MdbHLH162 connects the gibberellin and jasmonic acid signals to regulate anthocyanin biosynthesis in apple

Anthocyanins are secondary metabolites induced by environmental stimuli and developmental signals.The positive regulators of anthocyanin biosynthesis have been reported,whereas the anthocyanin repressors have been neglected.Although the signal transduction pathways of gibberellin(GA)and jasmonic acid(JA)and their regulation of anthocyanin biosynthesis have been investigated,the cross-talk between GA and JA and the antagonistic mechanism of regulating anthocyanin biosynthesis remain to be investigated.In this study,we identified the anthocyanin repressor MdbHLH162 in apple and revealed its molecular mechanism of regulating anthocyanin biosynthesis by integrating the GA and JA signals.MdbHLH162 exerted passive repression by interacting with MdbHLH3 and MdbHLH33,which are two recognized positive regulators of anthocyanin biosynthesis.MdbHLH162 negatively regulated anthocyanin biosynthesis by disrupting the formation of the anthocyanin-activated MdMYB1-MdbHLH3/33complexes and weakening transcriptional activation of the anthocyanin biosynthetic genes MdDFR and MdUF3GT by MdbHLH3 and MdbHLH33.The GA repressor MdRGL2a antagonized MdbHLH162-mediated inhibition of anthocyanins by sequestering MdbHLH162 from the MdbHLH162-MdbHLH3/33 complex.The JA repressors MdJAZ1 and MdJAZ2 interfered with the antagonistic regulation of MdbHLH162 by MdRGL2a by titrating the formation of the MdRGL2a-MdbHLH162 complex.Our findings reveal that MdbHLH162 integrates the GA and JA signals to negatively regulate anthocyanin biosynthesis.This study provides new information for discovering more anthocyanin biosynthesis repressors and explores the cross-talk between hormone signals.

Structural Optimization of Polyimide Foam via Composition with Hyperbranched Polymer Modified Fluorinated Carbon Nanotubes

The key to improve the foam’s performance is to optimize the cellular structure and its bulk-material composition.Here,the hyperbranched polymer modified fluorinated multi-walled carbon nanotube(HPMCNT-F)based on the nucleophilic reactions of MCNT-F was successfully prepared and used to composite with PI foams.The pristine MCNT shows a poor dispersity and weak interfacial interaction with PI matrix.While HPMCNT-F exhibits an excellent dispersity and effectively forms covalent/non-covalent interaction with PI matrix due to the surface-structure engineering,resulting in the enhancement of the PI bulk.Furthermore,HPMCNT-F works as a heterogeneous nucleation agent in PI foam to optimize the cellular structure.The enhancement of PI bulk and the optimizing of cellular structure result in the increase of compressive special strength of composite foam by 58.9%with a low loading of 1.6 wt%HPMCNT-F.Moreover,the hyperbranched polymers effectively prevent the thermal conduction among HPMCNT-F,and the isolated MCNTs effectively block thermal radiation through absorption and reflection the infrared waves.Thus,the thermal conductivity was reduced by 8.0%simultaneously.

Potential Medicinal Value of Rhein for Diabetic Kidney Disease

Diabetic kidney disease(DKD)is the primary cause of mortality among diabetic patients.With the increasing prevalence of diabetes,it has become a major concern around the world.The therapeutic effect of clinical use of drugs is far from expected,and therapy choices to slow the progression of DKD remain restricted.Therefore,research on new drugs and treatments for DKD has been a hot topic in the medical field.It has been found that rhein has the potential to target the pathogenesis of DKD and has a wide range of pharmacological effects on DKD,such as anti-nephritis,decreasing blood glucose,controlling blood lipids and renal protection.In recent years,the medical value of rhein in the treatment of diabetes,DKD and renal disease has gradually attracted worldwide attention,especially its potential in the treatment of DKD.Currently,DKD can only be treated with medications from a single symptom and are accompanied by adverse effects,while rhein improves DKD with a multi-pathway and multi-target approach.Therefore,this paper reviews the therapeutic effects of rhein on DKD,and proposes solutions to the limitations of rhein itself,in order to provide valuable references for the clinical application of rhein in DKD and the development of new drugs.

Volatiles of Shiraia fruiting body-associated Pseudomonas putida No.24 stimulate fungal hypocrellin production

Hypocrellins are major bioactive perylenequinones from Shiraia fruiting bodies and have been developed as efficient photosensitizers for photodynamic therapy.Pseudomonas is the second dominant genus inside Shiraia fruiting bodies,but with less known actions on the host fungus.In this work,the effects of bacterial volatiles from the Shiraia-associated Pseudomonas on fungal hypocrellin production were investigated.Pseudomonas putida No.24 was the most active to promote significantly accumulation of Shiraia perylenequinones including hypocrellin A(HA),HC,elsinochrome A(EA)and EC.Headspace analysis of the emitted volatiles revealed dimethyl disulfide as one of active compounds to promote fungal hypocrellin production.The bacterial volatiles induced an apoptosis in Shiraia hyphal cell,which was associated with the generation of reactive oxygen species(ROS).ROS generation was proved to mediate the volatile-induced membrane permeability and up-regulation of gene expressions for hypocrellin biosynthesis.In the submerged volatile co-culture,the bacterial volatiles stimulated not only HA content in mycelia,but also HA secretion into the medium,leading to the enhanced HA production to 249.85 mg/L,about 2.07-fold over the control.This is the first report on the regulation of Pseudomonas volatiles on fungal perylenequinone production.These findings could be helpful to understand the roles of bacterial volatiles in fruiting bodies and also provide new elicitation method using bacterial volatiles to stimulate fungal secondary metabolite production.

Characterization of Prealloyed Ti–6Al–4V Powders from EIGA and PREP Process and Mechanical Properties of HIPed Powder Compacts

Prealloyed Ti-6Al-4V powders were prepared by electrode induction melting gas atomization （EIGA） and plasma rotating electrode process （PREP） in this work. A comparative study of EIGA and PREP powders for hot isostatic pressing （HIping） compaction was conducted. Characterization of important technological parameters such as particle size distribution, powder surface morphology and flowability was carried out. Microstructure and mechanical properties of Ti- 6Al-4V powder compacts HIPed from EIGA and PREP powders were also investigated. The results showed that the EIGA powder has a finer average particle size and higher tap density, while the PREP powder has better flowability and less pores. Micropores can be observed in heat-treated EIGA powder compacts by X-ray tomography and the porosity was found to be about 0.02%. There are no micropores （≥4 μm） to be detected in heat-treated PREP powder compacts. Transgranular fracture mode as well as micropores contributes to the scatter in fatigue property of heat-treated PREP powder compacts. The respective advantages and disadvantages of both EIGA and PREP powders for producing Ti-based complex parts through HIping were also discussed.

Ultrafine VN nanodots induced generation of abundant cobalt single-atom active sites on nitrogen-doped carbon nanotube for efficient hydrogen evolution

Development of highly active and stable non-noble electrocatalysts with well-defined nanostructures is crucial for efficient hydrogen evolution reaction(HER). Herein, a novel three-dimensional(3D) selfsupported electrode consists of vanadium nitride(VN) nanodots and Co nanoparticles co-embedded and highly active single Co atoms anchored in N-doped carbon nanotubes supported on carbon cloth(VN-Co@CoSAs-NCNTs/CC) is fabricated via a one-step in situ nanoconfined pyrolysis strategy, which shows remarkable enhanced HER electrocatalytic activity in acidic medium. During pyrolysis, the formed VN nanodots induce the generation of atomic Co Nxsites in NCNTs, contributing to superior electrocatalytic activity. Experimental and density functional theory(DFT) calculation results reveal that the electrode has multiple accessible active sites, fast reaction kinetics, low charge/mass transfer resistances,high conductivity, as well as downshifted d-band center with a thermodynamically favorable hydrogen adsorption free energy(△G_(H·)), all of which greatly boost the HER performance. As a result, the VNCo@CoSAs-NCNTs/CC electrode displays superb catalytic performance toward HER with a low overpotential of 29 mV at 10 mA cm^(-2) in acidic medium, which could maintain for at least 60 h of stable performance. This work opens a facile avenue to explore low-cost, high performance, but inexpensive metals/nitrogen-doped carbon composite electrocatalysts for HER.

Breast non-mass-like lesions on contrast-enhanced ultrasonography: Feature analysis, breast image reporting and data system classification assessment

BACKGROUND Breast non-mass-like lesions(NMLs)account for 9.2%of all breast lesions.The specificity of the ultrasound diagnosis of NMLs is low,and it cannot be objectively classified according to the 5th Edition of the Breast Imaging Reporting and Data System(BI-RADS).Contrast-enhanced ultrasound(CEUS)can help to differentiate and classify breast lesions but there are few studies on NMLs alone.AIM To analyze the features of benign and malignant breast NMLs in grayscale ultrasonography(US),color Doppler flow imaging(CDFI)and CEUS,and to explore the efficacy of the combined diagnosis of NMLs and the effect of CEUS on the BI-RADS classification of NMLs.METHODS A total of 51 breast NMLs verified by pathology were analyzed in our hospital from January 2017 to April 2019.All lesions were examined by US,CDFI and CEUS,and their features from those examinations were analyzed.With pathology as the gold standard,binary logic regression was used to analyze the independent risk factors for malignant breast NMLs,and a regression equation was established to calculate the efficiency of combined diagnosis.Based on the regression equation,the combined diagnostic efficiency of US combined with CEUS(US+CEUS)was determined.The initial BI-RADS-US classification of NMLs was adjusted according to the independent risk factors identified by CEUS,and the diagnostic efficiency of CEUS combined with BI-RADS(CEUS+BI-RADS)was calculated based on the results.ROC curves were drawn to compare the diagnostic values of the three methods,including US,US+CEUS,and CEUS+BI-RADS,for benign and malignant NMLs.RESULTS Microcalcification,enhancement time,enhancement intensity,lesion scope,and peripheral blood vessels were significantly different between benign and malignant NMLs.Among these features,microcalcification,higher enhancement,and lesion scope were identified as independent risk factors for malignant breast NMLs.When US,US+CEUS,and CEUS+BI-RADS were used to identify the benign and malignant breast NMLs,their sensitivity rates were 82.6%,91.3%,and

Hepatocellular carcinoma(HCC),which makes up the majority of liver cancer,is induced by the infection of hepatitis B/C virus.Biomarkers are needed to facilitate the early detection of HCC,which is often diagnosed too late for effective therapy.The tRNA-derived small RNAs(tsRNAs)play vital roles in tumorigenesis and are stable in circulation.However,the diagnostic values and biological functions of circulating tsRNAs,especially for HCC,are still unknown.In this study,we first utilized RNA sequencing followed by quantitative reverse-transcription PCR to analyze tsRNA signatures in HCC serum.We identified tRF-Gln-TTG-006,which was remarkably upregulated in HCC serum(training cohort:24 HCC patients vs.24 healthy controls).In the validation stage,we found that tRF-Gln-TTG-006 signature could distinguish HCC cases from healthy subjects with high sensitivity(80.4%)and specificity(79.4%)even in the early stage(Stage I:sensitivity,79.0%;specificity,74.8%;155 healthy controls vs.153 HCC patients from two cohorts).Moreover,in vitro studies indicated that circulating tRF-Gln-TTG-006 was released from tumor cells,and its biological function was predicted by bioinformatics assay and validated by colony formation and apoptosis assays.In summary,our study demonstrated that serum tsRNA signature may serve as a novel biomarker of HCC.
