Enhanced potency of an IgM-like nanobody targeting conserved epitope in SARS-CoV-2 spike N-terminal domain

Almost all the neutralizing antibodies targeting the receptor-binding domain(RBD)of spike(S)protein show weakened or lost efficacy against severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)emerged or emerging variants,such as Omicron and its *** suggests that highly conserved epitopes are crucial for the development of neutralizing ***,we present one nanobody,N235,displaying broad neutralization against the SARS-CoV-2 prototype and multiple variants,including the newly emerged Omicron and its ***-electron microscopy demonstrates N235 binds a novel,conserved,cryptic epitope in the N-terminal domain(NTD)of the S protein,which interferes with the RBD in the neighboring S *** neutralization mechanism interpreted via flow cytometry and Western blot shows that N235 appears to induce the S1 subunit shedding from the trimeric S ***,a nano-IgM construct(MN235),engineered by fusing N235 with the human IgM Fc region,displays prevention via inducing S1 shedding and cross-linking virus *** to N235,MN235 exhibits varied enhancement in neutralization against pseudotyped and authentic viruses in *** intranasal administration of MN235 in low doses can effectively prevent the infection of Omicron sub-variant BA.1 and XBB in vivo,suggesting that it can be developed as a promising prophylactic antibody to cope with the ongoing and future infection.

Drogue detection for autonomous aerial refueling via hybrid pigeon-inspired optimized color opponent and saliency aggregation

Drogue detection is one of the challenging tasks in autonomous aerial refueling due to the requirement for accuracy and *** detection based on image intrinsic cues can achieve fast detection,but with poor *** studies reveal that optimization-based methods provide accurate and quick solutions for saliency *** paper presents a hybrid pigeon-inspired optimization method,the optimized color opponent,that aims to adjust the weight of color opponent channels to detect the drogue *** can optimize the weights in the selected aerial refueling scene offline,and the results are applied for drogue detection in the scene.A novel algorithm aggregated by the optimized color opponent and robust background detection is presented to provide better precision and *** results on benchmark datasets and aerial refueling images show that the proposed method successfully extracts the saliency region or drogue and exhibits superior performance against the other saliency detection methods with intrinsic *** algorithm designed in this paper is competent for the drogue detection task of autonomous aerial refueling.

Flow instability of supercritical hydrocarbon fuel in a multi-channel cooling system

Thermally-induced flow instabilities are a critical issue in multi-channel regenerative cooling *** particular,the interactions between Density-Wave Oscillations(DWO)and Flow Maldistribution(FMD)can result in complex and disastrous instability *** study investigates the instability behaviors of hydrocarbon fluid in a four-channel system with a constant heat flux ratio using both frequency-and time-domain *** the heat flux increases,the in-tube flow sequentially destabilizes in each channel and converges to new equilibrium states,leading to the emergence of FMD *** also causes the system eigenvalue to change repeatedly from negative to positive rather than increasing ***,the system eigenvalues are between those of the two most unstable channels,indicating that the stability behavior of the entire system is dictated by the most unstable *** FMD occurs,flow oscillations are activated in channels with weak stability,and the in-tube flow is observed to evolve into various flow patterns,including stable flow,self-sustained oscillation,oscillation divergence,quasiperiodic oscillation,and oscillation *** novel instability mode of oscillation excursion involves a spontaneous transition of operating *** oscillates from an equilibrium state and then stabilizes at a new operational state after oscillation-induced ***,the newfound stable state may also be only temporary,with the in-tube flow regressing to the initial state,resulting in quasi-periodic oscillation.

Single-cell mass spectrometry studies of drug metabolism heterogeneity and primary resistance to gefitinib in non-small cell lung cancer cells

Patients with epidermal growth factor receptor(EGFR)wild-type non-small cell lung cancer(NSCLC)often show primary resistance to gefitinib *** is thus necessary to study the metabolism of gefitinib in NSCLC cells to comprehensively reveal the reasons for the primary resistance of ***,we develop a platform for studying drug metabolism heterogeneity based on single-cell mass spectrometry(sDMH-scMS)by integrating living-cell electrolaunching ionization MS(ILCEI-MS)and high-performance liquid chromatography-MS(HPLC-MS)analysis,and the primary resistance of NSCLC cells to gefitinib was studied using this *** ILCEI-MS analysis showed that approximately 11.9%of NSCLC single cells contained the gefitinib metabolite M11;HPLC-MS detection diluted the intensity of M11 in subpopulations and concealed the heterogeneity of drug metabolism in tumor single *** intensity of gefitinib in EGFR wild-type A549 cells was markedly lower than in mutant PC9 cells,and the intensity of gefitinib metabolites was significantly higher than in PC9 cells,suggesting that the primary resistance of NSCLC cells is related to gefitinib ***,the combination of gefitinib and the drug-metabolizing enzyme inhibitorα-naphthoflavone was shown to overcome the primary resistance of the NSCLC ***,the results of this study are expected to be applicable for clinical drug resistance diagnosis and treatment at the single-cell level.

A Systematic Review and Performance Evaluation of Open-Source Tools for Smart Contract Vulnerability Detection

With the rise of blockchain technology,the security issues of smart contracts have become increasingly *** the availability of numerous smart contract vulnerability detection tools,many face challenges such as slow updates,usability issues,and limited installation *** challenges hinder the adoption and practicality of these *** paper examines smart contract vulnerability detection tools from 2016 to 2023,sourced from the Web of Science(WOS)and Google *** systematically collecting,screening,and synthesizing relevant research,38 open-source tools that provide installation methods were selected for further *** a developer’s perspective,this paper offers a comprehensive survey of these 38 open-source tools,discussing their operating principles,installation methods,environmental dependencies,update frequencies,and installation *** on this,we propose an Ethereum smart contract vulnerability detection *** framework enables developers to easily utilize various detection tools and accurately analyze contract security *** validate the framework’s stability,over 1700 h of testing were ***,a comprehensive performance test was performed on the mainstream detection tools integrated within the framework,assessing their hardware requirements and vulnerability detection *** results indicate that the Slither tool demonstrates satisfactory performance in terms of system resource consumption and vulnerability detection *** study represents the first performance evaluation of testing tools in this domain,providing significant reference value.

Invasion of fall armyworm led to the succession of maize pests in Southwest China

The invasive fall armyworm Spodoptera frugiperda(***)invaded Asia in 2018,colonizing the tropical and southern subtropical regions as well as migrating with the monsoons into Northeast Asia during spring and *** has resulted in widespread infestations,with significant impacts on maize production in various Asian *** studies have shown that the invasion of this pest can alter the species relationships of maize pests,but the actual impact on maize pest management is still *** study investigated the changes in maize pest occurrence and pesticide use in the annual breeding areas of *** in Yunnan Province and the Guangxi Zhuang Autonomous Region of China during 2017-2021,based on surveys and interviews with small farmers in maize *** results showed that *** has emerged as the dominant species among maize pests after invasion and colonization,replacing traditional pests such as Ostrinia furnacalis,Spodoptera litura,Agrotis ypsilon,and Rhopalosiphum *** variety of pesticides used for maize pest control has changed from chlorpyrifos,lambda-cyhalothrin,and acetamiprid to emamectin benzoate-based pesticides with high effectiveness against ***,the frequency of maize pest chemical applications has increased from an average of 5.88 to 7.21 times per season,with the amounts of pesticides used in summer and autumn maize being significantly higher than in winter and spring maize,thereby increasing application costs by more than 35%.The results of this study clarified the impact of *** invasion on maize pest community succession and chemical pesticide use in tropical and south subtropical China,thereby providing a baseline for modifying the regional control strategies for maize pests after the invasion of this relatively new pest.

Mutations in the SARS-CoV-2 spike receptor binding domain and their delicate balance between ACE2 affinity and antibody evasion

Intensive selection pressure constrains the evolutionary trajectory of SARS-CoV-2 genomes and results in various novel variants with distinct mutation *** mutations,particularly those within the receptor binding domain(RBD)of SARS-CoV-2 spike(S)protein,lead to the functional alteration in both receptor engagement and monoclonal antibody(mAb)***,we review the data of the RBD point mutations possessed by major SARS-CoV-2 variants and discuss their individual effects on ACE2 affinity and immune *** single amino acid substitutions within RBD epitopes crucial for the antibody evasion capacity may conversely weaken ACE2 binding ***,this weakened effect could be largely compensated by specific epistatic mutations,such as N501Y,thus maintaining the overall ACE2 affinity for the spike protein of all major *** predominant direction of SARS-CoV-2 evolution lies neither in promoting ACE2 affinity nor evading mAb neutralization but in maintaining a delicate balance between these two ***,this review interprets how RBD mutations efficiently resist antibody neutralization and meanwhile how the affinity between ACE2 and spike protein is maintained,emphasizing the significance of comprehensive assessment of spike mutations.

Electrochemical energy storage and conversion techniques that exhibit the merits such as high energy density,rapid response kinetics,economical maintenance requirements and expedient installation procedures will hold a pivotal role in the forthcoming energy storage technologies *** recent years,aqueous zinc-ion batteries（AZIBs） have garnered substantial attention as a compelling candidate for large-scale energy storage systems,primarily attributable to their advantageous featu res encompassing cost-effectiveness,environmental sustainability,and robust safety ***,one of the primary factors hindering the further development of AZIBs originates from the challenge of cathode ***,the three mainstream types of mainstream cathode materials,in terms of manganese-based compounds,vanadium-based compounds and Prussian blue analogues,surfer from the dissolution of Mn2+,in the low discharge voltage,and the low specific capacity,*** strategies have been developed to compensation the above intrinsic defects for these cathode materials,including the ionic doping,defect engineering,and materials ***,this review first provides a systematic summarization of the zinc storage mechanism in AZIBs,following by the inherent merit and demerit of three kind of cathode materials during zinc storage analyzed from their structure characteristic,and then the recent development of critical strategies towards the intrinsic insufficiency of these cathode *** this review,the methodologies aimed at enhancing the efficacy of manganese-based and vanadium-based compounds are emphasis ***,the article outlines the future prospective directions as well as strategic proposal for cathode materials in AZIBs.
