Background:Buyang Huanwu decoction(BHD)is a traditional Chinese medicine herbal formula used for treating hypertension,particularly in the later stages of hypertension when it is associated with intracerebral hemorrhage.This study aims to investigate the treatment mechanism of BHD to provide a basis for its clinical application in hypertension treatment.Methods:Network pharmacology analysis and cell culture experiments were performed to explore the potential proteins and mechanisms of action of BHD against hypertension.Bioactive compounds related to BHD were screened,and relevant targets associated with hypertension and BHD were retrieved.Molecular docking technology was used to identify the effective signaling pathway based on the Kyoto Encyclopedia of Genes and Genomes and protein-protein interaction network cores.Lastly,the effects and mechanisms of BHD on salt-sensitive hypertensive endothelial cells were investigated.Results:Ninety-three potential therapeutic targets for BHD and salt-sensitive hypertension were found to be closely associated with the PI3K/Akt/eNOS signaling pathway and oxidative stress.Cell experiments further indicated the pivotal role of endothelial cells in hypertension,and validation analysis showed that BHD significantly preserved cell morphology,suppressed oxidative stress reactions,activated the PI3K/Akt/eNOS signaling pathways,preserved normal endothelial cell function,and reduced cell apoptosis.Conclusion:BHD effectively activates the PI3K/Akt/VEGF signaling pathway,attenuates oxidative stress-induced injury in endothelial cells exposed to high salt levels,and mitigates apoptosis,supporting the use of traditional Chinese medicine BHD in the treatment of salt-sensitive hypertension.

Limitation of the Application of Heat Tracer in Investigating Groundwater and River Interactions under Dynamic Flow Conditions

The groundwater (GW) in the floodplain riparian area frequently interacts intensely with surface water (SW). Heat as a tracer is one of the hot research fields in investigating GW-SW interactions， and analytical approaches have been proposed for the calculation of exchange flow velocity. However， few studies have considered the effects of very dynamic flow conditions and monitoring instrumentation on the calculation with field measured data. Herein， taking the middle reaches of the Heihe River as the study area， different types of monitoring wells were constructed under the riverbed and near the river， and multiple methods (Darcy’s law， heat tracing， and isotopic mixing methods) were employed to trace the exchanges between the river and groundwater. The results indicate that different methods demonstrate diverse information with obvious unevenly distributed flux along the vertical direction. And the combination of multiple methods has an important role in studying the interaction between GW and SW. Fully screened wells produce intraborehole flow and disturb the heat transport， which is relevant to flow velocity， and further affects the temperature distribution， impacting the temperature-based flow velocity calculation. Dynamic flow conditions aggravate riverbed sediment disturbances， e.g.， scour and deposition， and additionally affect the interaction and monitoring data.

Identification of (-)-bornyl diphosphate synthase from Blumea balsamifera and its application for (-)-borneol biosynthesis in Saccharomyces cerevisiae

Borneol is a precious monoterpenoid with two chiral structures,(-)-borneol and(+)-borneol.Bornyl diphosphate synthase is the key enzyme in the borneol biosynthesis pathway.many(+)-bornyl diphosphate synthases have been reported,but no(-)-bornyl diphosphate synthases have been identified.Blumea balsamifera leaves are rich in borneol,almost all of which is(-)-borneol.In this study,we identified a high-efficiency(-)-bornyl diphosphate synthase(BbTPS3)from B.balsamifera that converts geranyl diphosphate(GPP)to(-)-bornyl diphosphate,which is then converted to(-)-borneol after dephosphorylation in vitro.BbTPS3 exhibited a K m value of 4.93±1.38μM for GPP,and the corresponding k cat value was 1.49 s−1.Multiple strategies were applied to obtain a high-yielding(-)-borneol producing yeast strain.A codon-optimized BbTPS3 protein was introduced into the GPP high-yield strain MD,and the resulting MD-B1 strain produced 1.24 mg⋅L^(-1)(-)-borneol.After truncating the N-terminus of BbTPS3 and adding a Kozak sequence,the(-)-borneol yield was further improved by 4-fold to 4.87 mg⋅L^(-1).Moreover,the(-)-borneol yield was improved by expressing the fusion protein module of ERG20 F96W-N127W-YRSQI-t14-BbTPS3K2,resulting in a final yield of 12.68 mg⋅L^(-1) in shake flasks and 148.59 mg⋅L^(-1) in a 5-L bioreactor.This work is the first reported attempt to produce(-)-borneol by microbial fermentation.

受到成像设备、传输环境等因素的影响,导致激光遥感图像存在明显的模糊现象,为恢复激光遥感图像质量提出基于视觉传达的模糊激光遥感图像重建方法。利用视觉传达技术获取模糊激光遥感图像,从纹理、统计分布、频率等方面提取图像特征,以此作为图像重构规律,通过图像配准、去模糊、噪声等步骤,得出模糊激光遥感图像重建结果,以可视化的形式输出。通过测试实验得出结论:与传统重建方法相比,优化方法得出重建图像峰值信噪比最高达到了38 dB,结构相似性最高达到了0.94,表明优化方法在重建质量方面具有明显优势。

Authenticated Digital Currency Redaction with Stronger Privacy and Usability

With the promotion of digital currency,how to effectively solve the authenticity,privacy and usability of digital currency issuance has been a key problem.Redactable signature scheme(RSS)can provide the verification of the integrity and source of the generated sub-documents and solve the privacy problem in digital currency by removing blocks from the signed documents.Unfortunately,it has not realized the consolidation of signed documents,which can not solve the problem of merging two digital currencies.Now,we introduce the concept of weight based on the threshold secret sharing scheme(TSSS)and present a redactable signature scheme with merge algorithm(RSS-ma)using the quasi-commutative accumulator.Our scheme can reduce the communication overhead by utilizing the merge algorithm when transmitting multiple digital currency signatures.Furthermore,this can effectively hide the scale of users’private monetary assets and the number of transactions between users.While meeting the three properties of digital currency issuance,in order to ensure the availability of digital currency after redacting,editors shall not remove the relevant identification information block form digital currency.Finally,our security proof and the analysis of efficiency show that RSS-ma greatly improves the communication and computation efficiency when transmitting multiple signatures.

RNA-binding proteins(RBPs)-RNA networks have contributed to cancer development.Circular RNAs(circRNAs)are considered as protein recruiters;nevertheless,the patterns of circRNA-protein interactions in colorectal cancer(CRC)are still lacking.Processing bodies(PBs)formed through liquid-liquid phase separation(LLPS)are membrane-less organelles(MLOs)consisting of RBPs and RNA.Previous evidence suggests a connection between PBs dynamics and cancer progression.Despite the increasingly acknowledged crucial role of RBPs and RNA in the accumulation and maintenance of MLOs,there remains a lack of specific research on the interactions between PBs-related RBPs and circRNAs in CRC.Herein,we identify that MEX-3 RNA binding family member A(MEX3A),frequently upregulated in CRC tissues,predicts poorer patient survival.Elevated MEX3A accelerates malignance and inhibits autophagy of CRC cells.Importantly,MEX3A undergoes intrinsically disordered regions(IDRs)-dependent LLPS in the cytoplasm.Specifically,circMPP6 acts as a scaffold to facilitate the interaction between MEX3A and PBs proteins.The MEX3A/circMPP6 complex modulates PBs dynamic and promotes UPF-mediated phosphodiesterase 5A(PDE5A)mRNA degradation,consequently leading to the aggressive properties of CRC cells.Clinically,CRC patients exhibiting high MEX3A expression and low PDE5A expression have the poorest overall survival.Our findings reveal a collaboration between MEX3A and circMPP6 in the regulation of mRNA decay through triggering the PBs aggregation,which provides prognostic markers and/or therapeutic targets for CRC.

本文以苯基磷酰二氯、香草醛和4-氨基苯酚为主要原料,合成了一种含磷席夫碱阻燃剂(PH-VAN-AP),并采用红外光谱、核磁共振氢谱和核磁共振磷谱证明其结构符合预期。将DOPO与PH-VAN-AP作为复配阻燃剂PH/DOPO加入环氧树脂(EP)中,通过垂直燃烧(UL-94)、极限氧指数(LOI)和锥形量热测试(CCT)对其阻燃性能进行了表征,并利用热失重和差示扫描量热分析了环氧树脂的热稳定性、热降解过程及玻璃化转变温度(T_(g))。结果表明,PH/DOPO兼有气相和凝聚相2种阻燃机理,能有效提高EP的燃烧性能。具体而言,当仅添加PH/DOPO至体系的3%(质量分数)时,顺利通过了UL-94测试并达到V-0等级,其LOI由24.7%提高至34.8%。此外,峰值热释放速率(PHRR)和总热释放(THR)分别降低了257 kW/m^(2)和8 MJ/m^(2),材料的残留量提高了6.8%。添加3%的PH/DOPO,环氧树脂的Tg由149.5℃降至148.1℃,变化不大,表明PH-VAN-AP一定程度上改善了由于DOPO加入EP后导致T_(g)下降的问题。

抗战时期,电化教育被视作中国宣传抗战建国和启发民智的重要手段。1942年5月,滇缅公路被截断,电化教育转入顿挫状态,袖珍片放映逐渐取代电影放映,成为电化教育中最为重要的媒介分发系统,并重塑了电化教育的实践网络。研究发现袖珍放映机在大后方的应用和普及,一方面是与电力、交通、邮政等基础设施相互交织和共同演进;另一方面,人和社会组织是其技术系统的重要组成部分,通过袖珍放映机转译其行动方案,对战争宣传和教育普及具有举足轻重的作用。

BACKGROUND SmaRCA4 is a component of chromatin remodeling of SWItch/sucrose-nonfermenting(SWI/SNF)complexes and plays an essential role in oncogenesis.SmaRCA4-deficient malignancies arising from the gastrointestinal tract are rare and have a poor prognosis.There is no standard treatment for advanced and undifferentiated SmaRCA4-deficient duodenal malignancies.Programmed death 1(PD-1)antibodies,known as immune checkpoint inhibitor antibodies,potentially play a role in treating gastrointestinal tract malignancies.CASE SUMmaRY We present two patients with SmaRCA4 deficiency and TP53 gene mutation in advanced undifferentiated carcinomas of the duodenum.For both patients,SmaRCA4 deficiency was confirmed by immunohistochemical staining for the BRG1 protein,while TP53 gene mutations were observed via next-generation sequencing.Both patients were administered chemotherapy in combination with an anti-PD-1 antibody.The two patients exhibited completely different responses to treatment and had different prognoses.Case 1 experienced rapid progression after PD-1 infusion and chemotherapy,case 2 experienced a remarkable response after treatment,and the progression-free survival was more than 6 months.CONCLUSION This study described our clinical and pathological observations of SmaRCA4-deficient advanced undifferentiated carcinoma of the duodenum.PD-1 combined with chemotherapy showed a certain efficacy in select patients,providing options for treating these highly malignant tumors.Patients with liver metastases had a worse prognosis than did those with only lymph node metastasis.
