Development,Validation and Characterization of Genic Microsatellite Markers in Urochloa Species

Urochloa (syn. Brachiaria) cultivars represent 85% of tropical pastures occupying 114 million hectares of cultivated grasslands in Brazil. Despite the commercial importance of the Urochloa species, low molecular information is available and is far from saturating the genome. Investigating and obtaining more markers associated to characteristics of difficult and late expression can benefit in accelerating breeding programs of more important species. Aiming to increase the number of molecular markers, genic microsatellite markers were obtained from transcriptome of U. decumbens and analyzed for their cross-amplification to U. brizantha, U. humidicola and U. ruziziensis. Genic microsatellite markers were isolated from a transcriptome obtained of U. decumbens “Basilisk” roots. Specific primers were designed for one hundred loci, and 32 were polymorphic presenting polymorphism informative content values ranging of 0.12 to 0.81 (mean 0.54). Amplified microsatellite regions yield an average of 4.44 alleles per locus (ranging of 1 to 13). Cross-amplification to U. brizantha, U. humidicola and U. ruziziensis were successfully performed, although the number of the loci transferred varied among them. Multiple Correspondence Analysis revealed three distinct groups separating accessions and species. Four genetic markers presented high potential to distinguish sexual and apomictic accessions of Urochloa and must be further investigated. The genic markers identified in this study are the first set of expressed sequence tagged molecular markers for Urochloa species.

AIM:To evaluate the effects of soy supplementation on insulin resistance,fatty liver and alanine aminotransferase(ALT) levels in non-diabetic patients with chronic hepatitis C(CHC).METHODS:In a prospective,randomized and singleblinded clinical trial,we compared patients with CHC who had casein as a supplement(n = 80)(control group),with patients who consumed a soy supplement diet(n = 80) [intervention group(IG)].Both groups received 32 g/d of protein for 12 ***:Patients' baseline features showed that 48.1% were overweight,43.7% had abdominal fat accumulation,34.7% had hepatic steatosis and 36.3% had an homeostasis model assessment index of insulin resistance(HOMA-IR) ≥ *** analysis showed that protein supplementation diet reduced hepatic steatosis in both groups;however,significant reductions in ALT levels occurred in the soy *** regression modeling indicated that in the presence of severe fibrosis(F3/F4),g glutamyl transferase elevation and high density lipoprotein(HDL) reduction,the intervention group had 75% less chance of developing hepatic steatosis(OR= 0.25;95% CI:0.06-0.82) and 55% less chance of presenting with an ALT level ≥ 1.5 × the upper limit of normal(ULN)(OR = 0.45,95% CI:0.22-0.89).Soy treatment did not have any effect on insulin resistance(OR = 1.92;95% CI:0.80-4.83),which might be attributed to the fact that the HOMAIR values at baseline in most of our patients were in the normal *** hepatic fibrosis,an ALT level > 1.5 × ULN and visceral fat were predictors of an HOMA-IR ≥ *** IG group had a reduced risk of an ALT level > 1.5 × *** HOMA-IR ≥ 3.0 and HDL < 35 mg/dL were also risk factors for increased ***:Soy supplementation decreased ALT levels and thus may improve liver inflammation in hepatitis C virus(HCV) patients;it also reduced hepatic steatosis in a subgroup of patients but did not change insulin *** should be considered in the nutritional care of HCV patients.

A comparative analysis of three different methods for the estimation of the basic reproduction number of dengue

The basic reproduction number,R0,is defined as the expected number of secondary cases of a disease produced by a single infection in a completely susceptible population,and can be estimated in several *** example,from the stability analysis of a compartmental model;through the use of the matrix of next generation,or from the final size of an epidemic,*** this paper we applied the method for estimating R0 of dengue fever from the initial growth phase of an outbreak,without assuming exponential growth of cases,a common assumption in many *** used three different methods of calculating R0 to compare the techniques'details and to evaluate how these techniques estimate the value of R0 of dengue using data from the city of Ribeir^ao Preto(SE of Brazil)in two *** results of the three methods are numerically different but,when we compare them using a system of differential equations developed for modeling only the first generation time,we can observe that the methods differ little in the initial growth *** conclude that the methods predict that dengue will spread in the city studied and the analysis of the data shows that the estimated values of R0 have an equal pattern overtime.

ESWL for difficult bile duct stones:A 15-year single centre experience

Endoscopic mucosal resection of large hyperplastic polyps in 3 patients with Barrett’s esophagus

AIM： To report the endoscopic treatment of large hyperplastic polyps of the esophagus and esophagogastric junction （EGJ） associated with Barrett＇s esophagus （BE） with low-grade dysplasia （LGD）, by endoscopic rnucosal resection （EMR）. METHODS： Cap fitted EMR （EMR-C） was performed in 3 patients with hyperplastic-inflammatory polyps （HIPs） and BE. RESULTS： The polyps were successfully removed in the 3 patients. In two patients, with short segment BE （SSBE）（≤ 3 cm）, the metaplastic tissue was completely excised. A 2 cm circumferential EMR was performed in one patient with a polyp involving the whole EGJ. A simultaneous EMR-C of a BE-associated polypoid dysplastic lesion measuring 1 cm × 10 cm, was also carried out. In the two patients, histologic assessment detected LGD in BE. No complications occurred. Complete neosquamous reepithelialization occurred in the two patients with SSBE. An esophageal recurrence occurred in the remaining one and was successfully retreated by EMR. CONCLUSION： EMR-C appears to be a safe and effective method for treating benign esophageal mucosal lesions, allowing also the complete removal of SSBE.

Carbon sequestration capability of Fagus sylvatica forests developing in the Majella National Park (Central Apennines,Italy)

Terrestrial ecosystems represent a major sink for atmospheric carbon(C) and temperate forests play an important role in global C cycling, contributing to lower atmospheric carbon dioxide(CO2) concentration through photosynthesis. The Intergovernmental Panel of Climate Change highlights that the forestry sector has great potential to decrease atmospheric CO2concentration compared to other sectoral mitigation activities. The aim of this study was to evaluate CO2sequestration(CO2S)capability of Fag us sylvatica(beech) growing in the Orfento Valley within Majella National Park(Abruzzo,Italy). We compared F. sylvatica areas subjected to thinning(one high-forest and one coppice) and no-management areas(two high-forests and two coppices). The results show a mean CO2S of 44.3 ± 2.6 Mg CO2ha-1a-1,corresponding to 12.1 ± 0.7 Mg C ha-1a-1the no-managed areas having a 28% higher value than the managed areas. The results highlight that thinning that allows seed regeneration can support traditional management practices such as civic use in some areas while no management should be carried out in the reserve in order to give priority to the objective of conservation and naturalistic improvement of the forest heritage.

Tumor budding predicts response to anti-EGFR therapies in metastatic colorectal cancer patients

AIM:To investigate whether the evaluation of tumor budding can complement K-RAS analysis to improve the individualized prediction of response to anti-epidermal growth factor receptor based therapies in metastatic colorectal cancer (mCRC) patients. METHODS:Forty-three patients with mCRC treated with cetuximab or panitumumab were entered into this study. According to the Response Evaluation Criteria in Solid Tumors criteria, 30 patients had stable or progressive disease (non-responsive), while 13 patients had a partial response. Tumor buds were evaluated from whole tissue sections stained for pan-cytokeratin, evaluated in the densest region using a 40 × objective and "high-grade" tumor budding was defi ned as 15 buds/high-power f ***: Tumor buds and K-RAS mutation both correctly classif ied 68% of patients. All patients with K-RAS mutation (n=7) or high-grade tumor budding (n=11) were non-responsive, of which 4 patients had both features. All 13 partial responders were K-RAS wild-type with low-grade tumor budding. Combined, the predictive value of K-RAS and tumor budding was 80%. Additionally, high-grade tumor budding was significantly related to worse progression-free survival [HR (95% CI): 2.8 (1.3-6.0, P=0.008)].CONCLUSION: If confirmed in larger cohorts, the addition of tumor budding to K-RAS analysis may represent an effective approach for individualized patient management in the metastatic setting.

AIM： To evaluate endoscopic mucosal resection （EMR） in patients with high-grade dysplasia （HGD） and/or intramucosal cancer （IMC） in Barrett＇s esophagus （BE）. METHODS： Between June 2000 and December 2003, 39 consecutive patients with HGD （35） and/or IMC （4） underwent EMR. BE 〉30 mm was present in 27 patients. In three patients with short segment BE （25.0%）, HGD was detected in a normal appearing BE. Lesions had a mean diameter of 14.8＋10.3 ram. Mucosal resection was carried out using the cap method. RESULTS： The average size of resections was 19.7± 9.4×14.6＋8.2 mm. Histopathologic assessment postresection revealed 5 low-grade dysplasia （LGD）（12.8%）, 27 HGD （69.2%）, 2 IMC （5.1%）, and 5 SMC （-12.8%）. EMR changed the pre-treatment diagnosis in 10 patients （25.6%）. Three patients with SMC underwent surgery. Histology of the surgical specimen revealed 1 TON0 and 2 TIN0 lesions. The remaining two patients were cancer free at 32.5 and 45.6 mo, respectively. A metachronous lesion was detected after 25 mo in one patient with HGD. Intra-procedural bleeding, controlled at endoscopy, occurred in four patients （10.3%）. After a median follow-up of 34.9 mo, all patients remained in remission. CONCLUSION： In the medium term, EMR is effective and safe to treat HGD and/or IMC within BE and is a valuable staging method. It could become an alternative to surgery.

A Local Ensemble Transform Kalman Filter Data Assimilation System for the Global FSU Atmospheric Model

Data assimilation is the process by which measurements and model predictions are combined to obtain an accurate representation of the state of the modeled system. We implemented a data assimilation scheme called LETKF （local ensemble transform Kalman filter） with FSUGSM （Florida State University Global Spectral Model） and made an experiment to evaluate the initial condition generated to numerical weather prediction to FSUGSM model. The LETKF analysis carries out independently at each grid point with the use of ＂local＂ observations. An ensemble of estimates in state space represents uncertainty. The FSUGSM is a multilevel （27 vertical levels） spectral primitive equation model, where the variables are expanded horizontally in a truncated series of spherical harmonic functions （at resolution T63） and a transform technique is applied to calculate the physical processes in real space The assimilation cycle runs on the period 01/01/2001 to 31/01/2001 at （00, 06, 12 and 18 GMT） for each day. We examined the atmospheric fields during the period and the OMF （observation-minus-forecast） and the OMA （observation-minus-analysis） statistics to verify the analysis quality comparing with forecasts and observations. The analyses present stability and show suitable to initiate the weather predictions.
