Comparison of bioavailability and transporters gene expression of four iron fortificants added to infant cereals

Iron deficiency is one of the most important nutritional disorders around the world. To reduce this health problem in infants, most countries have developed different iron fortification programs as a long-term strategy. However, it is necessary to select the best iron source to get the highest bioavailability with the lowest impact on the organoleptic properties of the foods. The aim of this research was to assess the in vitro bioavailability and intestinal cellular transport of four iron sources (electrolytic iron: EI, ferrous fumarate: FF, a micronized and dispersible ferric pyrophosphate: MDFP, and ferrous sulphate heptahydrated: FsH) used to fortify infant cereals. Fortified cereals were in vitro digested and soluble fractions were then incubated in Caco-2 cells to study iron retention, transport and uptake. Moreover, mRnA expression of iron transporters and ferritin formation were also measured in cells incubated with the iron standard salts. Iron solubility, uptake percentage, transport and uptake efficiencies by the Caco-2 cells were significantly higher in cereals fortified with FsH than with EI, obtaining intermediate values for the other two iron fortificants. MDFP induced higher expression of iron transporters Zip-14 and HEPH than other iron sources. FsH promoted the highest iron storage protein ferritin, followed by FF and MDFP, while EI presented the lowest in vitro bioavailability due to its low solubility. This work demonstrated that iron absorption, storage, and gene expression of transporters depend on the source of iron. These are important challenges to be taken into consideration when assessing the potential of iron fortificants.

Modeling and measuring glucose diffusion and consumption by colorectal cancer spheroids in hanging drops using integrated biosensors

As 3D in vitro tissue models become more pervasive,their built-in nutrient,metabolite,compound,and waste gradients increase biological relevance at the cost of analysis *** these gradients and the resulting metabolic heterogeneity requires invasive and time-consuming *** alternative is using electrochemical biosensors and measuring concentrations around the tissue model to obtain size-dependent metabolism *** our hanging-dropintegrated enzymatic glucose biosensors,we conducted current measurements within hanging-drop compartments hosting spheroids formed from the human colorectal carcinoma cell line *** developed a physics-based mathematical model of analyte consumption and transport,considering(1)diffusion and enzymatic conversion of glucose to form hydrogen peroxide(H_(2)O_(2))by the glucose-oxidase-based hydrogel functionalization of our biosensors at the microscale;(2)H_(2)O_(2)oxidation at the electrode surface,leading to amperometric H_(2)O_(2)readout;(3)glucose diffusion and glucose consumption by cancer cells in a spherical tissue model at the microscale;(4)glucose and H_(2)O_(2)transport in our hangingdrop compartments at the macroscale;and(5)solvent evaporation,leading to glucose and H_(2)O_(2)*** model relates the measured currents to the glucose concentrations generating the *** low limit of detection of our biosensors(0.4±0.1μM),combined with our current-fitting method,enabled us to reveal glucose dynamics within our *** measuring glucose dynamics in hanging-drop compartments populated by cancer spheroids of various sizes,we could infer glucose distributions within the spheroid,which will help translate in vitro 3D tissue model results to in vivo.

Aims sexes of dimorphic species often differ in ecophysiological traits and display spatial *** differences have been interpreted as an evolved response of the sexes to meet the specific resource demands associated with *** may differ not only in the amount of resources allocated to reproduction but also in the tim-ing of allocation to *** this study,we hypothesize that as a consequence of their specific resource demands for reproduction,the sexes of the dune plant Honckenya peploides differ in terms of tempo-ral patterns of water use efficiency and nitrogen use and *** Water use efficiency,as inferred from leaf carbon isotope discrimi-nation(Δ^(13)C),nitrogen use,estimated by leaf nitrogen isotope com-position(δ^(15)n),and the foliar carbon and nitrogen contents were measured in males and females at three different points in *** Findings Females had greater water use efficiency than males,regardless of *** ratio of n^(15) to n^(14) did not change with time in males,but significantly decreased in August for *** total n content in the leaf tissues of females decreased as the season progressed,while in males a decrease was only found from April to June and then it remained constant from June to August.A similar pattern,but reversed,was followed by the foliar C/n ***,nega-tive relationships between leafΔ^(13)C and n content were found at all times for males and only at the end of the season for ***,our hypothesis that sex-specific patterns of nitrogen and water use efficiency will depend on time was ***,our results highlight the importance of including time in studies of sex-ual dimorphism,and also the role that physiological specialization plays in meeting the specific demands associated with reproduction.

next-generation sequencing （nGs） technologies allow the cost-effective sequencing of whole genomes and have expanded the scope of genomics to novel applications, such as the genome-wide characterization of intraspecific polymorphisms and the rapid mapping and identification of point mutations. next-generation sequencing platforms, such as the Illumina Hiseq2ooo platform, are now commercially available at affordable prices and routinely produce an enormous amount of sequence data, but their wide use is often hindered by a lack of knowledge on how to manipulateand process the information produced. In this review, we focus on the strategies that are available to geneticists who wish to incorporate these novel approaches into their research but who are not familiar with the necessary bioinformatic concepts and computational tools. In particular, we comprehensively summarize case studies where the use of nGs technologies has led to the identification of point mutations, a strategy that has been dubbed ＂mapping-by-sequencing＇, and review examples from plants and other model species such as Caenorhabditis elegans, saccharomyces cerevisiae, and Drosophila mela- nogaster. As these technologies are becoming cheaper and more powerful, their use is also expanding to allow mutation identification in species with larger genomes, such as many crop plants.

Aims the limited water supply in the Mediterranean Basin will be exacerbated in the immediate future as result of severe drought ***,the study of the intraspecific variability in functional traits of plant species is important,because such variability modulates species’***,we aimed to analyse the variability in plasticity of physiological and structural traits of Arbutus unedo *** response to water availability and to determine whether the levels of phenotypic plasticity varied in plants of different *** We kept two sets of plants under contrasted water regimes(high water and low water)and calculated a phenotypic plasticity index to determine the plastic response to water *** compared the response among kind of traits and *** also per-formed analysis of variance to assess the trait and provenance *** Findings We found that biomass and key physiological traits decrease under drought conditions,while the water use efficiency *** differed between groups of traits,being higher in structural and gas exchange parameters,but the provenance did not account for signifi-cant variation in *** has evolved a‘good saver’strat-egy,involving decreasing growth and water use-when this resource is unavailable-together with the dissipation of excess of radiation that it cannot *** homogeneous response between provenances may be the result of a canalization *** strategy,based on a high degree of plasticity,is a probable determining factor in the success of the species over its wide range of environmental conditions and will be critical to withstand the ongoing environmental changes.

Electromagnetic-Energy Flow in Anisotropic Metamaterials: The Proper Choice of Poynting’s Vector

We study the controversy about the proper determination of the electromagnetic energy-flux field in anisotropic materials, which has been revived due to the relatively recent experiments on negative refraction in metamaterials. Rather than analyzing energy-balance arguments, we use a pragmatic approach inspired by geometrical optics, and compare the predictions on angles of refraction at a flat interface of two possible choices on the energy flux: and . We carry out this comparison for a monochromatic Gaussian beam propagating in an anisotropic non-dissipative anisotropic metamaterial, in which the spatial localization of the electromagnetic field allows a more natural assignment of directions, in contrast to the usual study of plane waves. We compare our approach with the formalism of geometrical optics, which we generalize and analyze numerically the consequences of either choice.

scheduling multi-tenant cloud workflow tasks with resource reliability

Resource reliability is crucial in scheduling workflow instances for different tenants. Both cloud resource reliability and precedence constraints in workflows bring about great challenges for these kinds of scheduling problems. In this paper, we construct a hybrid resource reliability model which is adaptively evaluated in every time window. The objective is to optimize the Qos(quality of service) for tenants which is measured by the introduced AIsE(all instance success entropy) index. A scheduling algorithmic framework is proposed for the studied workflows which consider cloud resource reliability. Deadline and budget division(BD) methods are presented to divide deadlines and budgets of instances into those of tasks. A tenant sequence method is developed to determine the order of tenants. A task allocation strategy is investigated to schedule tasks that are ready to appropriate available resources. Parameters and algorithm component candidates are statistically calibrated over a comprehensive set of random instances using the analysis of variance technique. The performance of the proposed algorithm is also evaluated in practical instances.

Gut microbiota-related complications in cirrhosis

Gut microbiota plays an important role in cirrhosis. The liver is constantly challenged with commensal bacteria and their products arriving through the portal vein in the so-called gut-liver axis. Bacterial translocation from the intestinal lumen through the intestinal wall and to mesenteric lymph nodes is facilitated by intestinal bacterial overgrowth, impairment in the permeability of the intestinal mucosal barrier, and deficiencies in local host immune defences. Deranged clearance of endogenous bacteria from portal and systemic circulation turns the gut into the major source of bacterialrelated complications. liver function may therefore be affected by alterations in the composition of the intestinal microbiota and a role for commensal flora has been evidenced in the pathogenesis of several complications arising in end-stage liver disease such as hepatic encephalopathy, splanchnic arterial vasodilatation and spontaneous bacterial peritonitis. The use of antibioticsis the main therapeutic pipeline in the management of these bacteria-related ***,other strategies aimed at preserving intestinal homeostasis through the use of pre-,pro-or symbiotic formulations are being studied in the last *** this review,the role of intestinal microbiota in the development of the most frequent complications arising in cirrhosis and the different clinical and experimental studies conducted to prevent or improve these complications by modifying the gut microbiota composition are summarized.

Polyarteritis nodosa(PAn) is one of the systemic vasculitis that affects the media wall of arteries of small and medium diameter. Diagnosis proves difficult due to the unspecific symptoms that dominate the clinical profile. liver involvement is very diverse, ranging from the development of cirrhotic liver disease to acute abdomen presentation that requires surgery because of liver rupture. The management of these patients requires an expert multidisciplinary team. There are several cases in the literature that describe a sudden liver rupture as the first manifestation of a PAn. In this paper we present the case of a 75 years old patient without any previous disease, who is subjected to major hepatic resection for spontaneous liver rupture.
