01)均显著升高;RIPK3表达在恢复期48 h达到峰值,在恢复期48 h(P<0.01)、72 h(P<0.01)、120 h(P<0.01)明显升高;p-RIPK1S166蛋白表达呈现先升高后降低的趋势,在恢复期72 h(P<0.01)显著升高;p-RIPK3S232表达在恢复期72 h达到峰值,运动后即刻(P<0.05)和12 h(P<0.05)明显降低,72 h(P<0.05)显著升高。结论:1)一次离心运动后恢复期比目鱼肌发生程序性坏死现象,在运动后48 h相对明显;2)一次离心运动后恢复期比目鱼肌程序性坏死现象可能由RIPK1/RIPK3途径介导。
