重组小反刍兽疫病毒血凝素蛋白纯化体系的优化

本试验利用发酵技术诱导表达了小反刍兽疫病毒H蛋白胞外区(PPRV tH),优化建立了一套纯化重组PPRV tH的完整工艺体系。通过发酵表达获得的10 L菌液,菌体湿重约达30 g/L,均质机破碎菌体获得包涵体;Tris-0缓冲液(pH=8.0)洗涤包涵体3次,每1 g包涵体加入5 mL含20 mmol/L咪唑的Tris-E缓冲液(pH=8.0)重悬,4℃温和旋转溶解过夜,离心收集上清;按体积比1∶2将层析填料Chelaing Sepharose Fast Flow与上清混匀,以流速1~2 mL/min加入层析柱;依次通过10倍柱体积含50、100、400和500 mmol/L咪唑的Tris-E缓冲液(pH=8.0),4℃静置2 h进行洗脱。优化后的纯化体系可得到较纯的重组PPRV tH蛋白,利用灰度分析可知纯化后的重组PPRV tH蛋白纯度能达到85%。本试验提供的纯化体系可快速、简便、低成本、大批量纯化重组PPRV tH蛋白,而且纯化获得的PPRV tH蛋白具有良好的反应原性。

近年来随着全球气候的变化、人类活动领域的拓宽、动物及动物产品国际贸易的频繁,全球新发动物疫病不断出现,促使动物疫情防控形势日益复杂严峻,严重威胁了畜牧业持续健康发展与人类生命健康。然而有效的疫苗接种和预防是控制动物疾病最有效和最经济的干预措施,与传统疫苗相比,mRNA疫苗由于其研制周期短、易于产业化、生产工艺简单、易对新变体作出反应、能更好诱导免疫反应的特点引起了广泛的关注和研究。本文从mRNA疫苗的类别及递送系统、免疫机理、mRNA的修饰以及动物传染病mRNA疫苗等方面进行概述从而为动物疫病新型疫苗研制提供一定的参考和方向。

2019年8月10日,第一例牛结节性皮肤病在我国新疆伊犁哈萨克自治州确诊,此后疫情迅速蔓延至国内其他多个省份,牛群感染后严重影响皮张质量和产奶量,给养殖户造成重大经济损失。该文梳理了现有牛结节性皮肤病应用较为广泛的诊断方法及防控措施的研究进展,主要从临床综合诊断、实验室诊断、防控难点和防控建议等方面进行阐述。

非酒精性脂肪性肝炎(NASH)是一种临床常见病、多发病,因其可能发生纤维化、进展为肝硬化甚至肝癌,日益受到人们的重视。吉林省名中医米金铎教授认为,老年性NASH的病因病机为脾虚痰凝、瘀阻肝络,病机关键是脾虚失运,痰瘀互结于肝,肝络失和而致肝病。临证采用通补结合的治法,组方在逍遥散疏肝健脾的基础上加以活血化瘀、祛风化痰的药物,以达脾健肝疏、痰化络通的临床疗效。提出从“虚、痰、瘀”的角度出发,探讨老年性NASH的病机和中医治法,为防治非酒精性脂肪性肝炎提供了新的思路。

Sweet potato leaf tips have high nutritional value,and exploring the differences in the metabolic profiles of leaf tips among different sweet potato varieties can provide information to improve their *** this study,a UPLC-Q-Exactive Orbitrap/MS-based untargeted metabolomics method was used to evaluate the metabolites in leaf tips of 32 sweet potato *** varieties with distinct overall metabolic profiles(A01,A02,and A03),two varieties with distinct profiles of phenolic acids(A20 and A18),and three varieties with distinct profiles of flavonoids(A05,A12,and A16)were *** addition,a total of 163 and 29 differentially expressed metabolites correlated with the color and leaf shape of sweet potato leaf tips,respectively,were identified through morphological *** comparison analysis of the phenotypic traits and a metabolite-phenotypic trait correlation analysis indicated that the color differences of sweet potato leaf tips were markedly associated with ***,the level of polyphenols was correlated with the leaf shape of sweet potato leaf tips,with lobed leaf types having higher levels of polyphenols than the entire leaf *** findings on the metabolic profiles and differentially expressed metabolites associated with the morphology of sweet potato leaf tips can provide useful information for breeding sweet potato varieties with higher nutritional value.

Laser powder bed fusion (LPBF) is a commonly used additive manufacturing (AM) method for efficiently producing intricate geometric components. This investigation examines factors such as pores, cellular structure, grain size, and inclusions from the manufacturing process that contribute to the corrosion resistance of LPBF DSS. Furthermore, the as-built LPBF duplex stainless steel (DSS) is primarily ferrite due to the rapid cooling process. Therefore, the transformation of ferrite to austenite after various heat treatments in LPBF DSS and its corresponding corrosion resistance are presented. Additionally, a new mixed powder method is proposed to increase the austenite content in the as-built LPBF DSS. This review also focuses on the passivation capability and pitting corrosion performance in LPBF and conventional DSS. This article summarizes the variations in microstructure between as-built and heat-treated LPBF DSS, with their impacts on corrosion resistance, offering insights for manufacturing highly corrosion-resistant LPBF DSS.

The flowers of ZhaoFen and RouFurong may contain essential oils with natural aromatic ingredients. In the present work, the chemical compositions of essential oil of Paeonia suffruticosa Andrews from the flowers of ZhaoFen and RouFurong grown only in China were investigated by GC-MS analysis. The results indicate that there are 27 constituents in ZhaoFen and 29 constituents in RouFurong, which account for 96.04% and 95.90% of the oils of ZhaoFen and RouFurong, respectively. The major components of the essential oils are character-rized by oxygenated terpenols, and their content is, respectively, 85.06% and 83.47%. The essential oil of Paeonia suffruticosa Andrews was reported for the first time on the aerial parts.
