Characterization of a novel RInG-Type Ubiquitin E3 Ligase GhRInG2 Differentially Expressed in Cotton Fiber

The ubiquitin-proteasome proteolysis pathway is responsible for the degradation of abnormal and short-lived proteins to regulate many important biochemical activities in eukaryotes. By employing affymetrix microarray analysis, we have identified a novel ubiquitin ligase E3 gene GhRInG2 that is differentially expressed between two Gossypium hirsutum lines-Texas Marker-1 (TM-1) and Chromosome Substitution Line CS-B25. The CS-B25 line has chromosome 25 from G. barbadense substituted into TM-1. The complete nucleotide sequences of GhRInG2 along with its 5’-flanking region were obtained by genomic walking. GhRInG2 was highly expressed in elongating fiber, and GUS expression directed by the GhRInG2 promoter was found in hypocotyls and young stems of transgenic Arabidopsis plants. Using a yeast two-hybrid assay GhRInG2 was found to interact with a PROTODERMAL FACTOR1 (GhPDF1) protein. GhPDF1 was expressed preferentially in immature ovules and fiber initials, and the GhPDF1 gene had been suggested to play a role in cell fate determination and fiber development. Pull down and plasmid swap assays further confirmed the interaction between GhRInG2 and GhPDF1. The expression and protein interaction data indicate that GhRInG2 may be involved in the turnover of GhPDF1 and participation in the transition from initiation to elongation stages during fiber development. Our data strongly suggest that the ubiquitin-proteasome pathway may regulate cotton fiber growth and development. The nucleotide sequence data of GhRInG2 in this article have been submitted to the Gen Bank nucleotide Sequence Data Bases under the accession number BankIt 1,742,008 SeqKM 108,000.

Molecular Microbial Diagnostic in Lung Transplant Recipients

The appearing of microbial lineages carrying multiple dangerous loci are results from the extensive use of antibiotics and has brought a huge increase in the infections-derived morbidity and mortality, which is critical in the hospital setting. Etiologic agents of these non easily tractable infections include bacteria, such as the genus Staphylococcus, but also fungi and virus. Alterations in the immune system allow these organisms to invade and affect the functionality of any tissue, organ or system of the human being. Pulmonary infection occurs as result of deficient lung systemic defence mechanisms which could be altered by medical treatments or by environmental factors. The infective agents commonly gain access to the lung by air, but also by blood or lymphatic system. Staphylococcus aureus strains that share antibiotic resistance and virulence factors represent the aetiological agent responsible of many cases of bacterial pneumonia, thoracic surgery postoperative infections, and diverse tissue infections, resulting in significant disease and morbidity in recipient patients after lung transplantation. The control and treatment of Staphylococcus infections, especially methicillin resistant strains, need for developing reliable and rapid methods of detection and characterization of these microorganisms. nowadays, new insights into the diagnostic and epidemiology of MRSA and other pathogenic staphylococci have been developed employing molecular methods. This has meant an important advance in the diagnostic and treatment plans of such infective bacteria.

Askin Tumor in Egyptian Patients;5 Years Experience at the national Cancer Institute, Cairo University

Background: Askin tumor is a primitive neuroectodermal tumor of the Ewing sarcoma family arising from soft tissues of the chest wall. It is the commonest chest wall malignancy in children and adolescents. Its diagnosis is complex and management requires a multidisciplinary work including chemotherapy for systemic disease and radiation therapy to assist local control which is achieved through surgery with or without reconstruction. Objective: To analyze report and understand the clinicopathological features, results and outcome of this tumor with assessment of early and late postoperative complications following resection and chest wall reconstruction. Materials and Methods: This is a retrospective analysis of 30 cases with chest wall ES/PnET presented to the national Cancer Institute;Cairo University between January 2011 and December 2015. All patients’ records were revised for age, sex, clinical presentation, imaging, pathology, operative notes, different treatment modalities given, early and late postoperative complications following surgical resection or reconstruction, and outcome including overall survival (OS) and disease free survival (DFS). Results: Our cohort included 30 patients with chest wall ES. The median follow up period (n = 30) was 33.7 months (ranging from 3.7 to 69.3 months). The median OS was 54.2 months with cumulative OS at 60 months which was 45.6%. The median DFS was 27.9 months with cumulative DFS at 60 months which was 40.6%. All cases were below 18 years with a male predominance (n = 19). The commonest affected sites were ribs (n = 22, 73.3%), the scapula (n = 5, 16.6%), the clavicle (n = 2, 6.66%) and the sternum (n = 1, 3.33%). 23 patients (76.6%) were presented with localized disease, and 7 patients (23.4%) were metastatic from the start. All patients received neoadjuvant chemotherapy (4 cycles of VAC/IE) followed by local control: either surgery (26 cases, 86.6%) or radical radiotherapy (3 cases, 10%). Magnetic Susceptibility and natural Gamma Radioactivity as Indirect Indicators of Impurities in Limestones of the Sabinas Basin: Possible Relationships with the Sedimentation Environment

This study presents an analysis of magnetic susceptibility and natural gamma radioactivity as indirect indicators of impurities in Cretaceous limestones of the Sabinas basin, as well as of the possible relation of these physical properties with the changes in the sedimentation environment. Both of these physical properties indicate changes in the degree of impurities and the mineralogical composition of this rock, principally in its organic matter and detritus content. Considering that the sedimentation environment determines the impurities in these rocks, possible environmental conditions at different levels of the Cretaceous were suggested using the magnetic susceptibility, total gamma radiation and potassium values, as well as the uranium and thorium concentration in the limestones. The analyses suggest variations in the depth of water table of the basin, which are mainly related to transgressions and marine regressions.

Electric,magnetic and microstructural features of the La_(2)CoFeO_(6) lanthanide ferrocobaltite obtained by the modified Pechini route

In the search for new materials with applicable magnetic properties in spintronic devices,the aim of this work is to report the synthesis of the lanthanide ferrocobaltite La_(2)CoFeO_(6) using the modified Pechini route;the experimental study of structural,morphological and magnetic properties,and the analysis of the electronic structure and bands are obtained in the framework of the Density Functional *** refinement of experimental X-ray diffraction patterns revealed the crystallization of this oxide material in a perovskite-like monoclinic structure,space group P2_(1)/n(#14).Scanning electron microscopy and atomic force microscopy images revealed that the surface morphology is essentially polycrystalline,with mean grain sizes between 177 and 188 *** dispersive X-ray spectroscopy suggests that the material obtained contains La,Fe,Co and O in the stoichiometric proportions expected by up to 98%.The magnetic susceptibility curves as a function of temperature indicated that the material is ordered ferromagnetically,showing strong irreversibility effects due to the disorder of the Fe and Co cations in the three crystallographic directions of the structure and to the strong distortions in the FeO_(6) and CoO_(6) *** hysteresis curves confirmed the ferromagnetic character of the material for all temperatures evaluated,up to room temperature.I-V response curves revealed a semiconductor-like behavior with a figure of merit exponent 1.53 of the varistor *** ferromagnetic semiconductor behavior suggests the potential applicability of the material in spintronic technological devices.

Optimizing the Global Digital Elevation Models(GDEMs) and accuracy of derived DEMs from GPS points for Iraq’s mountainous areas

Optimizing the combined horizontal and vertical accuracy of the well-known Global Digital Elevation Models(GDEMs) of various resolutions for each country and region especially in Iraq’s mountainous areas is still questionable. All the three GDEMs, approximately, have the same vertical accuracy with the Root Mean Square(RMSE) of ±7.3 m, ±7.6 m and ±6.5 m via 12 fixed Ground Control Points(GCPs) for the Advanced Land Observation Satellite Phased Array L-band Synthetic Aperture Radar(ALOS PALSAR 12.5 m), the Shuttle Radar Topographic Mission(SRTM 30 m) and the TerraSAR-X(the name of twin satellites) add-on for Digital Elevation Measurement(TanDEM-X 90 m) GDEMs respectively. Moreover, the percentage of outliers that are greater or smaller than ±10 m detection of the height extraction from both the ALOS PALSAR and SRTM Digital Elevation Models(DEMs) contains 16.7% and for TanDEM-X was 25%. In this paper,the special DEM is derived using 2123 handheld GPS points for Sulaymaniyah Governorate, Kurdistan region, Iraq. The height extraction by discarding the outliers of 58% gives the RMSE of ±8.0 m in the case of adding geoid heights(n) to the ellipsoidal heights(h) via the Earth Gravitational Model 2008(EGM2008)and ±5.6 m without adding n. It is expected that the derived DEMs will give more accurate results both horizontally and vertically in the mountainous areas when GPS observations are intensified. The horizontal accuracy is validated through extracting hierarchy stream types of the watershed map from the DEMs for higher than 100 pixels length. The ALOS PALSAR DEM extracted more numbers of stream orders than ***, based on the criteria of RMSE, outlier detection, and the number of extracted stream orders, the ALOS PALSER DEM is regarded as the optimal GDEM in comparison with the close accuracy of both the TanDEM-X and SRTM DEM.

Parasite Communities of the Pacific Mole Crab,Emerita rathbunae(Anomura:Hippidae),in Sandy Beaches from Guerrero and Michoacan,Mexico

The species composition and infection levels were determined for helminth parasites in the mole crab, Emerita rathbunae, collected from seven sandy beaches from Guerrero and Michoacán, México. A total of 494 crabs were collected between August and December of 2009. The number of crabs that were examined from each beach varied from 40 in Lázaro Cárdenas to 114 in El Revolcadero. The cephalothorax length varied significantly between the sampled beaches, from 32.9 ± 5.5 mm (Ixtapa) to 40.5 ±1.7 mm(Las Trancas). Four species of larval parasites were identified: 1 metacercaria (Microphallus nicolli), 1 cystacanth (Profilicollis sp.), 1 cestode (Trypanorhyncha) and 1 nematode (Proleptus sp.). Infection levels (prevalence and mean abundance) varied significantly between beaches, due to possible differences in the availability of final or intermediate hosts in beaches visited by tourist and those beaches not visited by tourists, as well as the size of individual hosts. Helminth communities, at levels of component and infracommunity, were characterized by a low number of species (3 to 4) and a high dominance by the metacercaria of M. nicolli. The body size of the hosts was positively correlated with the number of parasites and species richness of helminths, indicating that larger crabs accumulate a higher number of parasites during the lifetime, and that they harbor a higher number of species of helminth than smaller crabs.

A novel H7n9 influenza A virus has been discovered as the causative identity of the emerging acute respiratory infection cases in Shanghai,*** virus has also been identified in cases of infection in the neighboring area Hangzhou City in Zhejiang *** this study,epidemiologic,clinical,and virological data from three patients in Hangzhou who were confirmed to be infected by the novel H7n9 influenza A virus were collected and *** respiratory specimens and chicken feces from a contacted free market were tested for influenza virus by real-time reverse transcription PCR(RT-PCR) and *** clinical features of the three cases were similar featured with high fever and severe respiratory symptoms;however,only one of the patients died.A certain degree of diversity was observed among the three Hangzhou viruses sequenced from human samples compared with other reported H7n9 influenza A *** sequences of the novel avian-origin H7n9 influenza viruses from Hangzhou City contained important amino acid substitutions related to human *** of the Hangzhou viruses had gained a novel amino acid substitution(Q226I) in the receptor binding region of *** importantly,the virus sequenced from the chicken feces had a 627E substitution in the PB2 protein instead of the mammalian-adapted 627K substitution that was found in the PB2 proteins from the Hangzhou viruses from the three ***,the newly-emerging H7n9 virus might be under adaptation pressure that will help it "jump" from avian to human *** significance of these substitutions needs further exploration,with both laboratory experiments and extensive field surveillance.

Optical Fiber Torsion Sensor with Mechanically Induced Long Period Fiber Gratings in Rare-Earth Doped Fibers

In this work wavelength sensitivity in mechanically induced long period fiber gratings (MLPFG) is analyzed. This analysis is first carried out both in standard single-mode fiber SMF-28 and in Er-doped fibers. The mechanical analysis for both types of fibers under different torsion conditions is presented. In order to apply the torsion one of the fiber ends is fixed while torsion is applied on the other end. A MLPFG whose period is 503 μm is used to press the fiber after torsion is applied. This allows for micro curvatures to be formed on the fiber, which in turn generates a periodical index perturbation on it. Here, it was noted that the sensitive wavelength shift of the rejection bands is bigger for Er-doped fibers. For a torsion of 6 turns applied to 10 cm of doped fiber the wavelength peaks can be moved up to 25 nm, which is longer to what was detected on standard fibers. Therefore, by using Er-doped fibers to monitor torsion on structures will give more sensitive and accurate results than using standard fibers. These results can be employed for sensing applications, especially for small to medium size structures, which can be mechanical, civil or aeronautics.
