Modeling and measuring glucose diffusion and consumption by colorectal cancer spheroids in hanging drops using integrated biosensors

As 3D in vitro tissue models become more pervasive,their built-in nutrient,metabolite,compound,and waste gradients increase biological relevance at the cost of analysis *** these gradients and the resulting metabolic heterogeneity requires invasive and time-consuming *** alternative is using electrochemical biosensors and measuring concentrations around the tissue model to obtain size-dependent metabolism *** our hanging-dropintegrated enzymatic glucose biosensors,we conducted current measurements within hanging-drop compartments hosting spheroids formed from the human colorectal carcinoma cell line *** developed a physics-based mathematical model of analyte consumption and transport,considering(1)diffusion and enzymatic conversion of glucose to form hydrogen peroxide(H_(2)O_(2))by the glucose-oxidase-based hydrogel functionalization of our biosensors at the microscale;(2)H_(2)O_(2)oxidation at the electrode surface,leading to amperometric H_(2)O_(2)readout;(3)glucose diffusion and glucose consumption by cancer cells in a spherical tissue model at the microscale;(4)glucose and H_(2)O_(2)transport in our hangingdrop compartments at the macroscale;and(5)solvent evaporation,leading to glucose and H_(2)O_(2)*** model relates the measured currents to the glucose concentrations generating the *** low limit of detection of our biosensors(0.4±0.1μM),combined with our current-fitting method,enabled us to reveal glucose dynamics within our *** measuring glucose dynamics in hanging-drop compartments populated by cancer spheroids of various sizes,we could infer glucose distributions within the spheroid,which will help translate in vitro 3D tissue model results to in vivo.

Bats and marsupials as indicators of endemism in the Yungas forest of Argentina

Several studies have characterized the Yungas as a separate biogeographic unit, mainly based in floristic components. However, these characterizations were mainly qualitative and did not include faunal groups. The Yungas have been assumed as a region with rich floral and faunal diversity, but without testing how well they are described by animal distributions. Our study consists of a formal analysis of endemism based on distribution of small mammals in the southernmost portion of the Yungas. This area is biogeographically very interesting because the Yungas are comprised of discontinuous fragments of forests that extend into temperate arid and semiarid habitats. As a first approximation, we contrasted a group of volant species （bats） versus a group of non-volant species （marsupials）. Our results show that small mammals are efficient indicators of endemism in Yungas. Eighty percent of the species of small marsupials included in the analysis supported the identified areas as being zones of endemism. Regarding bats, almost 55 percent of the species supported a designation of endemism. The results also show that the areas we considered are congruent with the botanical definition of the Yungas of northwestern Argentina and their subdivisions, an assumption that had not been previously tested with a formal quantitative method. We also found that non-volant species are better indicators of endemism than volant ones at regional scales, but volant species are better indicators than was previously thought.

Rambutan growing in the state of Chiapas has brought about a demand for new plantations;however, this species has several biotic and abiotic factors that limit its production. These factors include phytosanitary problems, deficient agronomical handling, and production alternancy. Because of this, the objective of the present study was to evaluate the effect of the application of agronomical practices such as pruning, ringing, water stress, and their interaction on the rambutan alternancy. The experiment was carried out in the “la Chinita” commercial orchard located on KM. 4.5 the Huehuetan Station, municipality of Huehuetan, Chiapas, Mexico. We used 48 rambutan trees in production with an age of 14 years. Two production cycles were evaluated. The experiment was established under a divided plot experimental design in random complete blocks with a factorial arrangement and six replicates;each tree being a replicate. The study factors were: water stress, pruning, girdling, irrigation (control), and their combinations, resulting in eight treatments. The evaluation was done during flowering, fruit set, and harvest. In flowering, the treatments with permanent irrigation in both evaluated cycles (2010-2011 and 2011-2012) manifested a production alternancy behavior, with an average 36.4% flowering in the first evaluation cycle and increased to 82.1% in the second cycle. The treatments with water stress kept a proportional flowering during the first and second cycles of 97.9% and 95.3%, respectively. The water stress influenced the bunch weight (PR) variable fruit set with bigger fruits and higher bunch weight than the irrigated treatments. In production, pruning and girdling together with water deficit had no alternancy in both production cycles.

The objective of the present work is modeling the mobility habitus in the public transport system with low Co2 emission mechanics in the center of *** this sense,a documentary work was carried out with a review of sources indexed to national repositories,using the Delphi technique for the content analysis and the specification of the model.A study was conducted with a selection of 100 students to establish the reliability,linearity,sphericity,adequacy,and validity of an instrument from which a relationship of dependence between income and mobility habitus was *** limits of the search,selection,and processing of information are recognized,and it is recommended to extend the research to international repositories,as well as to employ a more sophisticated technique.
