This article presents and analyses the modular architecture and capabilities of CODE-DE(Copernicus Data and Exploitation Platform–Deutschland,***),the integrated German operational environment for accessing and processing Copernicus data and products,as well as the methodology to establish and operate the *** March 2017,CODE-DE has been online with access to Sentinel-1 and Sentinel-2 data,to Sentinel-3 data shortly after this time,and since March 2019 with access to Sentinel-5P *** products are available and accessed by 1,682 registered users as of March *** this period 654,895 products were downloaded and a global catalogue was continuously updated,featuring a data volume of 814 TByte based on a rolling archive concept supported by a reload mechanism from a long-term *** November 2017,the element for big data processing has been operational,where registered users can process and analyse data themselves specifically assisted by methods for value-added product *** 195,467 core and 696,406 memory hours,982,948 products of different applications were fully automatically generated in the cloud environment and made available as of March *** features include an improved visualization of available Sentinel-2 products,which are presented within the catalogue client at full 10 m resolution.

Prevalence and risk factors of nonalcoholic fatty liver disease in patients with inflammatory bowel diseases:A cross-sectional and longitudinal analysis

BACKGROUND Nonalcoholic fatty liver disease(NAFLD)is common in the German population,with an even higher prevalence in inflammatory bowel disease *** To investigate the risk factors for NAFLD in inflammatory bowel disease *** This monocentric retrospective study with a cross-sectional and a longitudinal part included 694 *** criteria were diagnosed inflammatory bowel disease,age≥18 years,availability of at least one abdominal *** with infectious or suspected alcoholic fatty liver disease were *** was defined by increased echogenicity at liver *** characteristics,disease activity and medications were analyzed as potential risk *** influencing the course of NAFLD were identified by a generalized linear mixed *** Forty-eight percent of Crohn’s disease(CD)patients and 44%of ulcerative colitis patients suffered from *** occurrence was associated with greater age,hypertension and body mass index(BMI)in both groups,and with higher disease activity and dyslipidemia in CD.2467 ultrasound results were included in the longitudinal *** factors for NAFLD were age,BMI,higher disease activity,bowel resection(s),endoscopic activity and azathioprine use in CD;and BMI and endoscopic activity in ulcerative *** NAFLD was highly prevalent in this cohort of German inflammatory bowel disease *** risk increased mainly with rising age and *** analysis provides a rationale for non-invasive liver screening in inflammatory bowel disease patients.

The Role of Quality Control in Targeted Next-generation Sequencing Library Preparation

Next-generation sequencing （NGS） is getting routinely used in the diagnosis of hereditary diseases, such as human cardiomyopathies. Hence. it is of utter importance to secure high quality sequencing data, enabling the identification of disease-relevant mutations or the conclusion of negative test results. During the process of sample preparation, each protocol for target enrichment library preparation has its own requirements for quality control （QC）; however, there is little evi- dence on the actual impact of these guidelines on resulting data quality. In this study, we analyzed the impact of QC during the diverse library preparation steps of Agilent SureSelect XT target enrichment and lllumina sequencing. We quantified the parameters for a cohort of around 600 samples, which include starting amount of DNA, amount of sheared DNA, smallest and largest fragment size of the starting DNA; amount of DNA after the pre-PCR, and smallest and largest fragment size of the resulting DNA; as well as the amount of the final library, the corresponding smallest and largest fragment size, and the number of detected variants. Intriguingly, there is a high tolerance for variations in all QC steps, meaning that within the boundaries proposed in the current study, a considerable variance at each step of QC can be well tolerated without compromising NGS quality.

The Mesenteric-Caval Fistula: First Results of a New Technique in a Transperitoneal Reconstruction of the Caval Vein by Fulminant Thrombosis of the Inferior Vena Cava Based on Homozygous Antithrombin III-Deficiency

Recurrent thrombotic occlusions are one major problem in patients with thrombosis of the inferior vena cava. Due to this, we report a new surgical strategy for the construction of aorto-caval (mesenteric-caval) fistula in a patient with homozygous Antithrombin III (ATIII)-Deficiency. The patient survived postoperatively and only surgical complications grade I and II (Clavien-Dindo classification) were reported after short-term and one year follow-up. After one year, the CT-angiography did not show any caval thrombosis or stenosis and no restriction or occlusion of the fistula. Thus, the mesenteric-caval fistula could be safely performed and resulted in a satisfactory patency.
