The existence of multigenic families in the mevalonate pathway suggests divergent functional roles for pathway components involved in the biosynthesis of plant sterols. Squalene epoxidases （SQEs） are key components of this pathway, and Squalene Epoxidase 1 （SQE1） has been identified as a fundamental enzyme in this biosynthetic step. In the present work, we extended the characterization of the remaining SQE family members, phylogenetically resolving between true SQEs and a subfamily of SQE-like proteins that is exclusive to Brassicaceae. Functional characterization of true SQE family members, Squalene Epox- idase 2 （SQE2） and Squalene Epoxidase 3 （SQE3）, indicates that SQE3, but not SQE2, contributes to the bulk SQE activity in Arabidopsis, with sqe3-1 mutants accumulating squalene and displaying sensitivity to ter- binafine. We genetically demonstrated that SQE3 seems to play a particularly significant role in embryo development. Also, SQE1 and SQE3 both localize in the endoplasmic reticulum, and SQE3 can functionally complement SQEI. Thus, SQE1 and SQE3 seem to be two functionally unequal redundant genes in the pro- motion of plant SQE activity in Arabidopsis.

Effect of climate and altitude on plant community composition and richness in Brazilian inselbergs

Plant communities are shaped by multiple factors along environmental gradients;however,studies are limited on how environmental filtering drives community composition and species richness on tropical *** evaluate the influence of altitude and climatic variables related to temperature and precipitation on plant community composition and species richness on Brazilian *** assume as a premise that both climate and altitude would induce changes on plant community composition and species richness at the local *** used plant inventory data from 370 sampling units across four inselberg sites in the Atlantic Forest of Espírito Santo State,south-eastern *** tested the univariate and multivariate effects of altitude and climate variables on community composition and species richness with multiple *** in species richness between inselbergs were evaluated using sample-based data to estimate rarefaction and extrapolation *** addition,differences in species composition and taxonomic beta diversity were examined via novel frequency-based metrics.A contrasting climate pattern was observed between the inselberg sites,with south sites being wet compared to the dry conditions found in northern *** richness by rarefaction showed a similar pattern within regional sites;however,there were marked differences between *** richness and beta diversity showed significant differences among sites,with higher values in southern sites than in northern *** a multi-model comparison between inselberg sites,altitude significantly influenced community composition and species richness and explained more variance than climate *** finding suggested that climate could act to some extent on these tropical inselbergs;however,altitude was a better predictor of plant community composition and species richness at the local level.

Mechanotransduction via the coordinated actions of integrins,PI3K signaling and Connexin hemichannels

Mechanical loading opens connexin 43(Cx43)hemichannels(HCs),leading to the release of bone anabolic molecules,such as prostaglandins,from mechanosensitive osteocytes,which is essential for bone formation and ***,the mechanotransduction mechanism that activates HCs remains ***,we report a unique pathway by which mechanical signals are effectively transferred between integrin molecules located in different regions of the cell,resulting in HC *** integrinα5 andαV were activated upon mechanical stimulation via either fluid dropping or flow shear stress(FSS).Inhibition of integrinαV activation or ablation of integrinα5 prevented HC opening on the cell body when dendrites were mechanically stimulated,suggesting mechanical transmission from the dendritic integrinαV toα5 in the cell body during HC *** addition,HC function was compromised in vivo,as determined by utilizing an antibody blockingαV activation andα5-deficient osteocyte-specific knockout ***,inhibition of integrinαV activation,but not that ofα5,attenuated activation of the phosphoinositide 3-kinase(PI3K)-protein kinase B(AKT)signaling pathway upon mechanical loading,and the inhibition of PI3K/AKT activation blocked integrinα5 activation and HC ***,HC opening was blocked only by an anti-integrinαV antibody at low but not high FSS levels,suggesting that dendriticαV is a more sensitive mechanosensor thanα5 for activating ***,these results reveal a new molecular mechanism of mechanotransduction involving the coordinated actions of integrins and PI3K/AKT in osteocytic dendritic processes and cell bodies that leads to HC opening and the release of key bone anabolic factors.

Juvenile hemochromatosis:HAMP mutation and severe iron overload treated with phlebotomies and deferasirox

Juvenile hemochromatosis(JH) is a rare condition classified as an autosomal recessive disorder that leads to severe iron absorption. JH usually affects people under the age of 30 and presents symptoms such as chronic liver damage,hypogonadotropic hypogonadism,cardiac diseases and endocrine dysfunctions. The present case reports a 29-year-old Brazilian woman with JH condition due to HAMP mutation(g.47G>A),treated with phlebotomies and deferasirox. She presented symptoms such as weakness,skin hyperpigmentation,joint pain in the shoulders and hands and amenorrhea. First laboratory tests showed altered biochemical parameters [serum ferritin(SF): 5696 ng/mL,transferrin saturation(TS): 85%]. After sessions of phlebotomies(450 mL every 15 d),the patient presented partial symptomatic improvements and biochemical parameters(SF: 1000 ng/mL,Hb: 11 g/dL). One year later,deferasirox(15 mg/kg per day) was introduced to the treatment,and the patient showed total symptomatic improvement,with significant clearing of the skin,SF: 169 ng/mL,and TS: 50%. Furthermore,after the combined deferasirox-phlebotomy therapy,magnetic resonance imaging measurements revealed normalized level for liver iron(30 μmol/g; reference value < 36 μmol/g). In conclusion,combined deferasirox-phlebotomy treatment was able to normalize iron levels and improve symptoms.

Serrated pathway in colorectal carcinogenesis

Serrated adenocarcinoma is a recently described subset of colorectal cancer(CRC),which account for about10%of all CRCs and follows an alternative pathway in which serrated polyps replace the traditional adenoma as the precursor lesion to *** polyps form a heterogeneous group of colorectal lesions that includes hyperplastic polyps(HPs),sessile serrated adenoma(SSA),traditional serrated adenoma(TSA)and mixed *** are the most common serrated polyp followed by SSA and *** distinct histogenesis is believed to have a major influence in prevention strategies,patient prognosis and therapeutic ***,serrated polyps exhibited also a distinct pattern,with KRAS and BRAF having an important contribution to its *** other molecular changes that have been implicated in the serrated pathway include microsatellite instability and the CpG island methylator *** the present review we will address the current knowledge of serrated polyps,clinical pathological features and will update the most recent findings of its molecular *** understanding of their biology and malignancy potential is imperative to implement a surveillance approach in order to prevent colorectal cancer development.

Biological oyster shell waste enhances polyphenylene sulfide composites and endows them with antibacterial properties

To date,there is no research that deals with biological waste as fillers in polyphenylene sulfide(PPS).In this study,oyster shells were recycled and treated to prepare thermally-treated oyster shells(TOS),which were used as PPS fillers to make new bio-based antibacterial composite *** effect of varying the content of TOS was studied by means of structure and performance ***/TOS composites were demonstrated to have an antibacterial effect on the growth of E coli and *** analysis showed that when the TOS content was≥30%and 40%,the composite materials had an apparent inhibition *** analysis showed that the antibacterial activity increased with the TOS *** transform infrared spectroscopy indicated the formation of hydrogen bonds between the molecular chains of TOS and PPS and the occurrence of a coordination *** 10%TOS,the composite tensile strength reached a maximum value of 72.5 MPa,which is 9.65%higher than that of pure *** trend of bending properties is the same as that of tensile properties,showing that the maximum property was reached for the composite with 10%*** the same time,the crystallinity and contact angle were the highest,and the permeability coefficient was the *** fatigue test results indicated that for the composite with 10%TOS,the tensile strength was 23%lower than static tensile strength,and the yield strength was 10%lower than the static yield *** results of the study showed that TOS not only could reduce the cost of PPS,but also could impart antibacterial properties and enhance the mechanical and,barrier properties,the thermostability,as well as the crystallinity.

Osteocytes regulate bone anabolic response to mechanical loading in male mice via activation of integrinα5

Physical mechanical stimulation can maintain and even increase bone ***,we report an important role of osteocytic integrinα5 in regulating the anabolic response of bone to mechanical loading using an Itga5 conditional gene knockout(cKO)mouse ***α5 gene deletion increased apoptotic osteocytes and reduced cortical anabolic responses to tibial compression including decreased endosteal osteoblasts and bone formation,and increased endosteal osteoclasts and bone resorption,contributing to the decreased bone area fraction and biomechanical properties,leading to an enlarged bone marrow area in cKO *** disruption of anabolic responses to mechanical loading was also detected in cKO trabecular ***,integrinα5 deficiency impeded load-induced Cx43 hemichannel opening,and production and release of PGE2,an anabolic factor,resulting in attenuated effects of the loading on catabolic sclerostin(SOST)reduction and anabolicβ-catenin ***,this study shows an indispensable role of integrinα5 in osteocytes in the anabolic action of mechanical loading on skeletal tissue through activation of hemichannels and PGE2-evoked gene ***α5 could act as a potential new therapeutic target for bone loss,especially in the elderly population with impeded mechanical sensitivity.
