The Orthopoxvirus genus,especially variola virus(VARV),monkeypox virus(MPXV),remains a significant public health threat *** development of therapeutic antibodies against orthopoxviruses is largely hampered by the high cost of antibody engineering and manufacturing ***-encoded antibodies have emerged as a powerful and universal platform for rapid antibody ***,by using the established lipid nanoparticle(LNP)-encapsulated mRNA platform,we constructed four mRNA combinations that encode monoclonal antibodies with broad neutralization activities against *** vivo characterization demonstrated that a single intravenous injection of each LNP-encapsulated mRNA antibody in mice resulted in the rapid production of neutralizing *** importantly,mRNA antibody treatments showed significant protection from weight loss and mortality in the vaccinia virus(VACV)lethal challenge mouse model,and a unique mRNA antibody cocktail,Mix2a,exhibited superior in vivo protection by targeting both intracellular mature virus(IMV)-form and extracellular enveloped virus(EEV)-form *** summary,our results demonstrate the proof-of-concept production of orthopoxvirus antibodies via the LNP-mRNA platform,highlighting the great potential of tailored mRNA antibody combinations as a universal strategy to combat orthopoxvirus as well as otheremerging viruses.

BACKGROUND Integrating conventional ultrasound features with 2D shear wave elastography(2D-SWE)can potentially enhance preoperative hepatocellular carcinoma(HCC)*** To develop a 2D-SWE-based predictive model for preoperative identification of *** A retrospective analysis of 884 patients who underwent liver resection and pathology evaluation from February 2021 to August 2023 was conducted at the Oriental Hepatobiliary Surgery *** patients were divided into the modeling group(n=720)and the control group(n=164).The study included conventional ultrasound,2D-SWE,and preoperative laboratory *** logistic regression was used to identify independent predictive factors for RESULTS In the modeling group analysis,maximal elasticity(Emax)of tumors and their peripheries,platelet count,cirrhosis,and blood flow were independent risk indicators for *** factors yielded an area under the curve of 0.77(95%confidence interval:0.73-0.81)with 84%sensitivity and 61%*** model demonstrated good calibration in both the construction and validation cohorts,as shown by the calibration graph and Hosmer-Lemeshow test(P=0.683 and P=0.658,respectively).Additionally,the mean elasticity(Emean)of the tumor periphery was identified as a risk factor for microvascular invasion(MVI)in malignant liver tumors(P=0.003).Patients receiving antiviral treatment differed significantly in platelet count(P=0.002),Emax of tumors(P=0.033),Emean of tumors(P=0.042),Emax at tumor periphery(P<0.001),and Emean at tumor periphery(P=0.003).CONCLUSION 2D-SWE’s hardness value serves as a valuable marker for enhancing the preoperative diagnosis of malignant liver lesions,correlating significantly with MVI and antiviral treatment efficacy.

Objective:To investigate the effects of ondonin on nuclear transcription factors and the relationship of which target genes with inflammatory factors in human BxPC-3 ***:BxPC-3 cells were treated with different concentrations of oridonin,and it viability curve were used to test the inhibition effect of oridonin on the BxPC-3 *** expression of cytokines in the BxPC-3 cells supernatant was detected by ELISA and the protein expression of nuclear transcription factors,such as NF-κB,AP-1,IL-33,STAT3,and Smads,and its taget gene proteins,like Survivin,VEGF,and MMP-2,in the BxPC-3 cells was detected by western blot and immunonuorescent ***:The viability curve of BxpC-3 cells treatment with oridonin exhibited a dose-dependent *** analysis revealed that oridonin can down-regulate the inflammation factor(IL-1β,IL-6,IL-33)in a dose-dependent *** blot,and immunonuorescent staining analysis suggested that oridonin can regulate proteins expression of the nuclear transcription factors(NF-κB,IL-33,AP-1,STAT3,Smads),and influenced corresponding inflammatory factors expression which related target ***:The results suggested that oridonin against the pancreatic cancer maybe by regulating the nuclear transcription factors and its targeted inflammatory factors.

We conducted a comprehensive review of existing prediction models pertaining to the efficacy of immune-checkpoint inhibitor(ICI)and the occurrence of immune-related adverse events(irAEs).The predictive potential of neutrophil-to-lymphocyte ratio(NLR)and platelet-to-lymphocyte ratio(PLR)in determining ICI effectiveness has been extensively investigated,while limited research has been conducted on predicting ***,the combined model incor-porating NLR and PLR,either with each other or in conjunction with additional markers such as carcinoembryonic antigen,exhibits superior predictive capabilities compared to individual markers *** and PLR are promising markers for clinical *** models ought to incorporate established efficacious models and newly identified ones,thereby constituting a multifactor composite ***,efforts should be made to explore effective clinical application approaches that enhance the predictive accuracy and efficiency.

Two novel phosphors LiBa_(4(1-x))Eu_(4x)Ta_(3)O_(12)(H-LBTO:xEu^(3+)) and Li_(0.25)Ba_(1-x)Eu_(x)Ta_(0.75)O_(3)(C-LBTO:xEu^(3+))were prepared successfully by a molten salt *** transformation between these two structures was realized by changing the sintering temperature or changing the Eu^(3+) ions concentration,which was also demonstrated by the X-ray diffraction(XRD),scanning electron microscopy(SEM),diffuse reflectance spectra(DRS),and photoluminescence excitation(PLE) analyses,Both the sintering temperature and the Eu^(3+)ions doping concentration have significant impact on the formation of the crystal *** these phosphors sintered at 1023 K exhibit two major luminescence lines at 594 and 614 nm under nearuV light of 395 nm excitation,corresponding to Eu^(3+)ions typical transitions of ^(5)D_(0)→^(7)F_(1) and ^(5)D_(0)→^(7)F_(2).The optimum concentration of Eu^(3+) ions is 9 mol% for C-LBTO:xEu^(3+) samples and the quenching interaction type is the nearest-neighbor ion *** thermal stability of the C-LBTO:0.09Eu^(3+)sample was investigated in detail and the device application further suggests that C-LBTO:0.09Eu^(3+) can be used as a red phosphor for near-UV excited w-LEDs in lighting.

Constructed wetlands(CWs)are widely applied for decentralized wastewater ***,achieving efficient removal of ammonia(NH+-N)has proven challenging due to insufficient *** this study,natural hematite(Fe,O3)was employed as a CW substrate(H-CWs)for the first time to drive anaerobic ammonia oxidation coupled with iron(I)reduction(Feammox).Compared to gravel constructed wetlands(G-CWs),ammonia removal was enhanced by 38.14%to 54.03%and nitrous oxide(N_(2)0)emissions were reduced by 34.60%in *** synergistic removal of ammonia and nitrate by H-CWs also resulted in the absence of ammoxidation *** and 15N isotope tracer incubations showed that Feammox accounting for approximately 40%of all ammonia removal in the *** enrichment of iron phosphate(Fe_(3)Fe_(4)(PO_(4))_(6))promoted the accumulation of the Feammox intermediate compound *** nanowires were observed on the surface of H-CW substrates as well,suggesting that the observed biological ammoxidation was most likely related to extracellular electron transfer(EET).Microbial and metagenomics analysis revealed that H-CWs elevated the integrity and enhanced the abundance of functional microorganisms and genes associated with nitrogen ***,the efficient ammonia removal in the absence of O2 together with a reduction in N_(2)O emissions as described in this study may provide useful guidance for hematite-mediated anaerobic ammonia removal in CWs.
