Colorectal cancer(CRC)is one of the most common human malignant diseases and the second leading cause of cancer-related deaths *** treatment of advanced CRC has improved significantly in recent *** the emergence of two targeted antibodies,cetuximab(Erbitux),an anti-epidermal growth factor receptor monoclonal antibody and bevacizumab(Avastin),a vascular endothelial growth factor monoclonal antibody,the treatment of metastatic CRC has entered the era of personalized *** and prognostic biomarkers have,and will continue to,facilitate the selection of suitable patients and the personalization of treatment for metastatic CRC(mCRC).In this review,we will focus primarily on the important progresses made in the personalized treatment of mCRC and discuss the potentially novel predictive and prognostic biomarkers for improved selection of patients for anti-cancer treatment in the future.

Cancer is the leading cause of death in China and depicting the cancer pattern of China would provide basic knowhows on how to tackle it more *** this study we have reviewed several reports of cancer burden,including the Global cancer statistics 2018 and Cancer statistics in China,2015,along with the GLOBCAN 2018 online database,to investigate the differences of cancer patterns between China,the United States(USA)and the United Kingdom(UK).An estimated 4.3 million new cancer cases and 2.9 million new cancer deaths occurred in China in *** to the USA and UK,China has lower cancer incidence but a 30%and 40%higher cancer mortality than the UK and USA,among which 36.4%of the cancer-related deaths were from the digestive tract cancers(stomach,liver,and esophagus cancer)and have relatively poorer *** comparison,the digestive cancer deaths only took up≤5%of the total cancer deaths in either USA or *** reasons for the higher mortality in China may be the low rate of early-stage cancers at diagnosis and non-uniformed clinical cancer treatment strategies performed by different *** is undergoing the cancer transition stage where the cancer spectrum is changing from developing country to developed country,with a rapidly increase cancer burden of colorectal,prostate,female breast cancers in addition to a high occurrence of infection-related and digestive *** incidence of westernized lifestyle-related cancers in China(*** cancer,prostate,bladder cancer)has risen but the incidence of the digestive cancers has decreased from 2000 to *** estimated 40%of the risk factors can be attributed to environmental and lifestyle factors either in China or other developed *** smoking is the single most important carcinogenic risk factor in China,contributing to~24.5%of cancers in *** infection is another important preventable cancer contributor which is responsible for~17%of *** prevent

Patients with high tumor mutational burden(TMB)levels do not consistently respond to immune checkpoint inhibitors(ICIs),possibly because a high TMB level does not necessarily result in adequate infiltration of CD8^(+)T *** bulk ribonucleic acid sequencing(RNA-seq)data from 9311 tumor samples across 30 cancer types,we developed a novel tool called the modulator of TMB-associated immune infiltration(MOTIF),which comprises genes that can determine the extent of CD8^(+)T cell infiltration prompted by a certain TMB *** confirmed that MOTIF can accurately reflect the integrity and defects of the cancer-immunity *** analyzing 84 human single-cell RNA-seq datasets from 32 types of solid tumors,we revealed that MOTIF can provide insights into the diverse roles of various cell types in the modulation of CD8^(+)T cell *** pretreatment RNA-seq data from 13 ICI-treated cohorts,we validated the use of MOTIF in predicting CD8^(+)T cell infiltration and ICI *** the components of MOTIF,we identified EMC3 as a negative regulator of CD8^(+)T cell infiltration,which was validated via in vivo ***,MOTIF provided guidance for the potential combinations of programmed death 1 blockade with certain immunostimulatory drugs to facilitate CD8^(+)T cell infiltration and improve ICI efficacy.

Significant progress has been made in wet adhesives for low salinity water,but exploration of general ionic adhesives for natural seawater is less developed because the high salinity could weaken interfacial bonding and shields electrostatic interactions,resulting in adhesion ***,the design of adhesives for natural seawater represents challenges less ***,a cationic polyelectrolyte(PECHIA)containing imidazolacetonitrile unit was explored to prepare adhesives enabled by natural *** combining the ion shielding effect with the“cation-dipole”interactions between PECHIA chains,aqueous solution of the PECHIA underwent coacervation and self-crosslinking in natural seawater,allowing for underwater adhesion to various substrates in *** instantaneous lap-shear and tensile adhesion strengths are 47 and 119 kPa,respectively,while the cured adhesive shows~739 k Pa tensile adhesion in natural *** design of PECHIA enables wet adhesives viable for applications in the diversified scenarios of natural seawater.
