Gene Expression Analysis of Extracellular Matrix and Cytokines after Uterine Artery Embolization

Arterial embolization of myomas (AEM) is an established option for the conservative treatment of uterine leio-myomas;it treats all present uterine nodules at once, is less invasive than other procedures and effective in controlling symptoms, and does not require long term hospitalizations. Nevertheless, the potential impact on endometrial morphological and functional outcomes after the procedure is still controversial based on reports of endometritis or eventual transient ischemia. This study evaluated endometrial reorganization in uterine leiomyoma patients, before and after AEM, through gene expression analyses of extracellular matrix and cytokines genes in theendometrial tissue. Eight patients with leiomyomas were evaluated before AEM and 6 months after. The examinations included transvaginal pelvic ultrasonography, dosing of the follicle-stimulating hormone, and endometrial biopsy during the second phase of the menstrual cycle. RNA was extracted from endometrial samples, cDNA was synthesized, and applied on PCR arrayTM plates to evaluate the expression of extracellular matrix (ECM) genes and cytokines and their receptors’ genes (CYT). The ECM overexpressed genes were MMP (1, 3, 10, 11, and 14), CTGF1, ICAM1, TBHS1, ITGA2, ITGA3, ITGB3, COL7A1, COL12A, SPP1, and TNC;AdaMTS8 was underexpressed. The CYT overexpressed genes were SPP1, BCL6, CXCL12, IL-8, and CEBPB;CXCL13 and CCL21 were underexpressed. The ECM results showed overexpression of proteases that are responsible for dysfunctions in the ECM, and of genes responsible for adhesion and membrane components. The CYT results showed overexpression of chemokines responsible for endometrial repair, and underexpression of cytokines involved in inflammatory processes in the endometrial tissue. AEM treatment did not negatively affect the endometrial function at 6 months after embolization. Educational Video Increases Maternal Self-Efficacy for Prevention of Diarrhoea in Young Children:A Randomised Clinical Trial

The aim of this study was to compare the scores of maternal self-efficacy for child diarrhoea prevention, between intervention and control groups, according to sanitary, social and demographic characteristics. A randomised clinical trial was carried out on 163 mothers of children under 5 years old. data collection was conducted during two months, at two different moments (M0 and M1);at the moment M0 only the intervention group watched the educational video to promote self-efficacy for child diarrhoea prevention;at M1 (two months later) the scale was applied to both groups. In the intervention group, greater mean self-efficacy scores were obtained compared with the control group. The following variables showed statistically significant associations (p < 0.05) with maternal self-efficacy: age 35 or over;high school education;working outside the home;residence with 4 to 5 people;house not of brick or mortar;public refuse collection;piped water supply;consumption of mineral water;and public sewerage network. After use of the video “Diarrhoea: you can prevent it”, the maternal self-efficacy increased, proving that this strategy is able to assist the nursing staff in educational practices.

Phosphodiesterase inhibitors for treatment of voiding dysfunction: An overview of experimental and clinical evidence

Recently,the focus of the origin of lower urinary tract symptoms(LUTS)has change from the prostate to the *** of the underlying mechanism associated with the origin of LUTS,alpha-blockers continue to be the most common medicine prescribed to treat LUTS due to benign prostatic obstruction(BPO).The newest class of drug introduced to treat LUTS/BPO is phosphodiesterase inhibitors(PDEi)and the aim of this study was to review the role of PDEi in the treatment of LUTS/*** this review,the first evidence was evaluated based on epidemiological studies followed by randomized clinical trials which provide evidence on the administration of PDEi in patients with LUTS/*** studies were also assessed to tentatively elucidate the association between LUTS and erectile dysfunction,and to elucidate the underlying *** is still controversy regarding the administration of PDEi due to the fear of detrusorimpairment,response to acute administration,and the effects of PDEi combined with *** this review,we conclude that treatment of BPO/LUTS with PDEi is beneficial,based on experimental studies,strong evidence and the large number of randomized clinical trials confirming their efficiency.

NaPi-IIb is a multiple passage protein membrane which is primarily responsible for phosphate uptake in the kidney and in the small intestine. Beyond its physiological functions, their involvement with carcinogenesis was initially described in mid-2003, due to its distinct level of expression in normal and tumor cells of the ovary. Although less common than cervical cancer, epithelial ovarian cancer is considered the most lethal gynecologic malignancy, which is mainly due to diagnosis in the advanced stages as a result of the absence of symptoms during the onset of the disease and the lack of tools for early detection. Here, we produce antibodies that are anti-synthetic peptides that are derived from the regions of second extracellular loop of NaPi-IIb, which is a non-overlapping portion of MX35 epitope. These two 15 distinct amino acid residue peptides, designated as Let#1 and Let#2, are engineered in a very thorough way to detect specific sites only in this isoform, thus excluding cross-reactivity with other carriers of the same family. The lack of immunogenicity of small peptides is surpassed by the conjugation with carrier proteins. Using immunochemical methods, we demonstrate that polyclonal antibodies that are mono-specific for the Let#1 and Let#2 peptides recognize proteins that express similar amino acid sequences during blood circulation. Additionally, using flow cytometry, we identify NaPi-IIb in NIH:OVCAR-3 cells. The clear identification of two shorter peptides on the extracellular loop of NaPi-IIb, which are far from the monoclonal antibody MX35-recognizing epitopes, adds new promising tools for ovarian cancer follow-up and staging.

AIM To develop metabonomic models(MMs), using 1 H nuclear magnetic resonance(NMR) spectra of serum, to predict significant liver fibrosis(SF: Metavir ≥ F2), advanced liver fibrosis(AF: METAVIR ≥ F3) and cirrhosis(C: METAVIR = F4 or clinical cirrhosis) in chronic hepatitis C(CHC) patients. Additionally, to compare the accuracy of the MMs with the aspartate aminotransferase to platelet ratio index(APRI) and fibrosis index based on four factors(FIB-4). METHODS Sixty-nine patients who had undergone biopsy in the previous 12 mo or had clinical cirrhosis were included. The presence of any other liver disease was a criterion for exclusion. The MMs, constructed using partial least squares discriminant analysis and linear discriminant analysis formalisms, were tested by cross-validation, considering SF, AF and C. RESULTS Results showed that forty-two patients(61%) presented SF, 28(40%) AF and 18(26%) C. The MMs showed sensitivity and specificity of 97.6% and 92.6% to predict SF; 96.4% and 95.1% to predict AF; and 100% and 98.0% to predict C. Besides that, the MMs correctly classified all 27(39.7%) and 25(38.8%) patients with intermediate values of APRI and FIB-4, respectively. CONCLUSION The metabonomic strategy performed excellently in predicting significant and advanced liver fibrosis in CHC patients, including those in the gray zone of APRI and FIB-4, which may contribute to reducing the need for these patients to undergo liver biopsy.

Tropical Isabella Grape Juices： Bioactive Compounds and Antioxidant Power Depends on Harvest Season

Grapes are important sources of antioxidants compounds and one of the most used varieties to elaborate juices is ＂Isabel＂, or ＂Isabella＂, as it is called in North-America. This study aimed to evaluate the antioxidant activity of Isabella grape juices from the tropical semi-arid climate produced in Brazil on March and September, 2010. Total phenolics and anthocyanins were determined, as well as the antioxidant capacity by DPPH, FRAP and hydrophilic ORAC methods. Test T was used to compare statistical difference at 5% of significance level. As expected, the results showed that the season can play a significant role on phenolics content and antioxidant power. Polyphenols content in juices varied from 82.9 ± 0.92 to 102.2± 1.59 mg GAE 100 mL1- and anthocyanins content ranged from 44.3 ± 2.01 to 129.5 ± 2.82 mg cyanidin-3-glucoside 100 mL-1. Regression analyses showed a high correlation of antioxidant capacity and anthocyanins content. Grapes from Sao Francisco River Valley demonstrated to be a good source of antioxidant for human diet.

Circulation of Immunosuppressive Viruses and Avian Encephalomyelitis Virus in Backyard Chicken Flocks

The objective of this study was to evaluate the circulation of Chicken Anemia Virus (CAV), Infectious Bursal Disease Virus (IBDV), Avian Reovirus (ARV) and Avian Encephalomyelitis virus (AEV) in properties of backyard chickens and carry out an epidemiological analysis. We evaluated 200 samples of chickens from 19 backyard chicken property. Only one property (P10) did not present serological titers for the diseases evaluated. This property is close to industrial farms as well as the other properties, however, P10 remained a few years without the breeding of chicks and these were the first poultry to be housed on site. This reinforces the importance of the fallow period for poultry production. The prevalence of virus-seroreactive birds was 78% (156/200), 64.5% (129/200), 78% (156/200), 78% (156/200) for CAV, IBDV, ARV and, EA, respectively. All the free-range farms studied are within a radius of 500 meters to 6 Km away from some establishments of industrial poultry. There was a correlation between serological titers for CAV and the frequency of disease in poultry (r = 0.6178). In places where birds are frequently sick, 30.76% reported that the disease occurs in chicks, 30.76% in broilers, 23.07% in broiler chickens and 7.69% in birds of all ages. Birds get sick more often in the summer period. The owners reported that the most common signs of disease were respiratory signs (snoring and nasal discharge) (46.15%), diarrhea (30.76%), and paralysis of wings and/or paws (38.46%). There was a correlation between the presence of untreated water in the property and serological titers for ARV (r = 0.5576). This report draws attention not only to high serological prevalence for the viruses studied but also important epidemiological aspects of backyard chicken diseases that may indirectly influence the industrial production.

Isothermal Transport (Core-Flow Type) of Heavy and Ultraviscous Oil in Curved Pipes: A Transient Study by CFD

In the scenery of the oil industry, the remaining resources associated with light oils have an increasingly smaller share in the natural energy resources available to man, and in return the importance of resources associated with heavy oils has increased significantly. One of the drawbacks of this type of oil is associated with its low mobility due to the high viscosity in reservoir conditions, making the transport in pipelines very difficult, especially through pumping methods that require high powers. Thus, the development of new techniques and optimization of some existing technologies, aiming at the commercial use of heavy oil accumulations plays an important role. A viable technique that has been used is the core annular flow, in which small amounts of water are injected close to the pipe wall, lubricating the oil core, reducing friction and decreasing the pressure drop during the flow. In this sense, this work aims to perform, numerically, an energetic and hydrodynamic analysis of a heavy oil-water two-phase flow, using the core-flow technique, in curved pipes, in the Ansys CFX software. Results of the velocity, pressure, and volume fraction distribution of the involved phases are presented and analyzed. It was observed that the proposed mathematical model was able to accurately represent the analyzed phenomena and that a reduction factor in the pressure drop of 28.4 was obtained as compared to the heavy oil single-phase flow.

Growth and Photosynthetic Pigments of Cotton Cultivars Irrigated with Saline Water

The cultivation of cotton is a relevant socioeconomic activity in the Brazilian agricultural scenario. In the Brazilian Northeast, however, production is limited by low rainfall, requiring water supplementation, a problem for the cotton cultivation practiced in the semi-arid region, due to the presence of saline ions in several water sources, from where the water is used for irrigation. It is necessary to identify cultivars that better tolerate saline stress in order to provide subsidies for their cultivation under such conditions. In this sense, the objective was to evaluate the tolerance of cotton cultivars to salinity. The experiment was conducted in a greenhouse, belonging to the Natural Resource Technology Center of the Federal University of Campina Grande, Campus I, Campina Grande, Brazil, in a completely randomized design in a 2 × 5 factorial scheme, with four replications, totaling 40 experimental units (BRS 368 RF and BRS SAFIRA) submitted to five saline levels (1.5, 3.0, 4.5, 6.0 and 7.5 dS·mˉ1). Irrigation with water saline of EC from 1.5 dS·mˉ1 affects adversely the growth of cotton cultivars, causing reductions in plant height, stem diameter, leaf area and number of leaves. Saline stress reduced the levels of the photosynthetic pigments of the cotton plants studied.

Impact of a physical activity program on plasma concentrations of adiponectin, leptin and ghrelin in overweight and obese schoolchildren: A randomized controlled trial

Introduction: Child obesity has important consequences on the child’s health and is related to the risk of developing chronic diseases. That brings up the need for research assessing the impact of obesity-and overweight-reduction programs with close attention to methodology aspects and precise indicators of the impact of such programs. Goal: Assessing the impact of a physical activity program on the plasma concentrations of adiponectin, leptin, and ghrelin in overweight and obese schoolchildren. Methods: Randomized controlled trial, double-blind, with 6-to 9-year-old overweight and obese school-children. A sample of 79 schoolchildren randomly distributed into control and intervention groups was subjected to a physical activity program. The levels of adiponectin, leptin, ghrelin, body mass index, body fat percentage, anthropometry, lipid profile and glycemia were evaluated. The program’s impact was assessed through multivariate linear regression models. Result: When comparing the groups, a significant difference was found for adiponectin, which shows an increase in the proportion of subjects with higher adiponectin concentrations for the intervention group and a reduction in body mass index and body fat percentage. Conclusion: The impact of the intervention program was positive in increasing adiponectin levels associated with a reduction in body mass index and body fat percentage.
