Design and aligner-assisted fast fabrication of a microfluidic platform for quasi-3D cell studies on an elastic polymer

While most studies of mechanical stimulation of cells are focused on two-dimensional(2D)and three-dimensional(3D)systems,it is rare to study the effects of cyclic stretching on cells under a quasi-3D microenvironment as a linkage between 2D and 3D.Herein,we report a new method to prepare an elastic membrane with topographic microstructures and integrate the membrane into a microfluidic chip.The fabrication difficulty lay not only in the preparation of microstructures but also in the alignment and bonding of the patterned membrane to other layers.To resolve the problem,we designed and assembled a fast aligner that is cost-effective and convenient to operate.To enable quasi-3D microenvironment of cells,we fabricated polydimethylsiloxane(PDMS)microwell arrays(formed by micropillars of a few microns in diameter)with the microwell diameters close to the cell sizes.An appropriate plasma treatment was found to afford a coating-free approach to enable cell adhesion on PDMS.We examined three types of cells in 2D,quasi-3D,and 3D microenvironments;the cell adhesion results showed that quasi-3D cells behaved between 2D and 3D cells.We also constructed transgenic human mesenchymal stem cells(hMSCs);under cyclic stretching,the visualizable live hMSCs in microwells were found to orientate differently from in a 3D Matrigel matrix and migrate differently from on a 2D flat plate.This study not only provides valuable tools for microfabrication of a microfluidic device for cell studies,but also inspires further studies of the topological effects of biomaterials on cells.

The metallurgical properties of the CaO–SiO_(2)–Al_(2)O_(3)–4.6wt%Mg O–Fe_(2)O_(3)slag system,formed by the co-treatment process of spent automotive catalyst(SAC)and copper-bearing electroplating sludge(CBES),were studied systematically in this paper.The slag structure,melting temperature,and viscous characteristics were investigated by Fourier transform infrared(FTIR)spectroscopy,Raman spectroscopy,Fact Sage calculation,and viscosity measurements.Experimental results show that the increase of Fe_(2)O_(3)content(3.8wt%–16.6wt%),the mass ratio of CaO/SiO_(2)(m(CaO)/m(SiO_(2)),0.5–1.3),and the mass ratio of SiO_(2)/Al_(2)O_(3)(m(SiO_(2))/m(Al_(2)O_(3)),1.0–5.0)can promote the depolymerization of silicate network,and the presence of a large amount of Fe_(2)O_(3)in form of tetrahedral and octahedral units ensures the charge compensation of Al^(3+)ions and makes Al_(2)O_(3)only behave as an acid oxide.Thermodynamic calculation and viscosity measurements show that with the increase of Fe_(2)O_(3)content,m(Ca O)/m(SiO_(2)),and m(SiO_(2))/m(Al_(2)O_(3)),the depolymerization of silicate network structure and low-melting-point phase transformation first occur within the slag,leading to the decrease in melting point and viscosity of the slag,while further increase causes the formation of high-melting-point phase and a resultant re-increase in viscosity and melting point.Based on experimental analysis,the preferred slag composition with low polymerization degree,viscosity,and melting point is as follows:Fe_(2)O_(3)content of 10.2wt%–13.4wt%,m(CaO)/m(SiO_(2))of 0.7–0.9 and m(SiO_(2))/m(Al_(2)O_(3))of 3.0–4.0.This work provides a theoretical support for slag design in co-smelting process of SAC and CBES.

Seasonal variation of the shape and location of the Luzon cold eddy

Using observational data from multiple satellites,we studied seasonal variations of the shape and location of the Luzon cold eddy(LCE)northwest of Luzon Island.The shape and location of the LCE have obvious seasonal variations.The LCE occurs,develops,and disappears from December to April of the next year.During this period,the shape of the LCE changed from a flat ellipse to a circular ellipse,and the change in shape can be reflected by the increase of the ellipticity of the LCE from 0.16 to 0.82.The latitude of center location of the LCE changes from 17.4°N to 19°N,and the change in latitude can reach 1.6°.Further study showed that seasonal variation of the northeast monsoon intensity leads to the change in the shape and location of the LCE.The seasonal variation of the LCE shape can significantly alter the spatial distribution of the thermal front and chlorophyll a northwest of the Luzon Island by geostrophic advection.

Left-Right Symmetry or Asymmetry of Cells on Stripe-Like Micropatterned Material Surfaces

Material surfaces can induce cell responses such as contact guidance, yet little attention has been paid to further cell orientation. Herein, we report an interesting phenomenon of cell orientation beyond the classic contact guidance on a stripe-like micropattern with cell-adhesive arginineglycine-aspartate （RGD） peptides on a nonfouling background decorated by poly（ethylene glycol）（PEG）. Such a micropattern with cell adhesion contrast led to significant contact guidance after cell seeding. What is more, the localized and elongated cells were found to be further orientated out of the adhesive stripes, and even an anticlockwise rotation was observed for rat mesenchymal stem cells （rMSCs）. The left-right asymmetry of rMSCs stood only in statistics, for we observed all cases including clockwise orientation, anticlockwise orientation or just keeping the orientation of previous contact guidance. We further found that human foreskin fibroblasts （HFFs） preferred a clockwise rotation, while human mesenchymal stem cells （hMSCs） and human umbilical vascular endothelial cells （HUVECs） exhibited no significant preference to either direction, which indicated that the left-right symmetry or asymmetry was cell-type dependent. The present report has partially confirmed the cell chirality and revealed its complexity, calling for further careful and comprehensive investigation of the challenging topic of cell chirality on material surfaces.

In this manuscript,we first report an ultrasensitive detection assay of microRNA by combing asymmetric polymerase chain reaction(A-PCR)and loop-mediated isothermal amplification(LAMP)technology.Using A-PCR obtained an extended single strand to form LAMP stem-loop structure under isothermal amplification conditions.We used miRNAs as a loop primer probe in LAMP reaction and completed its ultrasensitive and rapid detection.The established method furnished a fast,specific and efficient detection of target miRNA with a detection limit as low as 10 amol/L in 90 min.

A Blockchain-Based Authentication Protocol for WLAN Mesh Security Access

In order to deploy a secure WLAN mesh network,authentication of both users and APs is needed,and a secure authentication mechanism should be employed.However,some additional configurations of trusted third party agencies are still needed on-site to deploy a secure authentication system.This paper proposes a new block chain-based authentication protocol for WLAN mesh security access,to reduce the deployment costs and resolve the issues of requiring key delivery and central server during IEEE 802.11X authentication.This method takes the user’s authentication request as a transaction,considers all the authentication records in the mesh network as the public ledger and realizes the effective monitoring of the malicious attack.Finally,this paper analyzes the security of the protocol in detail,and proves that the new method can solve the dependence of the authentication node on PKI and CA.

Mixed matrix membrane containing metal oxide nanosheets for efficient CO_(2)separation

The two-dimensional(2D)nanosheet zinc cobaltate(ZnCo_(2)O_(4))was added into polyether block amide(Pebax)matrix to prepare mixing matrix membrane(MMM)for separating carbon dioxide(CO_(2))/methane(CH4)gas mixture.The 2D porous ZnCo_(2)O_(4)nanosheets were composed of chemically interconnected metal oxide nanoparticles.The ZnCo_(2)O_(4)nanoparticles in the nanosheets constructed large-quantity pores of 11.78 nm and provided abundant transfer channels for gas molecule.Moreover,the synergistic effect of bimetallic Zn^(2+)and Co^(2+)would promote the generation of oxygen vacancies(Oδ-),which could provide more CO_(2)(Cδ+)adsorption sites,thereby increased the selectivity of the membrane.The large aspect ratio of the ultra-thin ZnCo_(2)O_(4)nanosheets showed better dispersion in the membrane.The pure gas separation performance data showed the CO_(2)permeability and CO_(2)/CH4 selectivity of Pebax/ZnCo_(2)O_(4)membrane were 139.10 Barrer and 15.38,respectively,when the filling amount was 0.5 wt%.Compared with pure Pebax membrane,the separation performance(permeability and selectivity)were increased with 165.67%and 75.57%,respectively.

The promoting effect of health education on medical treatment seeking behavior in the context of healthy China:based on the comparison of migrants and registered local residents

The migrants are the important participant and contributor in China’s economic and social construction,but they still have many restrictions on their access to health services.This paper analyzes the differences between the migrants and the regis-tered local residents in looking for medical treatment and explores the relationship between health education and medical treatment seeking behavior,so as to find an appropriate way to improve the health level of the migrants.By using the compara-tive survey data of the floating population and registered population in 2017 and establishing the binary Logit model,this paper finds that the health education has a promoting effect on medical treatment seeking behaviors.In terms of the influence mechanism of health education on medical treatment seeking behavior,the mediat-ing effect of social participation factors is stronger in the migrants’group because making new friends and learning about local public health resources through health education is of great significance to the migrants.It is proposed to expand the cov-erage of health education among migrant groups and improve the accessibility of health education.At the same time,public medical and health services should be used as a window to guide the migrants to seek medical treatment in an orderly manner while assisting their social integration.

Orthodontic treatment monitoring involves using current images and previous 3D models to estimate the relative position of individual teeth before and after orthodontic treatment.This process differs from image-based object 6D pose estimation due to the gingiva deformation and varying pose offsets for each tooth during treatment.Motivated by the fact that the poses of molars remain relatively fixed in implicit orthodontics,we design an approach that employs multiview pose evaluation and bidirectional temporal propagation for jaw pose estimation and then employs an iteration-based method for tooth alignment.To handle changes in tooth appearance or location with weak texture across frames,we also introduce an instance propagation module that leverages positional and semantic information to explore instance relations in the temporal domain.We evaluated the performance of our approach using both the Shining3D tooth pose dataset and the Aoralscan3 tooth registration dataset.Our experimental results demonstrate remarkable accuracy improvements compared with existing methods.

Drought stress,mercuric chloride,andβ-mercaptoethanol effects on hydraulic characteristics of three cultivars of wolfberry(Lycium chinense)

In order to further understand the effects of drought stress on hydraulic characteristics and the relationship between hydraulic conductivity and aquaporins or water channels of root systems of three wolfberry cultivars(Lycium chinense Mill.),hydraulic conductivity of 2-yearold pot-grown seedlings was measured under drought stress,rewatering,and treatment with exogenous mercuric chloride andβ-mercaptoethanol.Under moderate and severe drought stress levels,the most significant decrease of hydraulic conductivity was 37.3%and 24.0%,respectively,in the‘Ningqi 5’cultivar compared with the nonstressed control.After rewatering,the rate of recovery in specific conductivity was most rapid in the‘Mengqi 1’cultivar,at 0.058 and 0.072 kg MPa^-1 m^-2 s^-1 h^-1 under moderate and severe drought stress levels,respectively.The‘Mengqi 1’cultivar had the highest recovery degree of hydraulic conductivity under two concentrations ofβ-ME(500 or 1000 lmol L^-1),reaching 82.4%and 88.5%,respectively,of the initial conductivity.The adaptive capacity of hydraulic conductivity in the‘Ningqi 5’cultivar was weaker than in the‘Ningqi 1’and‘Mengqi 1’cultivars under drought stress.The recovery capacity of hydraulic conductivity in‘Mengqi 1’cultivar was stronger than the‘Ningqi 1’and‘Ningqi 5’cultivars after rewatering.Aquaporins of the‘Ningqi 1’cultivar root systems had the highest binding affinity with mercuric chloride,which was the most likely cause in the decrease in hydraulic conductivity,whereas aquaporins of‘Mengqi 1’root systems had the weakest binding affinity.The inhibitory effect of mercuric chloride was readily eliminated byβ-mercaptoethanol in the‘Mengqi 1’cultivar.The hydraulic characteristics of this cultivar were more sensitive to drought,mercuric chloride andβ-mercaptoethanol than the other cultivars.
