Single-cell joint analysis of methylome and transcriptome reveals how the methylation regulates the transcriptional ***,traditional bench-top protocols for single-cell DNA methylation and RNA transcription co-detection are laborintensive,cost-ineffective and ***,we establish the DMF-sc MT-seq,a highly-efficient and cost-effective method to simultaneously analyze single-cell DNA methylation and transcriptional activity based on digital *** MT-seq automates the workflow of single-cell isolation,cellular hypotonic lysis,nucleic acid separation and methylome/transcriptome library construction in a contactless and addressable *** system ensures high accuracy(R>0.85),high gene detection ability(14,697 genes per cell at 4 million sequencing depth),and high CpG coverage(677,198 CpG sites per cell at 1million sequencing depth).By using DMF-sc MT-seq,the relationship of DNA methylation and RNA transcription under different genomic contexts is *** further apply DMF-sc MT-seq to study the dynamics of transcription regulation with methylation-inhibiting anti-tumor Decitabine,and identify the methylated promoter/gene body driven genes in response to Decitabine ***-sc MT-seq facilitates the construction of the correlation of DNA methylation and transcriptional activity at the single-cell level in a flexible,sensitive and accurate way,which is anticipated to be a powerful tool in studying single-cell biological systems.

In this paper, an HIV-1 infection model with intracellular delay, humoral immunity and immune impairment is investigated, in which both virus-to-cell infection and cell-to-cell transmission are *** basic reproduction ratio is calculated and the existence of feasible equilibria is established. By analyzing the distributions of roots of the corresponding characteristic equations, the local asymptotic stability of each of feasible equilibria is established. With the help of appropriate Lyapunov functionals and LaSalle’s invariance principle, it is proved that if the basic reproduction ratio is less than unity, the infection-free equilibrium is globally asymptotically stable, and the virus is eventually eliminated; if the basic reproduction ratio is greater than unity, the chronic-infection equilibrium is globally asymptotically stable. Finally, numerical simulations are carried out to illustrate the effects of some parameters on HIV-1 infection dynamics.

The ecological environment of the Yellow River Basin has become more fragile under the combined action of natural and manmade ***,the change mechanisms of ecological vulnerability in different sub-regions and periods vary,and the reasons for this variability are yet to be ***,in this study,we proposed a new remote sensing ecological vulnerability index by considering moisture,heat,greenness,dryness,land degradation,and social economy indicators and then analyzed and disclosed the spatial and temporal change patterns of ecological vulnerability of the Yellow River Basin,China from 2000 to 2022 and its driving *** results showed that the newly proposed remote sensing ecological vulnerability index had a high accuracy,at 86.36%,which indicated a higher applicability in the Yellow River *** 2000 to 2022,the average remote sensing ecological vulnerability index of the Yellow River Basin was 1.03,denoting moderate vulnerability *** intensive vulnerability area was the most widely distributed,which was mostly located in the northern part of Shaanxi Province and the eastern part of Shanxi *** 2000 to 2022,the ecological vulnerability in the Yellow showed an overall stable trend,while that of the central and eastern regions showed an obvious trend of *** gravity center of ecological vulnerability migrated southwest,indicating that the aggravation of ecological vulnerability in the southwestern regions was more severe than in the northeastern regions of the *** dominant single factor of changes in ecological vulnerability shifted from normalized difference vegetation index(NDVI)to temperature from 2000 to 2022,and the interaction factors shifted from temperature∩NDVI to temperature∩precipitation,which indicated that the global climate change exerted a more significant impact on regional *** above results could provide decision support for the ecological protection and restoration of the Y

Covalent organic frameworks(COFs),which are constructed by linking organic building blocks via dynamic covalent bonds,are newly emerged and burgeoning crystalline porous copolymers with features including programmable topological architecture,pre-designable periodic skeleton,well-defined micro-/meso-pore,large specific surface area,and customizable electroactive *** benefits make COFs as promising candidates for advanced electrochemical energy ***,for now,structure engineering of COFs from multiscale aspects has been conducted to enable optimal overall electrochemical performance in terms of structure durability,electrical conductivity,redox activity,and charge *** this review,we give a fundamental and insightful study on the correlations between multi-scale structure engineering and eventual electrochemical properties of COFs,started with introducing their basic chemistries and charge storage *** careful discussion on the significant achievements in structure engineering of COFs from linkages,redox sites,polygon skeleton,crystal nanostructures,and composite microstructures,and further their effects on the electrochemical behavior of COFs are ***,the timely cutting-edge perspectives and in-depth insights into COFbased electrodematerials to rationally screen their electrochemical behaviors for addressing future challenges and implementing electrochemical energy storage applications are proposed.

Effects of organic fertilizers on organic carbon accumulation in alkalized saline soil and silage maize yield

To understand the combined effect of organic and chemical fertilizers on soil carbon emissions and carbon balance of a farmland ecosystem,this study investigated the organic fertilizer nitrogen replacing different proportions of chemical fertilizer *** results showed that,compared to F_(100),the O_(15)F_(85) treatment increased the yield and net ecosystem productivity carbon sequestration of silage maize under mild,moderate,and severe salinization levels,as well as the contents of soil organic carbon,microbial carbon,and humin carbon,while reducing plant carbon *** O_(15)F_(85) treatment did not significantly increase soil carbon emissions(CEC),but O_(30)F_(70),O_(45)F_(55) and O_(100) treatments significantly increased *** soil carbon balance analysis showed that the farmland ecosystem was a“sink”for atmospheric CO_(2) under each *** O_(15)F_(85) treatment produced an“excitation effect”to enhance the carbon sink effect of silage maize farmland under mild,moderate and severe salinization levels while maintaining stable production and *** the O_(100) treatment increased the carbon sink of farmland under different salinization levels,the yield was significantly reduced and did not represent practical production *** analysis showed that soil organic carbon components and ecosystem carbon balance were closely related to soil total salt,pH and bulk density,while soil dissolved organic carbon,humus carbon components and carbon emissions were closely related to soil moisture and ***,the purpose of improving the carbon sink of saline-alkali land can be achieved through soil salt inhibition,soil structure remodeling and water supplement and warming regulation,which provides technical and theoretical support for reducing carbon emissions,achieving carbon neutrality and alleviating global warming.

The assembly of exogenous artificial architectures inside cells can regulate a series of biological events,which heavily relies on the development of spatiotemporally controlled molecular assembly *** herein report a designer deoxyribonucleic acid(DNA)nanostructure that enables light-mediated spatiotemporally dynamic assembly in living cells and consequently achieves efficient regulation of cell *** DNA nanostructure was constructed from i-motif moiety-containing branched DNA,photocleavable bond-containing linker,and tumor cell-targeting *** cellular uptake mediated by aptamers,under the spatiotemporal control of both UV light and late endosomal/lysosomal acidic environments,disassembly/reassembly of DNA nanostructure occurred via two rationally designed routes,generating microsized DNA *** a result,autophagy was significantly enhanced with the increase of DNA assembly *** enhanced autophagy showed an impact on related biological *** system is expected to be a powerful tool for the regulation of intracellular events and cellular behaviors.

Spinel LiNi_(0.5-x)Mn_(1.5+x)O_(4)(LNMO)has attracted intensive interest for lithium-ion battery due to its high voltage and high energy ***,severe capacity fade attributed to unstable surface structure has hampered its *** vacancies(OVs)tend to occur in the surface of the material and lead to surface structure reconstruction,which deteriorates the battery performance during electrochemical ***,we utilize high-temperature-shock(HTS)method to synthesize LNMO materials with fewer surface *** calcination drives lower surface OVs concentration,reducing the content of Mn^(3+)and surface reconstruction layers,which is beneficial to obtain a stable crystal *** LNMO material synthesized by HTS method delivers an initial capacity of 127 mAh·g^(-1) at 0.1 C and capacity retention of 81.6%after 300 cycles at 1 C,and exhibits excellent performance at low temperature.

A novel Nd-Fe-B type permanent magnet with excellent thermal stability was designed by Co replacing Fe in the main phase and the grain boundary *** remanence and coercivity temperature coefficient reach 0.058%/℃and 0.465%/℃in the te mperature range from 25 to 100℃,which are much lower than those of commercial Nd-Fe-B *** enhanced Curie temperature is obtained for the novel magnet due to the Co substitution,which significantly improves the operating *** microstructure result reveals that an amorphous phase exists in the intergranular grains which is probably responsible for the deterioration of intrinsic *** work can provide a reference for the design and optimization of components of sintered Nd-Fe-B magnets with excellent thermal stability.

Effect of titanium on the wettability between Fe-P-Ti alloy and Al2O3 substrate

It is of significance that the wettability between the molten alloy and the Al2O3affects the inclusion removal,nozzle clogging and refractory erosion. In the present work, the wettability between the Al2O3substrate and Fe–P–Ti alloys with different Ti content was investigated by using the sessile droplets method. The interfacial characteristics were investigated by scanning electron microscope（SEM）. The contact angles between the sintered Al2O3substrate and Fe–P–Ti alloys with 0, 0.05, 0.10 and 0.15 wt% Ti are 100.1°, 97.3°, 94.5°and 90.5°,respectively. The oxidation of [Al] and [Ti] in the molten alloys leads to the formation of the oxides on the droplets. With the increase of Ti content in the Fe–P–Ti alloys, the formed oxides change from Al2O3to Ticontaining oxides. The reaction between [Ti] and Al2O3substrate in the interface results in the formation of Ti-containing oxides. The quantity of the interfacial oxides increases with the increase of Ti content in the *** a result, the synergetic effect of the formation of the interfacial products and the oxides on the surface of droplets leads to the decrease of the contact angles between the Al2O3substrate and Fe–P–Ti alloys due to the increase of Ti content.
